Artigo de Periódico Acesso aberto (Open Access) Diagnóstico molecular da infecção por hemoplasmas em gatos domésticos naturalmente infectados da cidade de Belém, Pará(2013-09) SOUSA, Sinerey Karla Salim Aragão de; SAMPAIO JUNIOR, Francisco Dantas; SOUSA, Luciane Oeiras; SANTOS, Rafaelle Cunha dos; GONÇALVES, Evonnildo Costa; SCOFIELD, Alessandra; CAVALCANTE, Gustavo GóesMycoplasma haemofelis, 'Candidatus Mycoplasma haemominutum' and 'Candidatus Mycoplasma turicensis' are the causative agent of the feline mycoplasmosis, which could cause acute or chronic anemia. The aim of this work was to determine the occurrence of hemoplamas in domestic cats from Belém, Pará. To this, 201 cats were divided into three groups: Group A were composed by 101 stray cats captured by Zoonosis Control Center, group B were composed by 62 owners healthy cats and group C were composed by 38 owners cats that were suffering by some medical condition. Blood samples were collected to perform Polymerase Chain Reaction (PCR) to detect the DNA of these agents, which were sequenced and aligned. Statistical analysis was performed to detect association between the infection, the sex of the animals and experimental groups. The DNA of at least one of the hemoplasmas studied were detected in 19,9% (40/201) of the samples, being the DNA of 'Candidatus M. haemominutum' was found in 7.96% (16/201) of samples, M. haemofelis in 1.49% (3/201) of samples, while 'Candidatus M. turicensis' in 12.93% (26/201) of the samples. The DNA of these three agents was detected in cats from groups A and C, while in Group B was detected only 'Candidatus M. turicensis' and 'Candidatus M. haemominutum'. The influence of sex on hemoplasma infection was detected only between 'Candidatus M. haemominutum' and males. These findings showed that hemoplasma circulate among domestic cats in Belém, and 'Candidatus M. turicensis' and 'Candidatus M. haemominutum' were more common than M. haemofelis, especially in stray cats.Artigo de Periódico Acesso aberto (Open Access) Diagnóstico molecular e frequência de anticorpos anti-Leishmania infantum chagasi em cães do município de Belém, Pará(2014-03) SCHWANKE, Katiane; SILVA, Aryane Maximina Melo da; OLIVEIRA, Adlilton Pacheco de; SILVA, Michele Bahia do Vale; SILVEIRA, Fernando Tobias; SCOFIELD, Alessandra; CAVALCANTE, Gustavo GóesVisceral leishmaniasis is a disease whose etiological agent in Brazil is Leishmania infantum chagasi. Dogs are considered urban reservoirs of the disease, being an indicator of the human cases occurrence. The present study aimed to diagnose L. infantum chagasi infection in stray and owned dogs in Belém, Pará State, by polymerase chain reaction (PCR) and indirect immunofluorescence assay (IFA) using two different antigens. Venous blood samples from adult dogs, regardless of gender or breed, of different neighborhoods in Belém-PA, were collected in tubes with and without anticoagulant to obtain DNA and serum, respectively. These animals were divided into two groups: stray dogs captured by the Center for Zoonosis Control (Group A) and owned dogs (Group B). Sera were analyzed by IFA testing for IgG using two different antigens: 1) Bio-Manguinhos/Fiocruz antigen kit (Ag-PRO) containing promastigotes of Leishmania sp. (Complex Major-Like), 2) Instituto Evandro Chagas Antigen (Ag-AMA) consisting of amastigotes of L. infantum chagasi. The evaluation of the two antigens was performed considering positive the reactions above the 1:80 dilution. Already PCR was performed with DNA isolated from whole blood of animals and amplified with the primers RV1 and RV2. Of the 335 samples analyzed, 10.4% (35/335) were positive by IFA (Ag-PRO) and 0.9% (3/335) with the Ag-AMA. The distribution of positive samples is given as follows: Group A 14.8% (25/169) with Ag-PRO and 1.2% (2/169) with Ag-AMA; Group B 6% (10/166) with Ag-PRO and 0.6% (1/166) with Ag-AMA, being that all samples positive by IFA with Ag-AMA also reacted with Ag-PRO, and none of the samples detected DNA of L. infantum chagasi. The findings of this study indicate that Belém can still be considered non-endemic area for canine visceral leishmaniasis and the nature of the antigen influences the result of the IFA for the detection of anti-L. infantum chagasi antibodies in dogs, and the IFA using promastigotes of Leishmania major-like antigen should be used with caution as a confirmatory diagnostic on epidemiological studies in non-endemic areas.Artigo de Periódico Acesso aberto (Open Access) Infestation by Haematopinus quadripertusus on cattle in São Domingos do Capim, state of Pará, Brazil(Universidade Federal do Pará, 2012-09) SCOFIELD, Alessandra; CAMPOS, Karinny Ferreira; SILVA, Aryane Maximina Melo da; OLIVEIRA, Cairo Henrique Sousa de; BARBOSA NETO, José Diomedes; CAVALCANTE, Gustavo GóesSevere infestation with lice was observed on crossbred cattle (Bos taurus indicus × Bos taurus taurus) in the municipality of São Domingos do Capim, state of Pará, Brazil. Sixty-five animals were inspected and the lice were manually collected, preserved in 70% alcohol and taken to the Animal Parasitology Laboratory, School of Veterinary Medicine, Federal University of Pará, Brazil, for identification. The adult lice were identified as Haematopinus quadripertusus, and all the cattle examined were infested by at least one development stage of this ectoparasite. The specimens collected were located only on the tail in 80% (52/65) of the cattle, while they were around the eyes as well as on the ears and tail in 20% (13/65). Nits, nymphs and adults of the parasite were respectively collected from 98.46% (64/65), 38.46% (25/65) and 23.08% (15/65) of the animals examined. This is the first report of bovine pediculosis caused by H. quadripertusus in the state of Pará, Brazil. From February 2012 to August 2013, whole blood samples and blood smears were obtained from 45 free-living procyonids and from nine procyonids kept in captivity in wild life refuges and zoobotanical parks in the states of Amapá and Pará. Whole blood samples were collected and kept at -20ºC for the detection of T. evansi DNA by PCR using the RoTat 1.2 forward and RoTat 1.2 reverse primers. In addition, the blood smears were processed and examined for the presence of trypomastigote forms of T. evansi. T. evansi DNA was detected in 18.52% (10/54) of the procyonids, namely, in captive crab-eating raccoons and captive and free-living coatis in Pará State. No trypomastigote forms were observed in the blood smears. Each isolate was sampled from naturally infected adult chicken (Gallus gallus) from rural areas of three Brazilian municipalities: Seropédica (22º 48' S; 43º 41'W), in the state of Rio de Janeiro; Cruzeiro (22º 33' S; 44º 57'W), in the state of São Paulo; and Santa Bárbara do Tugúrio (21º 15' S; 43º 27' W), in the state of Minas Gerais. The blood samples taken from each infected chicken were inoculated in three groups of ten young chicken (21 days old). Blood smears of the experimentally infected chicken were sampled every two days until the 69th day in order to evaluate the parasitemia. For the morphological-descriptive and morphometric analyses, we measured 30 individuals from each of the intraerythocytic states, measures of the major (MD) and minor diameters (md), the estimation of morphometric index (Mi=md/MD) and size (T=pab, a= md/2; b=MD/2). The results indicated low and homogeneous parasitemia rates in the three strains, which showed differences among shape and size of the parasitic stadia displayed.Artigo de Periódico Acesso aberto (Open Access) Reemerging of natural infection by Trypanosoma evansi in horses in Arari, Marajó Island, Brazil(Universidade Federal do Pará, 2016-12) SILVA, Josileide Araújo da; DOMICIANO, Tarcísio Oliveira; MONTÃO, Daniele Pina; SOUSA, Paulo Geovani Silva; RAMOS, Leandro Lopes; PAREDES, Laura Jamille Argolo; GONZALEZ MONTEIRO, Silvia; RIVERO, Beatriz Riet Correa; SCOFIELD, Alessandra; BEZERRA JÚNIOR, Pedro; BEZERRA, Isis Abel; CERQUEIRA, Valíria DuarteTwo outbreaks of trypanosomiasis by Trypanosoma evansi in horses in the municipality of Chaves, Marajó Island, Pará State, Brazil are reported. The first outbreak occurred in April 2011 in a farm with 147 horses; of these, 47 (31.97%) got sick, and 40 (27.21%) died. The second outbreak occurred in May 2012 and involved nine properties. Of a total of 679 horses, 209 (30.07%) got sick, and 183 (26.97%) died. The main clinical signs observed in these horses were weight loss, abdominal edema, toe dragging, and pelvic muscle atrophy. Necropsy was performed in two horses, one from each outbreak. Macroscopic alterations were not seen in the horse from the first outbreak; however, the horse from the second outbreak exhibited emaciation, pallor and icterus, as well as enlarged spleen with prominent white pulp at cut surface. Histopathology of the central nervous system of both animals revealed mild to severe, diffuse lymphoplasmacytic encephalitis, with presence of Mott cells. Immunohistochemistry for T. evansi revealed structures similar to intralesional trypomastigote forms. Between November and December of 2013, an epidemiological survey was carried out in the municipalities of Cachoeira do Arari, Santa Cruz do Arari, Salvaterra, Soure, and Chaves. Only Santa Cruz do Arari and Chaves had reported cases of the disease. Blood samples were collected from 243 horses for detection of T. evansi DNA by polymerase chain reaction (PCR) and 20 were positive.
