Item Acesso aberto (Open Access) Copaiba oil effect on experimental jaw defect in Wistar rats(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2015-02) SILVA, Priscilla Flores; BRITO, Marcus Vinicius Henriques; PONTES, Flávia Sirotheau Corrêa; RAMOS, Suzana Rodrigues; MENDES, Laís Cordeiro; OLIVEIRA, Louize Caroline MarquesPURPOSE: To evaluate the effects of copaiba oil on jaw defects repair in Wistar rats treated with bioglass or adipose tissue. METHODS: A jaw defect was randomly created in forty-two rats and filled with bioglass or adipose tissue. The two groups (Gbio and Gcell) were subdivided in three subgroups with seven animals each according to gavage administration: control (distillated water), oil (copaiba oil) and melox (meloxicam). Euthanasia was performed after forty post-operative days. The bone formation was analyzed regarding the histological aspects. RESULTS: The osteoclasts activity was observed only in four subgroups (p=0.78). Regarding the osteoblasts presence, it was very similar between the subgroups, the difference was due to Gcell-melox (p=0.009) that presented less osteoblastic activity. The inflammatory cells were more evident in Gcell-melox subgroup, however, there was no difference in comparison with the other subgroups (p=0.52). Bone formation was observed in all subgroups, just two animals showed no bone formation even after 40 days. More than 50% of bone matrix mineralization was observed in 56% (23 animals) of the analyzed areas. The bone matrix mineralization was not different between subgroups (p=0.60). CONCLUSIONS: The subgroups that received copaiba oil showed bone repair, although not statistically significant in comparison to subgroups treated whit meloxicam or controls. Copaiba oil administered by gavage had no effect on bone repair in this experimental model.Item Acesso aberto (Open Access) Effect of andiroba oil on periodontitis in Wistar rats(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2013-06) CARMONA, Glaucia Babeto; TEIXEIRA, Renan Kleber Costa; BRITO, Marcus Vinicius Henriques; PONTES, Flávia Sirotheau Corrêa; ANDRADE, Eloisa Helena de Aguiar; FONSECA, Felipe Paiva; COSTA, Ricardo Miranda Brito; CARVALHO, Francideise MartinsPURPOSE: To evaluate the effects of andiroba oil on the periodontitis in rats. METHODS: The periodontitis was induced by the placement of cotton ligatures around the cervix of the second upper molars on fifteen rats, and waiting fifty days. The animals were randomly distributed into three groups: saline group, andiroba oil group and meloxican group, differentiated by substance used in the treatment of periodontitis. The groups received the respective substance by gavage for seven days, after the periodontitis induced. It was analyzed the score of inflammatory cells and the measurement from the cemento-enamel junction to the bone crest. RESULTS: The andiroba oil group (p=0.008) and meloxican group (p=0.0347) show a less score of inflammatory cells than saline group, however there weren't difference between them (p=0.2754). Regarding the analysis of measurement from the cemento-enamel junction to the bone crest, there was no difference between groups studied (p=0.3451). CONCLUSION: Andiroba oil decreased the quantity of inflammatory cells, however, it didn't have an effect on the measurement of alveolar bone loss, like the treatment with Meloxican®.Item Acesso aberto (Open Access) Effect of copaiba oil in hepatic damage induced by acetaminophen in rats(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2013-07) TEIXEIRA, Renan Kleber Costa; YAMAKI, Vitor Nagai; YASOJIMA, Edson Yuzur; BRITO, Marcus Vinicius HenriquesPURPOSE: To investigate the effects of copaiba oil on the hepatic damage induced by paracetamol. METHODS: Thirty six rats were distributed into six study groups (N=6): control group, that didn't receive the acetaminophen; Acetaminophen Group, that only received the acetaminophen; Prophylactic Copaiba Group 1, that received copaiba oil two hours before the acetaminophen; Prophylactic Copaiba Group 7, that received copaiba oil seven days, once by day, before the acetaminophen; Therapy Copaiba Group, that received the copaiba oil two hours afther the acetaminophen; and N-Acetyl-Cysteine Group, , that received the N-Acetyl-Cysteine two hours afther the acetaminophen. Euthanasia was performed after 24 hours. The serum levels of AST, ALT, alkaline phosphatase, GT, total bilirubin, direct bilirubin and indirect bilirubin and histological analisis were analized. RESULTS: The prophylactic copaiba group 7, therapy copaiba group and N-Acetyl-Cysteine Group showed amounts of AST and ALT similar to the control group; and the prophylactic copaiba group 1 showed similar levels to the acetaminophen group. There was no significant difference between the groups regarding the amount of alkaline phosphatase and GT (p>0.05). The therapy copaiba group showed the highest levels of bilirubin and was statistically different from the other groups (p<0.01) and this increased the costs of direct bilirubin. Regarding histopathology, the oil of copaiba administered prophylactic or therapeutic form for 7 days could decrease the amount of necrosis and inflammatory infiltrate. CONCLUSION: Copaiba oil administered prophylactically for seven days, and therapeutic could reduce liver damage caused by paracetamol similarly N-Acetyl-Cysteine, however, when treated with copaiba therapeutically showed increases in bilirubin, costs increasing fraction indirect.Item Acesso aberto (Open Access) Effects of hypertonic saline solution associated to remote ischemic perconditioning in kidney ischemia/reperfusion injury in rats(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2017-03) BRITO, Marcus Vinicius Henriques; YASOJIMA, Edson Yuzur; PÉRCARIO, Sandro; RIBEIRO JUNIOR, Rubens Fernando Gonçalves; CAVALCANTE, Lainy Carollyne da Costa; MONTEIRO, Andrew Moraes; COUTEIRO, Rodrigo Paracampo; RODRIGUES, Ivone Aline da Silva; SANTOS, Hellen Aparecida Geyer dosTo evaluate the effects of hypertonic saline solution associated to remote ischemic perconditioning in renal ischemia/reperfusion injury in rats. Methods: Twenty five male rats (Wistar) underwent right nephrectomy and were distributed into five groups: Sham group (S); Ischemia/Reperfusion group (I/R) with 30 minutes of renal ischemia; Remote ischemic perconditioning group (Per) with three cycles of 10 minutes of I/R performed during kidney ischemia; Hypertonic saline solution group (HSS) treated with hypertonic saline solution (4ml/kg); remote ischemic perconditioning + Hypertonic saline solution group (Per+HSS) with both treatments. After reperfusion, blood samples were collected for BUN and creatinine serum levels analyzes. TBARS were evaluated in plasma and renal tissue to assess oxidative stress. Kidney histopathological examination were performed. Results: Per+HSS group showed a lower degree of renal dysfunction in relation to I/R group, whereas the technique of remote ischemic perconditioning isolated or associated with saline solution significantly reduced oxidative stress and histological damage. Conclusion:Remote ischemic perconditioning associated or not to saline solution promoted reduction of acute renal injury induced by ischemia/reperfusion.Item Acesso aberto (Open Access) Immediate graft histological assay, post pig's liver transplantation(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2013-09) BRITO, Marcus Vinicius Henriques; ARAÚJO, Marialva Tereza Ferreira de; CUNHA, Luane Cristine Batista; OLIVEIRA, Antônio Jorge Barbosa de; RODRIGUES, Jacqueline Barreto; BRITO, Nathalya BotelhoPURPOSE: To describe the vascular and tissue histopathological changes in seven sequential experimental liver transplantations in pigs. METHODS: Fourteen female pigs, Sus domesticus species, with body mass between 5 and 8 kg were utilized. After the end of all anastomoses of the graft implantation in the receptor, the animal was monitored for 30 minutes, and at its end one of the biopsies was collected for histological analysis. The histological criteria utilized were: lytic hepatocyte necrosis, density of septal and portal inflammatory infiltrated, sinusoidal congestion and hemorrhage. The analysis was performed separately for the portal region in zone 1, 2 and 3. RESULTS: Among the structural changes undergone by the graft, those with greater frequency and intensity were vascular congestion and steatosis, which stood out in transplantations 5, 6 and 7. CONCLUSIONS: The technique demonstrated vascular alterations represented by vasocongestion, edema and minimum inflammatory reaction. In relation to the parenchyma, was observed macrovacuolar pan-acinar steatosis, focal lytic and occasional hemorrhages, beyond the accumulation of hemosiderin in Kuppfer's cells.Item Acesso aberto (Open Access) Lesão intestinal após isquemia-reperfusão: estudo comparativo usando sal tetrazólico (MTT) e histologia(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2001-03) BRITO, Marcus Vinicius Henriques; ARAÚJO, Marialva Tereza Ferreira de; ACÁCIO, Glayce June Sasaki; ACÁCIO, Glayce Josy Sasaki; REIS, José Maciel Caldas dosMany methods are used to evaluate ischemia reperfusion injury, but the most employed one is the histological study. However, it only demonstrates on mucosal tunic, the index of lesion and morphologic preserved cells, but not the index of viable functional cells, present in the sample. With this purpose, a colorimetric method was used, employing Methyl Thiazolyl Blue (MTT). The experiment was conducted in 30 Wistar male rats, distributed in 3 groups: Group Control (GC), Group ischemia and reperfusion-1 (GIR-1) and Group ischemia and reperfusion-3 (GIR-3), with 10 animals each. The Groups GIR-1 and GIR-3 were submitted to intestinal ischemia for 30 minutes by a false ligature of superior mesenteric artery, and submitted to euthanasia after 1 and 3 days of reperfusion, when material was picked for absorbency and histological procedures. It was observed a smaller proportion of viable cells and a larger degree of mucosal lesion in GIR-3 group (p<0.05), while GC group was the one with the larger proportion of viable cells and smaller degree of the mucosal injury (p<0.05). This way we concluded that MTT is as effective and reliable as the histological study at evaluating alterations produced by intestinal ischemia-reperfusion.Item Acesso aberto (Open Access) New experimental model of exposure to environmental tobacco smoke(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2013-12) BRITO, Marcus Vinicius Henriques; YASOJIMA, Edson Yuzur; SILVEIRA, Edvaldo Lima; YAMAKI, Vitor Nagai; TEIXEIRA, Renan Kleber Costa; FEIJÓ, Daniel Haber; GONÇALVES, Thiago BarbosaPURPOSE: To describe a new model to passive smoking for rodents. METHODS: Twenty rats were distributed into two study groups (N=10): control group (CG), that was not exposed to tobacco smoke and used as normal standard for biochemical and histological analysis; Experimental Group (EG), that Animals were exposed to the passive smoking; Euthanasia was performed after 14 days of exposure. The serum level of nicotine and histological analysis were performed. RESULTS: There was a statistical difference on the nicotine serum levels between Experimental and Control group, with level of 286 ±23 nanograma/mL in the EG and undetectable on CG (p<0.01). The histological study suggested the model efficacy producing alveolar destruction and emphysema in the EG compared with the insignificant lesions in the CG's lung. CONCLUSION: The model of exposure to environmental tobacco smoke for rodents induced easily the changes related to secondhand smoke.Item Acesso aberto (Open Access) Stem cells from adipose tissue improve the time of wound healing in rats(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2016-12) OHASHI, Camila Melo; CALDEIRA, Fabio Alves Morikawa; FEITOSA JUNIOR, Denilson José Silva; VALENTE, André Lopes; DUTRA, Paulo Roberto Witter; MIRANDA, Moysés dos Santos; SANTOS, Simone do Socorro Damasceno; BRITO, Marcus Vinicius Henriques; OHASHI, Otávio Mitio; YASOJIMA, Edson YuzurPURPOSE: To evaluate the Adipose Stem Cells (ACS) therapy efficacy on the time and quality of wound healing process in rats. METHODS: Nine male Wistar rats were randomly distributed into three groups I) 7 days of healing; II) 14 days of healing; III) 21 days of healing. Four incisions were made on the dorsal surface of each rat and then treated with intralesional ACS, meloxicam, and no treatment and ACS+meloxicam. Macroscopic evaluation was measured by percentage of healing and histopathological by hematoxylineosin was performed. RESULTS: All groups have the wound reduced during the three weeks (p<0.001) and after 14 days of healing had greater reduction than others. Wounds treated with ASC had accelerated healing in relation to no treatment and only meloxicam (p<0.001), excepting the ASC+Meloxicam that was similar (p=0.13). There was no difference in histopathological analysis between lesions. CONCLUSION: Adipose stem cell have benefits in reducing time of healing of experimental model of wound in rats, observed 7 days of after application.Item Acesso aberto (Open Access) Viabilidade celular da mucosa do intestino delgado de ratos, após correção de choque hipovolêmico com solução de NaCl 7,5%(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2003-08) BRITO, Marcus Vinicius Henriques; NIGRO, Amaury José Texeira; MONTERO, Edna Frasson de Souza; NASCIMENTO, José Luiz Martins do; SILVA, Paula Roberta Ferreira da; SIQUEIRA, Roberta Bianca PeresStudy the effect of the volemic correction with different solutions, in the mucous of the small bowel in rats. Were used 120 rats Wistar (Rattus norvegicus albinus), males, adults, seemingly healthy, with individual weight varying between 310 and 410g, originating from of the Instituto Evandro Chagas of Belém of Pará, submitted to an adaptation period of 15 days, receiving water and ration ad libitum, during the role experiment. For the research, ten animals were distributed, in groups and subgroups as following: Standard group (S), Shock group (Sh), Physiological Solution group (PS) and Hypertonic Solution Group (HS). The groups were divided in subgroups with 10 animals each, in agreement with the day of postoperative (DPO) foreseen for the euthanasis of the animals, (1st, 3rd or 7th DPO), being after this, picked material for cellular viability in every animals. RESULTS: The group PS took less quantity viable cells. The volemic correction with chloride of sodium solution at 7.5%, when compared the correction with chloride of sodium at 0.9% (isotonic solution), took the maintenance of larger amount of viable cells, in the small bowel in rats.
