    Antraquinonas e naftoquinonas do caule de um espécime de reflorestamento de Tectona grandi (Verbenaceae)

    The hexane extract of the bark of Tectona grandis (Verbenaceae) afforded two anthraquinones and two naphtoquinones. Their caracterizations were obtained through NMR spectroscopic techniques. This is the first phytochemical study of the bark of Tectona grandis reforestation specimen in Brazil. The main interest in this work is proving the presence of tectoquinone in reforestation specimen.

    The Amazonian Brazilian offers an appreciable potential of plants with therapeutic properties, although most are little known. In this way, with the objective of verifying the potentiality nutritional of medicinal herbs, a work was developed to determine the concentration of Ca, Mg, Fe, Cu and Zn in the leaves and in the teas of these species: Piper callosum Ruiz & Pav., Piperaceae, Mikania lindleyana DC., Asteraceae e Arrabidaea chica (Humb. & Bonpl.) B. Verl., Bignoniaceae. After the plants samples have been processed, they were submitted to digestion and soon afterwards the metals were analyzed in an spectrophotometer of Atomic Absorption. The results showed the follow yields: for the tea of Arrabidaea chica Ca were detected (6955 to 20058 mg/L), Mg (2390 to 3094 mg/L) and Fe (40 to 61 mg/L). For the tea of Mikania lindleyana besides the presence of high values of Ca (17722 to 22336 mg/L), Mg (4531 to 9370 mg/L) and Fe (20 to 87 mg/L) they were found from 7 to 16 mg/L of Cu and 9 to 41 mg/L of Zn. The tea of the Piper callosum presented 2036 to 4344 mg/L of Ca, 618 to 4023 mg/L of Mg and 39 to 60 mg/L of Fe. Being compared the results of the minerals with the values recommended by the Health World Organization, is possible that the present metals in the teas of the plants could contribute in the complementation of the people's alimentary diets that use these medicinal plants.

    Biotransformation of chalcones by the endophytic fungus Aspergillus flavus isolated from Paspalum maritimum trin

    The fungus Aspergillus flavus isolated as endophytic of the plant Paspalum maritimum Trin. was evaluated for its potential application in biotransformation reactions. The compounds chalcone (1), 3,4,5-trimethoxychalcone (2) and 2,3,4,4'-tetramethoxychalcone (3) were biotransformed, respectively, in dihydrochalcone (4), 3,4,5-trimethoxydihydrochalcone (5) and 2,3,4,4'-tetramethoxydihydrochalcone (6). The structures were elucidated by spectroscopic methods including 1D and 2D NMR techniques, and MS analysis. The dihydrochalcones 5 and 6 are new compounds.

    Biotransformation of sucrose into 5-hydroxy-2-hydroxymethyl-γ-pirone by Aspergillus flavus

    The sucrose hydrolysis and the preference of consumption of glucose instead of fructose were investigated for the production of 5-hydroxy-2-hydroxymethyl-γ-pyrone (HHMP) in the presence of Aspergillus flavus IOC 3974 cultivated in liquid Czapeck medium. Standardized 0.5g of pellets were transferred as inoculum into twelve conical flasks of 250 ml containing 100 ml of medium with different sucrose concentration, which was kept at 120 rpm and 28"C for 16 days without pH adjustment. Aliquots of 500μl of the broth culture were withdrawn at 24 h intervals and analyzed. The major yield of HHMP was 26g l-1 in 120g l-1 of sucrose. At these conditions, A. flavus produced an invertase capable of hydrolyzing 65% of total sucrose concentration in 24h, and an isomerase capable of converting fructose into glucose. In this work, it focused the preference for glucose and, then, of fructose by A. flavus and the strategy used to produce HHMP.

    A dehydrorotenoid produced by callus tissue culture and wild plant roots of Boerhaavia coccinea

    Calli cultures were established from leaves and stem of B. coccinea plantlet produced in vitro and analysed for isofl avonoid content. The quantification of 6,9,11-trihydroxy- 6a,12a-dehydrorotenoid isolated from the roots of Boerhaavia coccinea P. Miller collected from its natural environment, and the same metabolite produced in callus tissue culture of the same plant are described in this paper. The rotinary quantitative HPLC analysis indicated that callus culture produced the same isoflavonoid compound found in the roots of intact wild growing plant. The amount of the secondary metabolite produced in vitro was 955.35 􀈝g/g of dry cell weight, 2.5 times more than the highest amount concentration produced by the wild growing plant in its natural environment.

    Dihydroflavonols from the leaves of Derris urucu (Leguminosae): structural elucidation and DPPH radical-scavenging activity

    Derris urucu is an Amazonian plant with insecticide and ichthyotoxic properties. Studies with this species show the presence of flavonoids, mainly rotenoids, as well as stilbenes. The ethanol extract of the leaves of Derris urucu (Leguminosae) afforded three new dihydroflavonols named urucuol A (1), B (2) and C (3), and the dihydroflavonol isotirumalin (4). Their structures were elucidated by extensive analysis of 1D and 2D NMR, UV and IR spectra and MS data and comparison with literature data. The isolated compounds (1-4) were evaluated for DPPH• radical scavenging activity and showed a relatively lower antioxidant ability compared to the commercial antioxidant trans-resveratrol.

    Efeito fungitóxico in vitro do óleo resina e do óleo essencial de copaíba (Copaifera multijuga Hayne)

    In vitro fungitoxic effect of the oil-resin and the essential oil of copaiba (Copaifera multijuga Hayne). Copaifera multijuga Hayne oil, crude and fractions, was evaluated for its in vitro fungitoxic activities against five species of filamentous fungi belonging to the genus Aspergillus and three yeast species of the genus Candida. Oil-resin and essential oil concentrations ranging from 0.008 mg mL-1 to 1.6 mg mL-1 were used in qualitative and quantitative analyses. Samples were placed onto 5mm-diameter paper disks and distributed on Saboraud culture medium in Petri dishes inoculated with conidia and incubated at 28ºC during 10 days. A solution with 1.6 mg mL-1 of miconazole nitrate was used as positive control. Qualitative results indicated that the oil-resin presented good fungistatic activity; however, one of the essential oil fractions was highly effective against Candida parapsilosis IOC-2882, Aspergillus flavus IOC-3874 and A. tamarii IOC- 187, with inhibition halos of 16.0±1.4 mm, 19.5±2.1 mm and 12.5±3.5 mm, respectively. The quantitative evaluation indicated that 0.3 mg mL-1 of the oil-resin inhibited Aspergillus flavus and Candida parapsilosis growth, whereas 0.08 mg mL-1 of the essential oil fraction reached this same activity.

    Improvement of friable callus production of Boerhaavia paniculata Rich and the investigation of its lipid profile by GC/MS

    In this study, a protocol to induce high amount of friable callus of Boerhaavia paniculata RICH and a lipidomics technique were applied to investigate the profile of lipids to relate to those present in the roots of this plant that presented anti-inflammatory activity in the crude hexane extract. The callus culture was induced from seeds in solidified Murashige and Skoog medium containing different amounts of glucose and different concentrations of 2,4-Dichlorophenoxyacetic acid. The explants were kept in a germination chamber at 30±2°C with a photoperiod of 16 h under light intensity of 27 µmol m–2 s–1 for 4 weeks. The best results for friable callus formation and development of the biomass were obtained in the treatment containing 2.26 µM 2.4-D and glucose (1.5 %; w/v). Lipidomics techniques were applied in hexane fraction showing higher concentrations of the steroids β-sitosterol (3.53 mg/100 g dc–dry cells), and fatty acids, especially 2-hydroxy-tetracosanoic acid (0.34 mg/100 g dc), eicosanoic acid (86.25 mg/100 g dc), stearic acid (420.83 mg/100 g dc), tetradecanoic acid (10.74 mg/100 g dc) and linoleic acid (100.61 mg/100 g dc). The lipid profile of callus versus that found in the roots of wild plant is described in this work.

    Levels of As, Cd, Pb and Hg found in the hair from people living in Altamira, Pará, Brazil: environmental implications in the Belo Monte area

    Human hair was used as a bioindicator to evaluate the levels of As, Cd, Pb and Hg in the resident population of the "Volta Grande" area of the Xingu river, in the North of Brazil. The concentrations of As and Cd are in agreement with the values found in the literature, however, the Pb and Hg concentration is very different, which can suggest an inter-regional variation. Moreover, the high values found for these elements in the residents of the Canteiro Island can be attributed to environmental exposure of this population in an area with environmental impact from activities of gold mining. The application of these methods showed that the concentrations of the metals studied in this work can correctly classify two different localities in the Altamira district. The results reflect the impact of these elements on the health of the inhabitants of this auriferous area.

    A New Prenylisoflavone from the Antifungal Extract of Leaves of Vatairea guianensis Aubl.

    A new compound, 5,7,3'-trihydroxy-4'-methoxy-8-prenylisoflavone, was isolated from the leaves of Vatairea guianensis Aubl. (Fabaceae), together with two known isoflavones lupiwighteone and 5,7,4'-trihydroxy-3'-methoxy-8-prenylisoflavone. All isolated compounds were characterized based on infrared (IR), UV, 1H and 13C nuclear magnetic resonance (NMR), including 2D NMR analyses and high resolution mass spectrometry. The ethanolic extract from V. guianensis leaves displayed activity against Candida dubliniensis, C. albicans and C. krusei. However, the EtOAc fraction from that extract exhibited more significant activity than the ethanolic extract, showing antifungal activity for all fungi species investigated. The major compound 5,7,3'-trihydroxy-4'-methoxy-8-prenylisoflavone isolated from that EtOAc fraction was also active against C. parapsilosis and C. dubliniensis.

    Novel anthraquinone derivatives produced by Pestalotiopsis guepinii, an endophytic of the medicinal plant Virola michelii (Myristicaceae)

    A new anthraquinone derivative, named guepinone (1), along with the known substances isosulochrin (2) and chloroisosulochrin (3), were isolated from a rice culture of Pestalotiopsis guepinii, an endophytic fungus of Virola michelii. The compounds were identified by analysis of 1D and 2D NMR and MS spectral data. The antimicrobial activity of these compounds was evaluated and chloroisosulchrin (3) was the most active.

    Two compounds, (+)-catechin and epicatechin, were isolated from leaves of T. myrmecophyla, using chromatographic techniques. The structural identification was carried out on the basis of 1H and 13C NMR spectral data and comparison with literature data. The compounds (+)-catechin and epicatechin were submitted to germination inhibition and radicle and hypocotyl growth assays. Results showed some significant activities confirming the initial hypothesis about allelopathic properties of that plant.

    The use of fire and mowing as weed methods have proved inefficient. Although more efficient, synthetic chemical herbicides are usually related to environmental problems. Thus, the search for natural herbicides has become a priority matter. Based on these facts, research was conducted on Myrcia guianensis (pedra-ume-caá) including the isolation and identification of chemical substances with allelophatic activity. The allelophatic effects of extracts, fractions, essential oils, and isolated chemical substances (gallic acid and protocatechuic acid) on germination and radicle and hypocotyl growth of weed species Mimosa pudica (malícia) and Senna obtusifolia (mata-pasto) were analyzed. Extracts and fractions were analyzed at a concentration of 1%, essential oils at 15, 30, 45, 60 ppm and the isolated chemical substances at 15, 30, 45, 60 ppm. Malicia showed more sensitivity to the allelophatic effects of extracts and fractions. The essential oil inhibited the germination of M. pudica but stimulated that of S. obtusifolia. The allelophatic effects of the chemical substances were concentration dependent and the strongest activity was observed at 60 ppm.

    Many of the chemical compounds found in nature that are produced by plants or microorganisms can offer new and efficient ways of controlling pests in agriculture and agricultural practice, with the help of fungi. The objective of this work is to characterize the inhibitory potential for seed germination and the plantlet development of two weed species using extracts and compounds obtained from biomass produced by Pestalotiopsis guepinii, an endophytic fungus of the species Virola michelii. The bioassays were developed under controlled conditions at 25 ºC and 12-hour photoperiod for germination, and at 25 ºC and 12-hour photoperiod for root and hypocotyl development. The crude extracts were analyzed at a concentration of 1.0% (m/v). The results showed that the more polar extracts (MeOH-1 and MeOH-2) have the highest inhibitory potential, although the hexane and ethyl acetate extract effects were important, especially for seed germination. Comparatively, weed seed germination was more sensitive to the effects than plantlet development. Mimosa pudica was more affected by the inhibitory effects of the extracts. However, for seed germination of Senna obtusifolia, the extract MeOH-1 showed 100% inhibition. The compounds ergosterol and ergosterol peroxide showed an inhibitory potential always below 35%, not showing the inhibitory potential of the hexane extract from which they were isolated. When these compounds were tested together, little increase was observed in the inhibitory activity.

    Production of L-asparaginase by filamentous fungi

    L-asparaginase production was investigated in the filamentous fungi Aspergillus tamarii and Aspergillus terreus. The fungi were cultivated in medium containing different nitrogen sources. A. terreus showed the highest L-asparaginase (activity) production level (58 U/L) when cultivated in a 2% proline medium. Both fungi presented the lowest level of L-asparaginase production in the presence of glutamine and urea as nitrogen sources. These results suggest that L-asparaginase production by of filamentous fungi is under nitrogen regulation.

    Stilbenes from Deguelia rufescens var. urucu (Ducke) A. M. G. Azevedo leaves: effects on seed germination and plant growth

    The Amazon biodiversity may provide plants whose chemical substances are capable of controlling weeds. In this study we report the isolation and identification of five stilbenes from the leaves of "timbó vermelho" (Deguelia rufescens var. urucu): 4-methoxylonchocarpene (1); 3,5-dimethoxy-4´-hydroxy-3´-prenyl-trans-stilbene (2), lonchocarpene (3), 3,5-dimethoxy-4´-O-prenyl-trans-stilbene (4) and pterostilbene (5). Compounds 2 and 4 are new natural products although 2 has been previously cited as synthesis product. Potential allelopathic activity for 1, 2 and 4 was evaluated over seed germination and plant growth of Mimosa pudica weed. The observed effects on seed germination did not vary significantly (p > 0.05) when the analysis of phytotoxicity was performed with the substances alone, the maximum inhibition did not exceed 20%. The most intense inhibitions on radicle and hypocotyl development were found for compound 4 (p < 0.05). When tested in pairs, showed antagonism for seed germination and synergism for radicle and hypocotyl development.

    The lipidome, genotoxicity, hematotoxicity and antioxidant properties of andiroba oil from the Brazilian Amazon

    Andirobeira is an Amazonian tree, the seeds of which produce a commercially valuable oil that is used in folk medicine and in the cosmetic industry. Andiroba oil contains components with anti-inflammatory, cicatrizing and insect-repellant actions. However, virtually nothing is known of the safety of this oil for humans. The aim of this work was therefore to investigate the hematotoxicity, genotoxicity and mutagenicity of andiroba oil using the comet and micronucleus assays, and to assess its antioxidant properties and lipidome as a means of addressing safety issues. For the experiments, andiroba oil was administered by gavage for 14 consecutive days in nulliparous female Swiss mice randomly distributed in four groups: negative control and three doses of oil (500, 1000 and 2000 mg/kg/day). These doses were chosen based on recommendations of the OECD guideline no. 474 (1997). GC/MS was used to investigate the free fatty acid, cholesterol and triterpene content of andiroba oil in a lipidomic analysis. No clinical or behavioral alterations were observed throughout the period of treatment, and exposure to andiroba oil at the doses and conditions used here did not result in hematotoxic, genotoxic or mutagenic effects. Tests in vitro showed that oil sample 3 from southwestern of Brazilian Amazon had a high antioxidant capacity that may protect biological systems from oxidative stress, although this activity remains to be demonstrated in vivo.
