Artigo de Periódico Acesso aberto (Open Access) Anemia hemolítica causada por Indigofera suffruticosa (Leg. Papilionoideae) em bovinos(2001-03) BARBOSA NETO, José Diomedes; OLIVEIRA, Carlos Magno Chaves; PEIXOTO, Paulo Fernando de Vargas; BARBOSA, Imke Barbara Pfeifer; ÁVILA, Sandra Cristina de; TOKARNIA, Carlos Maria Antônio HubingerThe aereal parts of Indigofera suffruticosa Mill. (family Leg. Papilionoideae) were force-fed fresh to 6 bovines in daily doses of 10 to 40 g/kg. Cattle breeders of various parts of the Northeast of Brazil accuse this plant as the cause of a non-fatal disease characterized by hemoglobinuria. The disease occurs only in years when the plant proliferates well invading the native pastures. All experimental animals had hemoglobinuria, which was transitory, inspite continuation of the administration of the plant. Two animals had no further manifestations, a third animal showed only slight other manifestations, and the other three had additional symptoms of moderate intensity. These were apathy, whitish visible mucous membranes, rough hair coat, anorexia, descrease in frequency and intensity of the ruminal movements, tachycardia, positive venous pulse and dispnoea. Before the occurrence of the hemolytic crisis the urine had a bluish-green colour. None of the experimental animals died, but one was euthanized whilst showing hemoglobinuria. Post-mortem findings were anemia, the bladder containing wine-red urine, swollen dark-brown kidneys, liver on the outside and on the cut-surface bluish and with perceptible lobular design. The main histological changes were found in liver and kidney. In the liver there was coagulative necrosis and cloudy swelling and/or cytoplasmatic microvacuolization of the hepatocytes; in the kidney there was severe nephrosis, associated with large amounts of filtrate and/or hemoglobine in the Bowman spaces, in the tubules and also in the cytoplasm of the epithelial cells.Artigo de Periódico Acesso aberto (Open Access) Estudo comparativo de algumas provas funcionais do fluido ruminal e de metabólitos sangüíneos de bovinos e bubalinos(2003-01) BARBOSA NETO, José Diomedes; ÁVILA, Sandra Cristina de; DIAS, Regina Valéria da Cunha; BARBOSA, Imke Barbara Pfeifer; OLIVEIRA, Carlos Magno ChavesIn adult buffaloes and cattle in the Amazonian Region, fed elephant grass (Pennisetum purpureum), the following values of the ruminal fluid were determined: colour, odor, viscosity, sedimentation, flotation, pH, the reduction of methylene blue, glucose fermentation, total acidity, total number of protozoa, the amount of chlorides, reduction of nitrites and the predominant group of bacteria. In the blood serum, the amounts of chlorides, total proteins and urea were also established. A comparison between the parameters observed in buffaloes and cattle did not show significant differences, with exception of the amount of chlorides and the number and distribution of protozoa. In buffaloes the amount of chlorides was lower and the number of protozoa was higher than those observed in cattle; additionally in buffaloes the small protozoa predominated, to the contrary of cattle, in which the large protozoa predominated.Artigo de Periódico Acesso aberto (Open Access) Parâmetros sangüíneos e urinários, no pré e pós parto, de búfalas criadas em sistema exclusivo de pastejo(2003-06) OLIVEIRA, Carlos Magno Chaves; BARBOSA NETO, José Diomedes; BARBOSA, Imke Barbara Pfeifer; CARDOSO, Deugles PinheiroEight lactating buffaloes of the Murrah breed were used from 60 days before up to 60 days after calving, in order to evaluate alterations in the concentration of blood glucose, total plasmatic protein, haematocrit and the presence of ketonic bodies in the urine. The 5 to 10- year old buffaloes were clinically healthy and had had at least two lactations. During the experimental period the animals were maintained on Brachiaria brizantha pasture and supplemented with a mineral mixture and water. The media concentrations of blood glucose and the media values of the haematocrit diminished significantly after calving (p<0,05). The concentrations of total plasmatic protein did not show significant variation during the pre and post-calving period. The ketonic bodies in the pre-calving period were only detected in the urine of one buffaloe, but after the 32nd day of lactation ketonic bodies were detected in all animals. There was a direct relationship between the color of the urine positive by the Rothera test and blood glucose concentrations. It can be concluded that at the beginning of lactation the buffaloes had an energetic deficit, characterized by a decline of blood glucose concentrations and the presence of ketonic bodies in the urine, and that lactation caused a progressive decline of the hematocrit, but that the concentration of total plasmatic protein did not vary during the pre and post-calving period.Artigo de Periódico Acesso aberto (Open Access) The development of an enzyme-linked immunosorbent assay for Trypanosoma vivax antibodies and its use in epidemiological surveys(Universidade Federal do Pará, 2006-11) MADRUGA, Cláudio Roberto; ARAÚJO, Flábio Ribeiro de; CAVALCANTE, Gustavo Góes; MARTINS, Charles Ferreira; BARBOSA, Imke Barbara Pfeifer; RIBEIRO, Laura Rios; KESSLER, Raul Henrique; SOARES, Cleber Oliveira; MIGUITA, Midori; MELO, Elaine Silva de Pádua; ALMEIDA, Robson Ferreira Cavalcante de; LIMA JUNIOR, Manoel Sebastião da CostaThere are data indicating that the distribution of Trypanosoma vivax in the Brazilian territory is expanding with potential to reach other areas, where the vectors are present. The detection of anti-trypanosomal antibodies in serum provides important information of the trypanosomal status in cattle herds. For this reason, an enzyme-linked immunosorbent assay (Tv-ELISA-Ab) with crude antigen from one Brazilian isolate of T. vivax was developed and evaluated. The sensitivity and specificity were respectively 97.6 and 96.9%. In the evaluation of cross-reactions, three calves inoculated with T. evansi trypimastigotes blood forms showed optical densities (OD) under the cut-off during the whole experimental period, except one at 45 days post-inoculation. With relation to Babesia bovis, B. bigemina, and Anaplasma marginale, which are endemic hemoparasites in the studied area, the cross-reactions were shown to be 5.7, 5.3, and 1.1%, respectively. The first serological survey of Pantanal and state of Pará showed that T. vivax is widespread, although regions within both areas had significantly different prevalences. Therefore, this Tv-ELISA-Ab may be a more appropriate test for epidemiological studies in developing countries because the diagnostic laboratories in most countries may be able to perform an ELISA, which is not true for polymerase chain reaction.
