Item Acesso aberto (Open Access) Ajustes posturais antecipatórios e compensatórios em idosos com e sem lombalgia(Universidade Federal do Pará, 2021-03) GARCEZ, Daniela Rosa; CALLEGARI, Bianca; http://lattes.cnpq.br/0881363487176703; https://orcid.org/0000-0001-9151-3896; YAMADA, Elizabeth Sumi; http://lattes.cnpq.br/7240314827308306Chronic low back pain (DLC) is associated with changes in postural control and is highly prevalent in the elderly. Research shows that aging and DLC are described as important factors that affect postural control. The postural control impairments increase the risk of falls. Researches evaluating the postural control in elderly people with DLC are still necessary for greater effectiveness in balance rehabilitation programs to prevention falls in this population. The objective of this study is to verify whether anticipatory postural adjustments (APAs) and compensatory postural adjustments (CPAs) are affected by DLC in elderly people by assessing their postural control during a self-initiated perturbation paradigm induced by rapid upper arm movement when pointing to a target. Elderly people were divided into: Group with DLC (GDLC) (n = 15) and Control Group (CG) (n = 15). The participants’ lower limb muscle onset and center of pressure (COP) displacements were assessed prior to perturbation and throughout the entire movement. T0 moment (i.e., the beginning of the movement) was defined as the anterior deltoid (DEL) onset, and all parameters were calculated with respect to it. The rectus femoris (RT), semitendinosus (ST), and soleous (SOL) showed delayed onset in the GDLC group compared with the control group: RF (control: -0.094 ± 0.017 s; GDLC: -0.026 ± 0.012 s, t = 12, p < 0.0001); ST (control: - 0.093 ± 0.013 s; GDL: -0.018 ± 0.019 s, t = 12, p < 0.0001); and SOL (control: -0.086± 0.018 s; GDL: -0.029 ± 0.015 s, t = 8.98, p < 0.0001). In addition, COP displacement was delayed in the GDLC group (control: -0.035 ± 0.021 s; GDL: -0.015 ± 0.009 s, t = 3; p = 0.003) and presented a smaller amplitude during APA COPAPA [control: 0.444 cm (0.187; 0.648); GDLC: 0.228 cm (0.096; 0.310), U = 53, p = 0.012]. The GDLC group required a longer time to reach the maximum displacement after the perturbation (control: 0.211 ± 0.047 s; GDLC 0.296 ± 0.078 s, t = 3.582, p = 0.0013). This indicates that GDLC elderly patients have impairments to recover their postural control and less efficient anticipatory adjustments during the compensatory phase. Our results suggest that people with GDL have altered feedforward hip and ankle muscle control, as shown from the SOL, ST, and RT muscle onset. This study is the first study in the field of aging that investigates the postural adjustments of an elderly population with GDLC. Clinical assessment of this population should consider postural stability as part of a rehabilitation program.Item Acesso aberto (Open Access) Alterações hepáticas por exposição a baixas doses de metilmercúrio em macacos prego, Cebus apella (Linnaeus 1758)(Universidade Federal do Pará, 2011-09-16) SILVA, Márcia Cristina Freitas da; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219Cebus apella were exposed to 1,5 ppm methylmercury (methylHg) in the diet for 120 days. Hepatotoxicity was investigated, concentrations of mercury in total blood were monitored each 30 days using atomic absorption spectrometry with cold vapor Hg201, aspartate transaminase (AST), alanine transaminase (ALT) and total bilirubin (BT) were determined. Liver was fixed by formaldehyde 10% and prepared by histopathology protocols. Significant difference was observed in groups exposed and control about total mercury (Hgtotal) in the periods of 60, 90 (P < 0,05) and 120 days (P < 0,01). The histopathology revealed moderate steatosis and hydropic degeneration, common in methylHg exposed in other species. No Significant difference between the levels of AST (p= 0.38), ALT (p= 0.83) and BT (p= 0.07) in groups exposed and control. The Pearson correlation with Hgtotal was negative (AST r= -0,7; ALT r=0,07; BT r= -0,3 e p > 0,05), suggests another studies to clarify the alert levels of mercury concentrations and liver dosages.Item Acesso aberto (Open Access) Estudo do desenvolvimento auditivo da habilidade de fusão binaural(Universidade Federal do Pará, 2022-04) ARAÚJO, Francisca Canindé Rosário da Silva; SILVA FILHO, Manoel da; http://lattes.cnpq.br/2032152778116209Introduction: Binaural Interaction (BI) allows the introduction of auditory information (in the brain as a function of differences in perception of intensity or time of acoustic stimuli). Allows you to assess the action and integrated co- operative of the brainstem in lower understanding. As the maturation of the central nervous system occurs in the craniocaudal direction, the response to this ability may change during the course of development. Objectives: To normalize and compare the development with increasing age of response in the binaural diffusion test (BPT) with digital low-pass (LP) and high-pass (HP) filters in normative listeners. Methods: Prospective, cross-sectional and observational study. A total of 120 years were evaluated, in different age groups (6 to 8 years old, 10 to 12 years old, 14 to 16 years old and 20 to 30 years old) with TFB, filter at Fc 500/1700 Hz digital Finite Impulse Response type order 4096, with null phase and 5000 between 18 and 30 years with unfiltered speech material. Results: a progressive improvement in performance with increasing age (ANOVA (one-way): p<0.0001). There was a significant difference between the filtered words, age and the unfiltered p < (Dunnet: any filtered words,01). The difference between the age groups was significant (Tukey: p<0.01), less for the results obtained in the age groups of 6-8 and 10-12 years and of 14-16 and 18- 30 years. Discussion: IB is a skill that evolves with age development and NC fabrication. Final considerations: The interpretation of the TFB should take into account the performance by age group of the patients. This is important for future applications of these tests in people with Auditory Processing Disorder.Item Acesso aberto (Open Access) Investigação de ocorrência de alterações moleculares nos genes KRAS, HRAS, NRAS e BRAF em carcinoma papilífero da tireóide(Universidade Federal do Pará, 2018-01-25) PIMENTEL, Clebson Pantoja; OLIVEIRA, Edivaldo Herculano Correa de; http://lattes.cnpq.br/0094007714707651The most common cancer of the thyroid is the papillary carcinoma (PTC), which represents 80% of the cases of cancer affecting this gland. PTC is a malignant tumor, with slow evolution, found in any age, but with a higher occurrence in patients between 30-40 years old. The metabolic pathway MAPK is the most associated with PTC. Among the several proteins which have a role in this pathway, we highlight the ones encoded by the genes belonging to families RAS and BRAF. Considering that there are few clinical and genetic studies focusing on thyroid cancer from Brazilian Amazonian region, the aim of this study was to investigate the occurrence of alterations in genes HRAS, NRAS, KRAS and BRAF in patients with PTC treated in a public hospitals, from Belém (PA), seeking to make an association between the mutations found and the biochemical and clinical findings. To achieve this goal, polymerase chain reaction (PCR) and direct automatic sequencing were used. Statistical analyses were performed using the software SPSS version 21.0. Continuous data were expressed as means and standard deviation and categorical data were described in terms of percentages. Student t Test was used to evaluate the continuous variables, while Fisher exact test and Chi-square were used to analyze categorical variables. We considered p<0.05 as significant value in all the analyses. Our results showed that, among the analyzed genes, only BRAF showed a mutation, BRAFV600E, in 21 out of the 53 patients (16 female and 5 males, 39.6%). Additionally, a new mutation in codon 38 of gene K-RAS was found (p.D38E). Considering clinical data, we found a significant association between the BRAFV600E mutation and hoarseness, as well as between this mutation and lymph node metastasis. In addition, the observation of a new mutation in the K-RAS gene indicates that the number of gene changes involving the MAPK pathway is still incomplete. The data obtained can be used for a better pre-surgical evaluation of thyroid tumors, in order to increase the sensitivity for the detection of cancer and avoid unnecessary surgeries of lesions erroneously identified as malignant.Item Acesso aberto (Open Access) Método baseado me cromatografia líquida de alta eficiência para analisar a atividade dos transportadores de GABA no sistema nervoso central(Universidade Federal do Pará, 2022-08) MORAES, Edinaldo Rogério da Silva; SILVA, Anderson Manoel Herculano Oliveira da; http://lattes.cnpq.br/8407177208423247The GATs are the membrane proteins responsible for the uptake of GABA in the central nervous system. Alterations in GAT activity are implicated in several neurological diseases, including retinopathies. The present study describes an alternative method to determine GAT activity in tissue preparations of the central nervous system, using high performance liquid chromatography (HPLC) with fluorescence detection. The GABA concentration in the medium was determined using the o-phthaldehyde (OPA)-derivation protocol validated by the Brazilian Health Regulatory Agency (ANVISA) and the United States Food and Drug Administration (US-FDA). The GAT activity in the retinal preparations was determined through the evaluation of the GABA uptake, which was measured by assessing the difference between the initial and final concentrations of GABA in the incubation médium (Δ𝐺𝑎𝑏𝑎 = [𝐺𝑎𝑏𝑎] 𝑡_0 − [𝐺𝑎𝑏𝑎] 𝑡_𝑒𝑛𝑑). The evaluation of the GAT kinetics returned values of Km = 382.5 ± 32.2 μM and Vmax = 34 nmol/mg of protein. The data also demonstrated that the GABA uptake was predominantly Na+ and temperature-dependent, and was also inhibited by incubation with nipecotic acid, a substrate of GABA transporters. Taken together, these findings confirm that our approach provided a specific measure of GAT activity in retinal tissue. The data presented here thus validate, for the first time, an alternative, simple and sensitive method for the evaluation of GAT activity using high performance chromatography on preparations of the central nervous system.Item Acesso aberto (Open Access) Modulação dos sistemas GABAérgico e glutamatérgico na secreção hipotalâmica de ocitocina sob condições hiperosmóticas(Universidade Federal do Pará, 2014-04-07) GRISÓLIA, Alan Barroso Araújo; SILVA, Anderson Manoel Herculano Oliveira da; http://lattes.cnpq.br/8407177208423247; DINIZ, Domingos Luiz Wanderley Picanço; http://lattes.cnpq.br/9601463988942971In mammals, the osmolality of extracellular fluid is a main factor for maintenance hydro electrolyte balance. Thus, changes in osmolality are detected by specialized hypothalamic cell, thereby starting a neurochemical signaling with glutamatergic and GABAergic system involvement, which may trigger the oxytocin release. However, the way of GABA glutamate aminoacids relationship could modulate the oxytocin release under hyperosmolality is till poorly understood. In this context, the aim in present study was characterize the hypertonic medium effect in GABA glutamate extracellular levels and its relationship with oxytocin release in the hypothalamus in vitro. For this, male wistar rats (270-300g) were kept under standard laboratory conditions. After decapitation, the brain was quickly removed and the hypothalamic fragments were immediately dissected in cold Krebs Ringer Bicarbonate Glucose Buffer (KRBG), and were transferred to perifusion chambers containing KRBG isotonic (280 mOsm/Kg H₂O), at flow rate of 0.5-1.0 ml/min, medium effluent was collected every during 15 min. The hypertonic stimulation (340 mOsm/kg H2O) was performed during 3 minutes. Glutamate, GABA and oxytocin levels were determined by reverse phase high-performance liquid chromatograph (HPLC) system. The measurements of glutamate showed an increased release only after decrease in GABA concentration. This release temporal profile motivated us to add GABA (3 mM) during osmotic stimulation, resulting in blockage of glutamate release previously observed. Moreover the results showed oxtocin release by hypertonic solution may also depend on a GABA decrease. The present study suggests that oxytocin release stimulated by hypertonicity depends on altering the relationship GABA / glutamate.Item Acesso aberto (Open Access) Neurodegeneração crônica em modelo murino: ensaios comportamentais e neuropatológicos na doença prion em fêmeas adultas de camundongos albinos suíços(Universidade Federal do Pará, 2009-04-30) OLIVEIRA, Roseane Borner de; DINIZ, Cristovam Wanderley Picanço; http://lattes.cnpq.br/2014918752636286In the present report we described behavioral and neuropathological changes induced by ME7 prion agent inoculated into CA1 of the albino Swiss mice and confirmed previous descriptions in the murine model of prion disease C57Bl6J with two exceptions: 1) septal region present higher level of microglial activation and reactive astrocytosis 2) disease progression (from inoculation to death) is 4 weeks longer and on average, early behavioral changes start correspondently 4 weeks later in albino Swiss mice. Neuronal counts did not reveal any significant changes between the experimental groups. Comparative analysis of activated microglia and perineuronal nets by optical fractionators revealed significant differences between 15 and 18 weeks: the microglial total number increased in this period of time whereas perineuronal nets decreased (t test, two-tailed analysis p<0.05) Cluster and discriminant subsequent analysis applied to behavioral studies revealed that burrowing activity distinguished the occurrence of two subgroups with differential sensitivity to the ME7 agent: one group (40% of the subjects) where the disease progression is faster and the terminal stage is reached in 22 weeks and another one (60%) with slower progression and terminal stage at 26 weeks post-inoculation. The results are important for comparative studies of the immunoneuropathology of chronic neurodegenerative disorders in general and for prion disease itself.Item Acesso aberto (Open Access) Parâmetros vocais acústicos de sujeitos do sexo masculino antes, durante e após o processo de muda vocal(Universidade Federal do Pará, 2013-04-12) GUIMARÃES, Michelle Ferreira; SILVA FILHO, Manoel da; http://lattes.cnpq.br/2032152778116209The voice and the vocal ability are different in every individual and in every stage of life. There is a limited number of studies on the vocal quality of children and adolescents during puberty. The aim of this study was to quantify the following parameters of acoustic analysis of voice: fundamental frequency, jitter, shimmer, harmonic-to-noise ratio and intensity. To understand the relationship between the voice and the development of children and adolescents, these parameters were correlated with each other and with the degree of pubertal development in male subjects. Methods: The subjects of the study were 110 males, aged between 11 and 20 years, students from three Macapá schools, where the data collection was carried out. The subjects were divided into 4 groups, 32 subjects aged between 11 and 12 years composed the Group I, 29 subjects aged between 13 and 15 the Group II, the Group III consisted of 30 subjects aged between 16 and 18 years and Group IV by subjects aged between 19 and 20 years. All subjects underwent computer-based voice recording using a unidirectional microphone. The following phonation tasks were requested: saying sustained vowel /é/, counting from 1 to 10 and reading a pre-established paragraph. Then, the subjects were assessed by a physician for characterization of pubertal development according to the stages described by Tanner. The voice analysis was performed using the voice acoustic program Vox Metria ®. Results: The subjects showed the following F0 average during sustained vowel: 223.28 Hz, 249.86 Hz, 122.63 Hz and 127.61 Hz for Groups I, II, III and IV respectively. The mean of F0 during the sequential speech was 217.09 Hz, 246.18 Hz, 117.27 Hz and 123.42 for Groups I, II, III and IV respectively. Shimmer showed increased values for the four groups. Jitter, intensity and harmonic noise ratio remained within normal limits established by the acoustic voice program used. Most subjects were in G3 (n = 38, 34.5%) and G4 (n = 42, 38.2%) and P3 (n = 34, 31%) and P4 (n = 36 , 32.7%) stages of pubertal development. The degree of pubertal development was correlated with F0 during speech (p <0.001) and with F0 during sustained vowel (p <0.001) and this correlation was statistically significant between G2 and G5, and G3 and G5. Conclusion: Subjects aging up to 15 years showed acoustic vocal parameters typical of the childish voice. The decrease of F0 occurs between 16 and 20 years old, but the voice is still in process of stabilization, with increased values of shimmer. F0 is the only parameter correlated with the degree of pubertal development. The last stage of voice change, in the studied population, is a late event in relation to pubertal development.Item Acesso aberto (Open Access) Variabilidade alélica e expressão do gene ABCA4 em sujeitos diagnosticados com a maculopatia de Stargardt: associação com a função e estrutura da retina e morfologia celular granulocítica(Universidade Federal do Pará, 2015-06-24) CARVALHO FILHO, Nelson Monte de; SOUZA, Givago da Silva; http://lattes.cnpq.br/5705421011644718; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219The autosomal recessive Stargardt's maculopathy is characterized by symmetrical progressive loss of central vision in patients’ first and second decades of life. It is also part of the clinical diagnosis the presence of yellowish-white color specific macular lesions found in the parafoveal and foveal regions, besides lipofuscin deposition generating hypofluorescence and atrophy of choriocapillaris and retinal pigment epithelium. Visual acuity of the Stargardt’s degeneration carriers decreases with their progress and tend to stabilize between 20/200 (1.0 logMAR) to 20/400 (1.3 logMAR). This thesis aimed to investigate the occurrence of association between allelic variants (mutations) and ABCA4 gene expression levels with the peripheral granulocytes morphology and clinical phenotypes, electrophysiological and retinal structure in patients harboring Stargardt’s maculopathy. Four non-synonymous and three synonymous allelic variants were identified among eleven selected subjects. The allelic combination L1395P/D1817E was found in 64% of the 22 chromosomes analyzed suggesting the occurrence of founder effect. There were no association between genotypes and clinical phenotypes analyzed. The differential values obtained for ABCA4 gene expression between patients and controls, as well as the prevalence of banded neutrophils in the peripheral blood circulation suggest the possibility they could serve as biomarkers for Stargardt. The eye fundus and angiographic images were used for the classification in the stages I, II and III of retinal impairment in subjects investigated. The thickness values of the central region of the macula among patients were much lower than those obtained for the controls, indicating a loss of photoreceptor layer. Full-field electroretinographic findings of light adapted eyes during the course of time enabled the characterization of the functional losses in investigated subjects.
