Item Acesso aberto (Open Access) Chromosomal diversity in three species of electric fish (Apteronotidae, Gymnotiformes) from the Amazon Basin(2014-12) SILVA, Fernando Henrique Ramos; PIECZARKA, Julio Cesar; CARDOSO, Adauto Lima; SILVA, Patrícia Corrêa da; OLIVEIRA, Jonas Alves de; NAGAMACHI, Cleusa YoshikoCytogenetic studies were carried out on samples of Parapteronotus hasemani, Sternarchogiton preto and Sternarchorhamphus muelleri (Apteronotidae, Gymnotiformes) from the Amazon basin. The first two species exhibited both a 2n = 52 karyotype, but differed in their karyotypic formulae, distribution of constitutive heterochromatin, and chromosomal location of the NOR. The third species, Sternarchorhamphus muelleri, was found to have a 2n = 32 karyotype. In all three species the DAPI and chromomycin A3 staining results were consistent with the C-banding results and nucleolar organizer region (NOR) localization. The 18S rDNA probe confirmed that there was only one pair of ribosomal DNA cistron bearers per species. The telomeric probe did not reveal interstitial telomeric sequences (ITS). The karyotypic differences among these species can be used for taxonomic identification. These data will be useful in future studies of these fishes and help understanding the phylogenetic relationships and chromosomal evolution of the Apteronotidae.Item Acesso aberto (Open Access) Cytogenetics of the Brazilian Bolitoglossa paraensis (Unterstein, 1930) salamanders (Caudata, Plethodontidae)(2014-09) SILVA, Jéssica Barata da; SUAREZ, Pablo; NAGAMACHI, Cleusa Yoshiko; CARTER, Timothy Frederick; PIECZARKA, Julio CesarPlethodontid salamanders of genus Bolitoglossa constitute the largest and most diverse group of salamanders, including around 20% of living caudate species. Recent studies have indicated the occurrence of five recognized species in the Brazilian Amazon Rainforest. We present here the first cytogenetic data of a Brazilian salamander, which may prove to be a useful by contribution to the cytotaxonomy of the genus. Specimens were collected near the “type” locality (Utinga, Belém, PA, Brazil). Chromosomal preparations from duodenal epithelial cells and testes were subjected to Giemsa staining, C-banding and DAPI/CMA3 fluorochrome staining. All specimens showed a karyotype with 13 bi-armed chromosome pairs (2n = 26). Nucleolar Organizer Regions, evidenced by CMA3, were located distally on the long arm of pair 7 (7q). DAPI+ heterochromatin was predominantly centromeric, with some small pericentromeric bands. Although the C-banding patterns of other Bolitoglossa species are so far unknown, cytogenetic studies conducted in other Plethodontid salamanders have demonstrated that pericentromeric heterochromatin is a useful cytological marker for identifying interspecific homeologies. Species diversification is usually accompanied by chromosomal changes. Therefore, the cytogenetic characterization of Bolitoglossa populations from the middle and western Brazilian Amazon Basin could identify differences which may lead to the identification of new species.Item Acesso aberto (Open Access) Karyotypic and morphological divergence between two cryptic species of Eigenmannia in the Amazon basin with a new occurrence of XX/XY sex chromosomes (Gymnotiformes: Sternopygidae)(2015-06) SILVA, Danillo dos Santos; PEIXOTO, Luiz Antônio Wanderley; PIECZARKA, Julio Cesar; WOSIACKI, Wolmar Benjamin; READY, Jonathan Stuart; NAGAMACHI, Cleusa YoshikoEigenmannia species are widely distributed in the Neotropics, with eight valid species currently recognized. Populations of Eigenmannia from three locations in the eastern Amazon were investigated using cytogenetic and morphological techniques, revealing two taxa designated here as Eigenmannia sp. "A" and Eigenmannia sp. "B". The species differ in three morphometric characters, two meristic characters, and one osteological character. Eigenmannia sp. "A" presents 2n = 34 (22 m/sm+12 st/a) and Eigenmannia sp. "B" presents 2n = 38 (14 m/sm+24st/a) and simple differentiated sex chromosomes of the type XX/XY. In both species the Constitutive Heterochromatin (CH) rich in A-T bases is distributed in the centromeric region of all chromosomes. Eigenmannia sp. "B" also presents CH blocks in the interstitial region of chromosome pairs 8, 9 and X which are positively stained with CMA3, indicating G-C rich regions. The NOR is located on the short arm of chromosome pair 17 of Eigenmannia sp. "A" and on the short arm of pair 14 of Eigenmannia sp. "B". FISH with rDNA probes hybridized to different-sized regions between homologs, suggesting heteromorphism. The differentiation of the X chromosome in Eigenmannia sp. "B" could be the result of amplification of repetitive DNA sequences.Item Acesso aberto (Open Access) Profile of micronucleus frequencies and nuclear abnormalities in different species of electric fishes (Gymnotiformes) from the Eastern Amazon(2013) MELO, Karina Motta; ALVES, Ingrid Reale; PIECZARKA, Julio Cesar; DAVID, José Augusto de Oliveira; NAGAMACHI, Cleusa Yoshiko; GRISOLIA, Cesar KoppeThe frequency of spontaneous micronucleus (MN) formation in fish species needs to be determined to evaluate their usefulness for genotoxic biomonitoring. The definition of a good bioindicator takes into account the current knowledge of its metabolic traits as well as other factors including its feeding behavior and relationship to the environment. In this study, we compared the basal frequencies of micronucleated erythrocytes and nuclear abnormalities (NA) among different species of the fish Order Gymnotiformes (Rhamphichthys marmoratus, Steatogenys elegans, Sternopygus macrurus, Parapteronotus hasemani, Gymnotus mamiraua, Gymnotus arapaima, Brachyhypopomus beebei, Brachyhypopomus n. sp. BENN) sampled in several localities of the Eastern Amazon. A baseline of MN and NA frequency in these fish was determined, enabling the identification of potentially useful species as models for genotoxicity studies. Only one impacted sample collected at a site in the River Caripetuba showed a significant number of NAs, which may be due to the release of wastewater by neighbouring mining industries and by the burnt fuel released by the small boats used by a local community. Our results may provide support for further studies in areas of the Eastern Amazon affected by mining, deforestation and other anthropogenic activities.Item Acesso aberto (Open Access) X1X1X2X2/X1X2Y sex chromosome systems in the Neotropical Gymnotiformes electric fish of the genus Brachyhypopomus(2015-06) CARDOSO, Adauto Lima; PIECZARKA, Julio Cesar; NAGAMACHI, Cleusa YoshikoSeveral types of sex chromosome systems have been recorded among Gymnotiformes, including male and female heterogamety, simple and multiple sex chromosomes, and different mechanisms of origin and evolution. The 1X1X2X2/X1X2Y systems identified in three species of this order are considered homoplasic for the group. In the genus Brachyhypopomus, only B. gauderio presented this type of system. Herein we describe the karyotypes of Brachyhypopomus pinnicaudatus and B. n. sp. FLAV, which have an X1X1X2X2/X1X2Y sex chromosome system that evolved via fusion between an autosome and the Y chromosome. The morphology of the chromosomes and the meiotic pairing suggest that the sex chromosomes of B. gauderio and B. pinnicaudatus have a common origin, whereas in B. n. sp. FLAV the sex chromosome system evolved independently. However, we cannot discard the possibility of common origin followed by distinct processes of differentiation. The identification of two new karyotypes with an X1X1X2X2/X1X2Y sex chromosome system in Gymnotiformes makes it the most common among the karyotyped species of the group. Comparisons of these karyotypes and the evolutionary history of the taxa indicate independent origins for their sex chromosomes systems. The recurrent emergence of the X1X1X2X2/X1X2Y system may represent sex chromosomes turnover events in Gymnotiformes.
