Dissertação Acesso aberto (Open Access) Ação do metabólito secundário 5-hidroxi-2-hidroximetil gama-pirona isolado de fungos do gênero Aspergillus sobre monócitos humanos in vitro(Universidade Federal do Pará, 2012-06-15) COSTA, Josineide Pantoja da; SILVA, Edilene Oliveira da; http://lattes.cnpq.br/7410116802190343The 5-hydroxy-2- hydroxymethyl-gamma-pyrone (HMP) is a secondary metabolite synthe-sized by some species of fungi from Aspergillus, Penicillium and Acetobacter genera. The HMP has several applications, being used as antioxidant, tyrosinase inhibitor, protective agent against radiation and antitumor. Recently, it was also shown that this metabolite acts as a macrophage activator. However, the effect of HMP in human monocytes is unknown. Thus, the aim of this study was to evaluate the effects of HMP on the cell viability and differentia-tion of human blood monocytes in vitro. Human peripheral leucocytes were obtained from blood bag donated from Fundation Hemocenter of Para State. Cell isolation was performed using HISTOPAQUE® 1077-density-gradient. Monocytes were treated for 24, 48 and 72 hours with 50 and 100 μg/mL of HMP. The ultrastructural analysis of treated monocytes showed spreading ability, high number of cytoplasmatic projections and vacuoles, features that are often observed in activating cells. Immunofluorescence analysis of the expression of surface protein specific for the macrophage (F4/80), demonstrated that human monocytes treated with 50 and 100 μg/mL for 48 and 72 h showed the similar pattern of expression of proteins to that of human monocytes differentiated by macrophage colony-stimulating factor (M-CFS). The viability test used showed that HMP has no citotoxicity effect on human mon-ocytes when treated with 50 and 100 μg/mL of HMP. These results demonstrate a new role for HMP as an immunomodulator agent, inducing the differentiation of monocytes into macrophages.Dissertação Acesso aberto (Open Access) Análise morfológica in vitro da ação de antifúngicos em cepas de Fonsecaea pedrosoi(Universidade Federal do Pará, 2014-02-21) MASSOUD JUNIOR, Heleno Ramos; SALGADO, Claudio Guedes; http://lattes.cnpq.br/2310734509396125Choromoblastomycosis (CBM) is a disease caused by traumatic implantation of many species of melanized fungi. The State of Pará is the major endemic area in Brazil and Fonsecaea pedrosoi is the major etiological agent. The treatment is not standardized and many forms of interventions are related in the literature. In the other hand, the in vitro susceptibility test to antifungal drugs may help in the therapeutic choice and in the identification of resistant strains. The objective of this work is to evaluate the in vitro susceptibility of 20 F. pedrosoi clinical isolates to itraconazole (ITZ), ketoconazole (KCZ), fluconazole (FCZ) and terbinafine (TBF) as well as the possible morphological alterations induced by ITZ or TBF in the Minimal Inhibitory Concentration (MIC) and high concentrations. The tests were performed according to the Clinical and Laboratory Standards Institute (CLSI, M38-A2 document) recommendations. The final concentrations of ITZ, TBF and KCZ in each test were to 16 to 0.03 μg/mL. To FCZ the final concentrations were to 64 to 0.125 μg/mL. The MIC was defined as the lowest drug concentration that inhibit 100% the visual growth when compared to the non-treated group after five days of incubation at 30°C. ITZ proved to be the most effective drug in vitro against F. pedrosoi (CIM 90= 1μg/mL). TBF showed a low drug activity with 70% of the isolates with MIC ≥ 0.5 μg/mL. The conidia morphological analysis revealed an increasing in the diameter, an interruption of the cellular division and the formation of little chains after the treatment with ITZ in the MIC. At the high concentration used in the susceptibility test we noticed an irregular shape, a detachment of pigmented material from the cell wall and a vacuolization. Rupture in cell wall and amorphous conidia were observed at 32 μg/mL and 64 μg/mL. Significant alterations were not observed after treatment with TBF at the same concentrations. Moreover, the 5-fluorocytocise (5-FC) and FCZ do not stop the conidia growth at high concentrations. However, ultrastructure alterations were noticed after treatment with 5-FC 64 μg/mL. Thus, it is suggested a different morphological pattern after ITZ or TBF treatment during the in vitro susceptibility test. In synthesis, ITZ shown better in vitro antifungal activity while 5-FC only provoked structures alterations in the highest concentration tested.Dissertação Acesso aberto (Open Access) Avaliação da susceptibilidade de camundongos BALB/c e Swiss, hamster e Proechimys roberti à infecção por Leishmania (Viannia) Naiffi e Leishmania (Viannia) Lindenbergi(Universidade Federal do Pará, 2005) SODRÉ, Roberta Nice Salgado; ISHIKAWA, Edna Aoba Yassui; http://lattes.cnpq.br/3074963539505872Leishmania (Viannia) naiffi and Leishmania (Viannia) lindenbergi are species that cause cutaneous leishmaniasis in Amazonia and present great similarity in its isoenzymatic profile, monoclonals antibodies and production of unapparent infection in hamsters. The fact of not having a highly susceptible experimental model to the infection for L. (V.) naiffi and L. (V.) lindenbergi, the objective of this study was evaluate the susceptibility of BALB/c and Swiss mice, hamsters and Proechimys roberti to the infection for those two species. It was prepared inoculums with salivary glands and without glands for each group of animals, associated to the promastigotes. The experimental animals, of both sexes, were inoculated intradermally in the dorsal surface of back feet and they were observed for 90 days. In the period of 30, 60 and 90 days after inoculation, the animals were sacrificed and different fragments of skin of the inoculation place were used in the culture, microscopic exam and polymerase chain reaction (PCR). It was not possible to observe lesions in the animals inoculated with L. (V.) naiffi and L. (V.) lindenbergi even at the presence or absence of salivary glands. As well as, forms amastigotes during 30, 60 and 90 days after the inoculation. In the culture, all the animals inoculated with L. (V.) lindenbergi haven’t developed promastigotes. For the other hand, BALB/c mice inoculated with L. (V.) naiffi presented positively when sacrificed 30 days after inoculation. PCR presented low sensibility compared to the culture. This way, we concluded that L. (V.) naiffi and L. (V.) lindenbergi is species that present low infectivity and none of the animals used in the study experimental model can be considered highly susceptible to the infection for those two species.Dissertação Acesso aberto (Open Access) Avaliação do efeito protetor da prolactina em linfócitos expostos a ação do metilmercúrio(Universidade Federal do Pará, 2012-05-23) JESUS, Maria Izabel de; DINIZ, Domingos Luiz Wanderley Picanço; http://lattes.cnpq.br/9601463988942971Mercury can be found in several forms, as being organic methylmercury (MeHg), considered the most toxic. Readily absorbed orally, accumulates in the food chain and is amplified in aquatic carnivores, especially in fish, hence the greater risk to populations that preferentially feed on them, such as riparian Amazonian populations. The neurotoxic effect of this form of mercury has been widely demonstrated by epidemiological and experimental studies. Some of these studies have also shown that hormones and antioxidants may act by protecting the body against the deleterious effects of mercury. Prolactin is a hormone that has such protective action, but also acts as a proinflammatory cytokine. Since MeHg can also act as an immunotoxic substance, we have studied the cytoprotective action of PRL in continuous cultures of strain B95-A primate lymphocytes in order to assess their vulnerability to MeHg and its responsiveness to the action of PRL. In order to assess the functional integrity of lymphocytes exposed to MeHg we used to test color reaction for 3 - (4,5-dimethylthiazol-2-yl) -2,5-diphenyl tetrazolium bromide (MTT), which detects activity mitochondrial metabolism. To evaluate the immune response of lymphocytes, measures of tumor necrosis factor alpha (TNF α) concentration in the middle were performed by ELISA. It is a proinflammatory cytokine released in response to cellular injury from different causes, including oxidative stress, one of the most obvious acute effects of MeHg, and this cytokine also be able to answer prolactin regulation in human lymphocytes. After 18 hours of cultivation exposure to increasing concentrations of the metal (0.1, 1, 5, 10 and 50 mM) showed significant decrease in dose-dependent cell viability from 1 mM (35%) and progressively up to 50 mM (80%), when few intact cells were found in the cultivation. A biphasic effect in a "bell" shaped occurred in the release of TNF-α, where lower concentrations of MeHg inhibited (0.1 and 1 mM) stimulated the intermediate (5 mM) and the two largest (10 and 50 mM) returned to inhibit. Prolactin also decreased the cell viability by about 30% only at the highest dose (10 nM).Moreover, at a dose of 1 nM prevented PRL 40% decrease in cell viability due to exposure to 5 mM MeHg. This dose of 1 nM PRL was the only one to stimulate the release of TNF-α, but curiously reversed the release of this cytokine when combined with 5 mM of MeHg, concentrations that also stimulated the secretion of TNF-α. The results confirmed the toxicity of MeHg to lymphocytes of primates (strain B95-A) and its reversion by possible protective action of PRL. A biphasic effect on the secretion of TNF α resulted from MeHg exposure, suggesting the presence of different mechanisms of cytotoxic action resulting from mercury. Moreover, PRL was less effective in stimulating the secretion of that cytokine, reversing this response when the associated with MeHg. For the effectiveness and efficiency of this method are met, it is necessary for the establishment and application of standard rules of execution and reading of slides. Thus, it is essential to know the quality of smear microscopy available in a laboratory network. The aim of this study was to evaluate the quality of sputum for diagnosis and control of TB conducted in Units Laboratory (UL) belonging to the public laboratories in Belém, Pará, in 2007, 2008 and 2009. The evaluation was divided into three stages, corresponding to the year of manufacture of the sputum blades. Participants UL 18 that sent regularly to the Central Laboratory of Para, all smear slides prepared by them for External Quality Assessment of Smear (AEQB), which uses the method of blind rereading of smears. This method involves a technical evaluation of the smears, describing them as "Adequate" and "Inappropriate", besides the analysis of discrepant results False Positive (FP) and false negative (FN). During the study period, 4117 samples were processed by UL. In the first stage of evaluation, all 18 UL had blades that were classified as inadequate, and this number decreased to 12 UL in the 2nd stage of evaluation and UL for six at the 3rd step. Regarding disagreements, disagreements had 03 UL 03 UL FP and FN. The results obtained in this study reinforce the need to maintain and expand AEQB for all UL performing smear in the network of laboratories.Tese Acesso aberto (Open Access) Biodiversidade bacteriana durante a fermentação espontânea de frutos de açaí (Euterpe oleracea)(Universidade Federal do Pará, 2015-06-22) MOURA, Fábio Gomes; ROGEZ, Hervé Louis Ghislain; http://lattes.cnpq.br/5202118426597590Açai (Euterpe oleracea) fruits (AF) and a wide variety of products derived from this fruit are an example of success of a local and typically Brazilian product on the global market. The AF suffer spontaneous fermentation during postharvest due mainly to its high microbial load and the favourable intrinsic and extrinsic parameters. The biodiversity of microbial community in AF and their evolution between three geographical origins and two fermentation conditions were examined. Culture-independent methods based on 16S rRNA from fifteen samples revealed that Proteobacteria, Firmicutes, Actinobacteria, Bacteroidetes and Acidobacteria were the most abundant phyla. At genus level, were identified Massilia (taxon with more than 50 % of sequences remaining constant during the 30 h of fermentation), Pantoea (taxon with the highest increase during fermentation), Naxibacter, Enterobacter, Raoultella and Klebsiella, forming the carposphere bacterial microbiota of AF. Attributes related to plant growth promotion (siderophores) and others compounds e.g. poly-3-hydroxybutyrate (PHB) and violacein in response to stress conditions could improve the Massilia preponderance. Beta diversity showed that quality parameters of AF (pH, soluble solids, titratable acididy and lipids) and elemental analysis (C, N, H and C/N ratio) were unable to establish microbial patterns in AF. This research offers new insights and perspectives of the indigenous bacterial community composition on AF as a function of spontaneous fermentation during postharvest.Tese Acesso aberto (Open Access) Caracterização da resposta imune citocínica na infecção humana pelo vírus oropouche e sua relação com o padrão de soroconversão e a presença de sintomas(Universidade Federal do Pará, 2011-12-19) OLIVEIRA, Euzébio de; VASCONCELOS, Pedro Fernando da Costa; http://lattes.cnpq.br/0973550817356564This thesis is the first global study that researches and analyzes the immune response of cytokine in human infections by Orthobunyavirus Oropuche virus. The study used 320 samples of human serum. Sixty were from the Blood Bank (negative control) and 260 were obtained from two outbreaks of the Oropouche virus in the State of Pará and Amapá (Brazil). The latter was divided into 8 subgroups for better data accuracy. The collected samples were analyzed for clinical data/symptoms with serologic testing by titration of antibodies by the hemagglutination inhibition (IgM/IgG) and the detection cytokines plasma levels by flow cytometry. This allowed for the technical description of cytokine. The data obtained allowed for the observation of the characteristics and the behavior of the cytokines signatures expressed by patients by the presence or not of the virus. This also allowed for the observation of changes to serum through the presence of specific symptoms such as fever, chills, headache and dizziness. This led to the following conclusions a) there is a pattern in the synthesis of pro-inflammatory and regulatory cytokines; b) there is a balance in the profile of the immune response between pro-inflammatory cytokines (Th1) and modulators (Th17); c) an infection by the Oropouche virus alters the production of cytokines in individuals; d) the results also show that whem comparing individuals no responders with early responders, there was an increase of IL-1β and decreased IL- 12; no responders with late responders, there was a decrease of IL-8, and increased IFN-α, IL-23 and IL-17; No responders occurred early responders compared with the increase IL-4 and IFN-g; However, when compared early responders and late responders, decreased IFN-α and IL-6; early responders generally showed a decrease in IL-10 and late responders showed an increase in IL-5; e) The results also show the expression of IL-5 in patients who showed symptoms specific for Oropouche infection (fever, chills, headache and dizziness), suggesting this signal to be directly associated with pathogenesis of the virus; f) there is a need to complement this research with more studies such as those related to the expression of chemokines.Dissertação Acesso aberto (Open Access) Cinética da infecção pelo arbovírus piry em modelo murino: a resposta do hospedeiro adulto(Universidade Federal do Pará, 2011-09-30) SANTOS, Zaire Alves dos; DINIZ, Cristovam Wanderley Picanço; http://lattes.cnpq.br/2014918752636286In the present report, a member of a group of RNA South American viruses found in Brazil, that causes febrile disease in humans and encephalitis in neonate and adult murine models, was selected as a model to study encephalitis outcomes in adult albino Swiss mice. In mice housed under standard conditions with free access to water and food, we induced viral encephalitis by intranasal inoculation of Piry virus–infected brain homogenate and correlated neuropathological features. We quantified the cellular inflammatory response in the septal region using a stereologically based unbiased method with clinical signs and neuroinvasion, comparing the outcomes with those of animals inoculated with uninfected brain homogenate. Three-month-old female mice maintained in standard environment received an equal volume of Piry virus infected or normal brain homogenates into the nostrils. From the 1st to 8th days post-instillation (dpi), five subjects from the infected colony were fixed and processed to detect viral antigens and microglia. Control subjects were sacrificed in the 5th dpi and processed for the same markers. After Piry virus encephalitis induced microglial activation and neuroinvasion of glial cells and neurons mainly in the olfactory pathways early in the disease (2 – 4 dpi), but also included hippocampus, cerebellum and brain stem nuclei later on (5 - 8 dpi). The correlation of the host cellular inflammatory quantitative response in the septal area with clinical signs and neuroinvasion, revealed that the number and the morphology of microglias changed early in the disease before neuroinvasion had reached the septal region and clinical signs had appeared. Great variability in clinical symptoms intensity and survival rate were found in the outbred albino Swiss mice strain as compared with previous report in the inbred C57Bl6 strain suggesting less isogenic background. Taken together, our previous and present report dedicated to investigate Piry virus encephalitis progression in the outbred albino Swiss mice strain may open a new field of investigation of the genetics, anatomical and immune substrates of tropical sublethal arbovirus encepahlitis.Dissertação Acesso aberto (Open Access) Determinação de alguns parâmetros da biologia reprodutiva e produtiva da paca fêmea (Agouti paca Linnaeus, 1744) criada em cativeiro(Universidade Federal do Pará, 2002) LAMEIRA, Ana Paula Guimarães; GUIMARÃES, Diva Anelie de Araújo; http://lattes.cnpq.br/2891287458034896; OHASHI, Otávio Mitio; http://lattes.cnpq.br/5547874183666459This work had the objective of estimating basics reproductive characteristics of Agouti paca reared in captivity, the determination of the oestrous cycle period, gestational period, parturition interval, first postparturition oestrous, number of offsprings and sexual proportion. The observations were done using a colony of 13 caged animais in the Biotério of Universidade Federal do Pará, and the data were obtained with colpocitological techniques. These results indicate that this specie is polyestrous with continous reproduction. The oestrous cycle was categorized in four phases: proestrous, oestrous, metaestrous and diestrous, and its period was 32,5 ± 3,69 days (n = 20). As for the gestation period, two results were obtained: a) 147, 5 ± 2,83 days (n = 2) for females that had its last oestrous registered with sperm presence and, b) 146,7 ± 6,43 days (n = 3) for females that had just the last oestrous before the parturition. New borns were observed and weights and sexes were registered. Their weight was 605,9 ± 87,47 g (n = 12) for females and 736,7 ± 108,41 g (n = 14) for males. At birth the youngs are quite developed and soon show active movements, opened eyes, body completely covered by hair, and a capacity to eat solids within two days. From 38 births occurred at Biotério, all of them are single, although the A paca is been able to produce more than one offsprings per parturition. The parturition interval was 187,3 ± 8,48 days (n = 15) and within 35,6 ± 5,22 days (n = 5) occurred the first oestrous postparturition. These results can be used as basic orientation to future works with proposes to study reproductive parameters from selvage animals using biothecnology. In addition to, the application of an appropiate management in A. Paca creation, can become in the future, aliment qualified source and lucrative source too. Eight newborn mice two days old of the species Mus musculus and Swiss albino variety were inoculated intracerebrally with brain homogenate infected with Flavivirus species (DENV3 genotype III). Another group of animals was used as control (uninfected) and inoculated with an equal volume of non-infectious brain homogenate and maintained under the same conditions of those infected. After 7 days after infection the mice were sacrificed and patients have had their brains processed for immunostaining of astrocytes and microglia. We quantified the glial and astrocitic immune response in the stratum lacunosum molecular (Lac Mol), radiatum (Rad) and pyramidale (Pir) of hippocampus and in the stratum molecular of dentate gyrus (DGMol) using the optical ractionator to estimate the number of microglias and astrocytes in the hippocampus of infected and control animals. Intense reactive astrocytosis and microglial activation were associated with clinical signs of meningoencephalitis in neonate subjects. Although the number of activated microglia to be higher in infected than in control subjects in the GDMol (Inf:738,95 } 83,07 vs Cont: 232,73 } 70,38; p = 0,0035), Rad (Inf: 392,49 } 44,13 vs Cont: 62,76 } 15,86; p = 0,0004) the number of total microglias (activated or not) was different only in the stratum radiatum (Inf: 6.187,49 } 291,62; Cont: 4.011,89 } 509,73;p = 0,01). On the other hand the total number of astrocytes was higher in control than in infected subjects only in the DGMol (Inf: 8.720,17 } 903,11; Cont: 13.023,13 }1.192,14; p = 0,02). Taken together the results suggest that the immune innate response in neonate mice after encephalitis induced by DENV3 genotype III is associated with a higher increase in the activated microglias than astrocytes in a regional and laminardependent fashion. This mycosis occurs predominantly in Amazon region and affect all populations groups. Histologically, there is a chronic inflammatory reaction characterized by the intense fibroplastia and histiocytosis, abundant number of macrophages, multinucleated giant cells and presence of large numbers of yeast. Macrophages are phagocytic cells that participate in the recognition and response to pathogens through phagocytosis, antigen presentation to T lymphocytes and cytokine production. Langerhans cells (LC) are a group of dendritic cells (DC) derived from bone marrow, which are located mainly in the suprabasal layer of the epidermis. Studies on hostfungal interaction in Jorge Lobo's disease are scarce. Therefore, this study constitutes an important step towards a better understanding of L. loboi biology and pathogenesis, enabling in vitro culture of this organism endemic in Amazon region and study of their antigenic properties, as well as new therapeutic approaches. The objective of this study was to analyze the in vitro interaction between non-activated peritoneal macrophages and/or Langerhans cells isolated from BALB/c mice and L. loboi, calculating infection, phagocytosis and cellular fusion rates; also measuring the production of TNF, IL-4, IL-6, IL-10 and IL-12. Results showed that L. loboi is phagocytosed by macrophages but not by LC. Phagocytosis and infection rates in the interaction between macrophages and L. loboi was similar to interaction between macrophages, Langerhans cells and L. loboi at all times analyzed. The mean number of yeast cell per macrophage was almost equal among interactions and over time, with mean variation from 1.2 to 1.6 yeast/macrophages. No giant cells formation in coculture studied. No significant difference occurred in IL-4, Il-2 and IL-10 production between interactions studied. TNF levels decrease in interaction between macrophages and L. loboi while adding LC induces increased production of TNF-α, especially after 48 hours. LC negatively modulate IL-6 production by macrophage and L. loboi also inhibits this production by macrophages alone or cocultured with LC. L. loboi stimulated significantly IL-12 production by macrophages co-cultured with LC, but not for LC alone or macrophages.Tese Acesso aberto (Open Access) Papilloma vírus humano: prevalência, distribuição e importância como fator preditivo para metástases linfonodais em carcinoma do pênis(Universidade Federal do Pará, 2013-07-09) FONSECA, Aluizio Gonçalves da; BURBANO, Rommel Mario Rodriguéz; http://lattes.cnpq.br/4362051219348099The etiology of squamous cell carcinoma of the penis (CEP) is related to multiple risk factors such as the presence of foreskin, poor hygiene, chronic dermatitis and smoking. However, the risk factor most extensively studied, is infection by human papilloma virus (HPV). It is well established that this plays an important etiologic role in cancers of the uterine cervix, however, its association with CEP has been the focus of debate, demonstrating viral presence ranging from 15% to 80% of cases, suggesting that only a subset of these tumors is caused by HPV. The prognosis is negatively influenced mainly by the presence of metastases in inguinal lymph nodes. Thus, the surgical aproach of them, is of vital importance in curing the disease. On the other hand, the available methods for staging these nodes are imprecise and lymphadenectomy are accompanied by significant morbidity. A description of other histological markers is scarce because of the rarity of these tumors. The objective of this study is to evaluate the prevalence, distribution and association between HPV and histological parameters of poor prognosis, in order to determine their possible predictive value for inguinal metastases, as well as prognostic factors already described. Paraffin tumor specimens from 82 patients with squamous cell carcinoma of the penis were tested for prevalence and genotype distribution of HPV by PCR. The HPV status was correlated with histopathological factors and metastatic inguinal. It was also evaluated the influence of various histological tumor factors, in lymphonodal disease free survival at 5 years. Follow-up ranged from 1 to 71 months (mean 20 months). HPV DNA was detected in 60.9% of the sample having the most prevalent viral type 11 and 6 (64% and 32%, respectively). There was no significant correlation independent of the histological variables of poor prognosis with HPV status. The probability of lymphonodal disease free survival at 5 years, was not affected by HPV (log rank test = 0.45). The only independent factors for pathological inguinal metastases were T stage ≥ T1b-T4 (p = 0.02), lymphovascular invasion (p = 0.04) and infiltrative invasive front (p=0, 03). Both EBV and HP are classified are class 1 carcinogens by the World Health Organization, and a substantial number of individuals become co-infected in adulthood. These two pathogens may have synergic potential to cause perpetual chronic gastritis. The purpose of this paper was to verify the prevalence of HP and Epstein-Barr virus in children and adolescents. Material and Methods: Transversal descriptive study. The gastric mucosa of 64 children and adolescents was analyzed through the Urease Test to diagnose HP and the PCR technique to detect H. pylori’s cagA strain, the in situ hybridization technique to detect EBV and the pathological analysis to determine the histopathological characteristics. Results: The prevalence of HP and EBV found by this study was 53.1% and 3.1, respectively. Most of the patients infected by HP (94.3%) presented gastritis in the upper gastrointestinal endoscopy, with enanthemathous gastritis being the most commonly found type. In the histopathological analysis, most patients (97.1%) presented some level of gastritis, 80% of which classified as moderate chronic gastritis. Positive cagA strains were found in 64.7% of the patients infected with HP and all of them had gastritis, with predominance of moderate chronic gastritis (54%); however, there was no statistically significant correlation between these findings. There was also no statistically significant association between infection by HP and EBV in the studied population. The low prevalence of EBV in this analysis suggests that this virus is not an etiological agent in gastric mucosa lesions. To our knowledge, this is the first study that relates these two infectious agents in the gastric mucosa of children and adolescents in northern Brazil. Gastric carcinoma is among the few malignant neoplasms in which infectious agents play an important role. The aim of the present work was to research the prevalence and the association of Helicobacter pylori and Epstein-Barr virus infection in gastric adenocarcinoma in a northern population of Brazil. A hundred twenty-five samples of gastric adenocarcinoma were analyzed by PCR to detect H. pylori and pathogenic genotype H. pylori-cagA+, by in situ hybridization to detect EBV, and by histopathological analysis to determine epidemiologic and clinico-pathological data. It was observed a higher frequency in male patients (68%) as much as older patients (78%). Prevalence to H. pylori was 88%, and it was considered high when compared to early studies in northern region of Brasil. To EBV the prevalence was 9,6%. Patients H. pylori-cagA+ showed increased relative risk to intestinal type adenocarcinoma. The case’s frequency to III and IV stages of the disease was 82,4%, demonstrating that the diagnostic to this neoplasia has been done late. The urease positive cases presented a higher than four-fold relative risk (OR=4,231) to H. pylori-cagA+, the more pathogenic genotype. The water quality supplied to the population can be evaluated by physical, chemical and microbiological parameters. This study aimed to evaluate the increase in knowledge water quality for human consumption in Brazil using the scientometric method. A systematic search for information was carried out in the Scielo, Scopus, Web of Science and Google academic databases. We seek to evaluate the physical, chemical and microbiological water quality for human consumption in Brazil through a systematic literature review. We obtained 843 concatenated articles, of which 62 articles met inclusion criteria. There was an investigation of the water quality in 10 sources of water abstraction, 20 numbers of species of microorganisms were classified, being 15 species of bacteria and 6 of protozoa, were Escherichia coli, Salmonella spp, and for protozoa Giardia spp. were the most frequent. The variables that most favor the environment for the emergence of pathogenic microorganisms found in the studies were: Dissolved Oxygen (DO), hydrogenic potential (pH), Nephelometric Turbidity Unit (UNT), Electrical Conductivity (EC), Temperature (T) and Demand Oxygen Biochemistry (BOD). The Northeast region presented variables with higher values than expected, as well as the highest number of publications. Although we have detected an increase in the number of studies, some questions such as demographic aspects, socioeconomic conditions and environmental health still remain unanswered and are important questions to be considered in future studies.
