This study aimed to evaluate these limitations using a quantitative approach in leprosy patients in Belém (Pará, Brazil). Methods: This epidemiological, cross-sectional study measured the sensory impairment of smell and taste through the use of a questionnaire and evaluated activity limitations of daily life imposed by leprosy through the Screening of Activity Limitation and Safety Awareness (SALSA) Scale. Data were collected from 84 patients and associations between the degree of disability and clinical and epidemiological characteristics were assessed. Results: The majority of patients were men (64.3%), married (52.4%), age 31-40 years old (26.2%), had primary education (50%), and were independent laborers (36.9%). The multibacillary operational classification (81%), borderline clinical form (57.1%), and 0 degrees of physical disability (41.7%) were predominant. SALSA scores ranged from 17 to 59 points, and being without limitations was predominant (53.6%). The risk awareness score ranged from 0 to 8, with a score of 0 (no awareness of risk) being the most common (56%). Evaluation of smell and taste sensory sensitivities revealed that 70.2% did not experience these sensory changes. Patients with leprosy reactions were 7 times more likely to develop activity limitations, and those who had physical disabilities were approximately four times more likely to develop a clinical picture of activity limitations. Conclusions: Most patients showed no sensory changes, but patients with leprosy reactions were significantly more likely to develop activity limitations. Finally, further studies should be performed, assessing a higher number of patients to confirm the present results.Tese Acesso aberto (Open Access) Associação entre marcadores da resposta inflamatória e a imunopatogênese de agentes infecciosos de natureza viral (Vírus da dengue, HTLV-1 e HTLV-2) e bacteriana (Chlamydia trachomatis e Chlamydia pneumoniae)(Universidade Federal do Pará, 2010-11-29) FEITOSA, Rosimar Neris Martins; ISHAK, Ricardo; http://lattes.cnpq.br/5621101706909450The genetic basis of diseases is frequently studied aiming the polymorphisms of cytocine genes. The present study investigated markers of the inflammatory response associated to the course of infection and disease caused by viruses and bacteria. Serum levels (measured by an ELISA assay) and the polymorphisms (using PCR, RFLP and allele specific PCR) of TNF-α (-308), TNF-β (+252), IFN-γ (+874) and C reactive protein were measured among persons with febrile disease, infected by dengue virus (n=80), not infected by DV (100), a group of HTLV infected (30 symptomatic and 47 asymptomatic), a group with coronary disease (58 seroreactive to Chlamydia and 31 with negative serology) and a control group (99 persons with no reaction to DV, HTLV and Chlamydia). No group showed association with demographic informations. Dengue virus 3 (66.2%) and HTLV-1 (90% symptomatic and 76.6% asymptomatic persons) were the most frequent agents found among their groups. The majority of those with coronary disease (65.1%) presented antibodies to Chlamydia (39.6% to C. trachomatis and C. pneumoniae, 58.6% solely to C. trachomatis and 1.7% to C. pneumoniae). Statistically significant levels of differences were found among: (i) serum levels of TNF-β, IFN-γ and PrtCR of positive and negative dengue and control groups (p< 0,01); (ii) serum levels of TNF-α, TNF-β and IFN-γ of HTLV (including its types) and control groups; (iii) serum levels of TNF-α, TNF-β, IFN-γ and PrtCR among patients with coronary disease, serum reactive to Chlamydia, and the control group; (iv) the presence of antibodies to C. trachomatis and C. pneumoniae and the control group comparing TNF-β, IFN-γ and PrtCR. Genotypic frequency distributions were statistically significant for the polymorphisms: (i) of TNF-α (p=0,0494) and IFN-γ (p= 0,0008) genes among positive, negative and control dengue groups and to IFN-γ (p= 0,0007) among groups DEN 1, DEN 2, DEN 3 and controls; (ii) of IFN-γ gene (p= 0,0023) among the group of patients with coronary disease and sero reactivity to C. trachomatis e C. pneumoniae, as well as to the mono reactants in the comparison between the positivity to C. trachomatis and the control group.Dissertação Acesso aberto (Open Access) Caracterização antigênica, físico-química e biológica do vírus BE AR 701405, obtido a partir de mosquitos da espécie Psorophora (Jan) ferox, capturados em Altamira – Pará(Universidade Federal do Pará, 2014-03-26) ARAÚJO, João Batista dos Santos; LEAL, Élcio de Souza; http://lattes.cnpq.br/1158983666415285The BE AR 701405 virus was isolated from a pool of Psorophora (Jan.) ferox mosquitoes captured in the municipality of Altamira, Para state, Northern Brazil, in 2006. The objective of this study was the physicochemical, biological and antigenic characterization of the BE AR 701405 virus in order to provide data for it taxonomic classification. Newborn mice inoculated by intracerebral route showed evidences of neurological manifestations, such as chill and motor disorders after the infection with the virus BE AR 701405. Cytophatic effect (CPE) was in infected Aedes albopictus C6/36 cells was not clearly observed, however in VERO cells CPE was observed after 48 hors post infection. The virus load was determined in 10-4,1 LD50/ 0.02 mL and the titer after DCE analysis was calculated in 10−2,6 LD50/0,02 mL. The BE AR 701405 virus reacted antigenically with Pixuna virus hiperimmune serum. In conclusion, the studied vírus was sensible to the DCA solvent suggesting that it is an enveloped virus. Furthermore, by serology this virus was identified as a member of the group A, genus Alphavirus, family Togaviridae, more closely related to Pixuna virus. In addition, newborn mice, as well as C6/36 and VERO cells demonstrated to be sensible to the infection by the BE AR 701405 vírus. Further studies are needed to better understand the antigenic relationship between the BE AR 701405 virus and Pixuna virus.Dissertação Acesso aberto (Open Access) Caracterização das infecções por norovírus nas hospitalizações pediátricas por gastrenterite na cidade de Belém, Pará(Universidade Federal do Pará, 2012-04-20) SIQUEIRA, Jones Anderson Monteiro; GABBAY, Yvone Benchimol; http://lattes.cnpq.br/1579859438466504The Norovirus (NoVs), Caliciviridae family, is related with acute gastroenteritis (AGE) in people of all ages groups. Its importance as cause of outbreaks have been confirmed, which occur mainly indoors. Its transmission is mainly by the fecal-oral route through contaminated water and food, and person to person contact. The disease usually is characterized by diarrhea, vomiting, nausea and abdominal cramps. The aim of this study was to demonstrate the importance of NoVs as pathogen associated with hospital admissions of children with AGE in Belém, Pará. The collection of fecal specimens occurred from May 2008 to April 2011, being only tested the samples with negative results for rotavirus. The enzyme immunoassay (EIA) and the reverse transcription-polymerase chain reaction (RT-PCR) were used for NoVs detection. The samples with positive results by EIA and negative by RT-PCR, were submitted to the semi-nested RT-PCR, and the ones with remain negative, were tested by the real-time PCR. A total of 483 samples were analyzed with a positivity of 35.4% (171/483). Adopting the RT-PCR as the reference method, the EIA had a sensitivity of 85.9% and a specificity of 93.4% with excellent reproducibility between them (kappa = 0.8, p <0.0001). The 22 samples positive only by EIA were tested first by semi-nested RT-PCR and after by real-time PCR, with a positivity of 63.6% (14/22) and 75% (6/8), respectively. The partial nucleotide sequencing of ORF1 region demonstrated the presence of GII.4d (80.8%-42/52), GII.7 (7.7%-4/52) and GII.b (11.5%-6/52) genotypes. Sequencing was performed in 64.3% (9/14) of the samples positive only by semi-nested RT-PCR, also corresponding to ORF1, which 55.6% (5/9) were classified as GII.4d and 44.4% (4/9) as GII.b. Of the six samples classified as GII.b, five were characterized as GII.3 when sequenced with primers specifics for the capsid region, suggesting the possibility of recombinants samples. A higher infection rate was observed in children under 2 years of age (90.1%-154/171) and the main symptoms were vomiting (95.8%-137/143) and dehydration (94.4%-118/25), considering the diarrhea was an inclusion criterion. Most infected children had more than 9 days of diarrhea (41.2%), 4 evacuations per day (43.9%) and more than 5 episodes of vomiting (90%) during hospitalization. Regarding seasonality, three peaks of positivity were observed in September and October 2008 (63.6%), and in February 2010 (62.1%). Any correlation with the climatic parameters of rainfall, humidity and temperature was demonstrated. The current work determined the complete small RNA segment and partial medium RNA segment nucleotide sequences for group C members. The full-length SRNA sequences ranged from 915 to 926 nucleotides in length, and revealed similar organization in comparison with other orthobunyaviruses. Based on the 705 nt of the N gene, group C members were distributed into 3 major phylogenetic groups, with the exception of Madrid virus that was placed outside of these 3 groups. Analysis of the Caraparu virus strain BeH 5546 revealed that it has an SRNA sequence nearly identical to that of Oriboca virus and is a natural reassortant virus. In addition, analysis of 345 nucleotides of the Gn gene for seven group C viruses and for strain BeH 5546 revealed a different phylogenetic topology, suggesting a reassortment pattern among them. These findings represent the first evidence for natural reassortment among the group C viruses, which include several human pathogens. In case of reactivation of the virus, the herpes zoster may occur. Analysis of the presence of IgG and IgM is critical to determine the prevalence of this virus in the Metropolitan Region of Belém. The study of specific nucleotide polymorphisms is used to define the genotypes of VZV. Analysis of ORFs 22, 38 and 54 identified genotypes of VZV according to the classification established in conference July 25, 2008 in Whitechapel, London / UK, where the strains of VZV detected and characterized by sequencing of SNPs were grouped into classes 1 through 5. To evaluate the prevalence of antibodies and describe the circulating genotypes was the aim of this study. The frequency of IgM and IgG antibodies in cases of chickenpox was 68.2% and 48.2%, respectively. Cases of herpes zoster showed prevalence of anti-VZV IgG and IgM of 87.5% and 12.5%, respectively. The genotypes 1 or 3 and 5 were present in 13 samples sequenced, and the European strain (class 1 or 3) was found in samples from all the cities studied. The ecologic aspected and clinic pictures are related with phylogeny and evolution of flaviviruses. This work aims the molecular characterization of Bussuquara (BSQV), Iguape (IGUV), Ilheus (VILH) e Rocio (VROC) flaviviruses genomes, determining their phylogenetic relationships with others members of genus Flavivirus. The study included: the full-length sequencing of the four Brazilian flaviviruses; analyzes of the predictive secondary structure of RNA and conserved sequences in the 3’NCR; determination of cleavage and glicosilation sites, cisteine residues and conserved motifs in the polyprotein; and similarity and phylogenetic analyzes. The BSQV, IGUV, VILH and VROC genomes present 10815 nt, 10922 nt, 10775 nt, and 10794 nt, respectively. The conserved standard sequences in 3’NCR of BSQV was RCS2-CS2-CS1, while to IGUV, ILHV and ROCV were CS3-RCS2-CS2-CS1. The secondary structure of RNA obtained for the Brazilian flaviviruses were similar to the other flaviviruses. The numbers of the glicosilation sites to PrM, E and NS1 proteins were distinct among the studied Brazilian flaviviruses, therefore the pattern 6,12,12 Cis residues and the cleavage sites were conserved. In the E protein, some singles mutations were observed in fusion peptide of BSQV, IGUV and ROCV, and the RGD motif were distinct for the flaviviruses under study. The motif that determines the MTase-SAM activity in NS5, as well as the helicase and protease activity in NS3 were conserved. Among the eight polimerase motifs in NS5, only the V, VI and VII motifs were observed single mutations in ILHV and ROCV. The similarity analyzes showed that BSQV presents high relationship with VIGU, while ILHV and ROV were more related among themselves, however those viruses were considerated distincts species. Based in the phylogenetic analyzes, molecular and biological characteristics, it was proposed the establishment of three distinct genetic groups: the Rocio group, grouping ILHV and ROCV, Bussuquara group formed by BSQV and Naranjal virus; and Aroa group, that include Aroa virus and IGUV.Dissertação Acesso aberto (Open Access) Caracterização morfológica e antigênica do vírus Juruaçá, isolado de morcego no estado do Pará(Universidade Federal do Pará, 2006) ARAÚJO, Tais Pinheiro de; VASCONCELOS, Pedro Fernando da Costa; http://lattes.cnpq.br/0973550817356564Juruaçá virus (BE AN 401933) was isolated from pooled organs of an unidentified bat captured during field work in Porto Trombetas, Oriximiná, Pará State, in 1982, and remains unclassified/ungrouped. The aims of this work were to classify Juruaçá virus in a viral taxon taking in account its morphological, physicochemical, antigenic and molecular properties, as well as, to describe the pathological alterations. This agent is pathogenic only for infant mice, and the animal’s death occurs with eight days post-inoculation. It’s not related to any of the arthropod borne viruses with which it has been tested by serological tests, such as complement fixation (CF) and hemagglutination inhibition (HI). Positive reactions were only observed with its homologous serum. Juruaçá virus is not sensitive to sodium desoxicolate and can hemagglutinate goose cells at pH 5.75. The virus didn’t show any cytophatic effect and immunofluorescence was negative in Vero and C6/36 cell lines, but it replicates in brain tissue primary culture cell line (astrocytes and microglias), confirmed by immunofluorescent assay (IFA). Culture of neuronal cells appears not to be infected by Juruaçá virus; however, infection of these cells was confirmed by imunohistochemistry. By transmission electron microscopy and negative stain, the virus is a spherical particle, with mean diameter of 23-30nm. Pathological alterations were observed mainly in the central nervous system of newborn mice experimentally infected with Juruaçá virus. The molecular results of RT-PCR suggest that Juruaçá virus is a possible new virus belonging to the family Picornaviridae, genus Enterovirus.Dissertação Acesso aberto (Open Access) Detecção e genotipagem de norovírus em diferentes amostras de água e esgoto não tratado na cidade de Belém, Pará, Brasil, 2008 a 2010(Universidade Federal do Pará, 2014) TEIXEIRA, Dielle Monteiro; GABBAY, Yvone Benchimol; http://lattes.cnpq.br/1579859438466504Enteric viruses excreted in feces from infected individuals dispersed in aquatic environments by sewage discharge. Among these viruses, the norovirus (NoV) is actually considered the main cause of gastroenteritis outbreaks worldwide, resulting from the ingestion of contaminated food and water as well as is also associated with hospitalizations. This research aimed to detect and partially characterize the human NoV (GI/GII) in different water matrices and in untreated sewage from Metropolitan Region of Belem. The study involved superficial waters from bay (Ver-o-Peso), river (Acai’s Port), stream (Tucunduba) and two lakes (Bolonha and Agua Preta), as well as treated water (WTP-Bolonha) and untreated sewage (SLP-UNA), monthly collected over two years . The water and sewage (2 liters) were initially concentrated on filtering membranes to obtain a final volume of 2 mL. The nucleic acid was extracted by silica method and submitted to semi nested RT-PCR (reverse transcription Polymerase chain reaction) using NoV GI and GII specific primers. The cDNA obtained after reverse transcription was also used to investigate the GI/GII by TaqMan® real time PCR. The positive samples for both molecular methods were analyzed for 5’end ORF2 by nested (for GI) and semi nested (for GII) in order to obtain amplicon for identification of circulating strains, being further purified using a commercial kit and submitted to molecular characterization in the automated sequencer. The obtained sequences were edited, aligned and compared to others available in gene bank (NCBI) and in the site NoV genotyping tool. In the period of November 2008 to October 2010, 168 water and sewage samples were collected and analyzed for NoV presence, obtaining a positivity of 33.9% (57/168) of which 21.1% (12/57) were positive only by TaqMan® real time PCR, 19.3% (11/57) only by semi nested and 59.6% (34/57) for both. Considering the two methodologies used, in the positive cases GI (82.5% - 47/57) was most frequent than GII (79.0% - 45/57). However, in most samples there was coexistence of the two genogroups (61.4% - 35/57), mainly in the Tucunduba and SLP-UNA samples, considered the most NoV contaminated sites. On the other hand, in WTP-Bolonha this agent was not found. Of 57 positive samples by TaqMan® real time PCR and/or semi nested RT-PCR, 53 were retested for 5’end ORF2, since four samples showed insufficient quantity of material which allowed a new analyze, so, in 47.2% (25/53) the NoV genome was detected, of these 12% (3/25) belonging to GI, 24% (6/25) to GII and 64% (16/25) for both. The most frequent GI and GII genotypes were GI.8 (n=8) and GII.4 (n=12), respectively, but others genotypes were also observed with lower incidence as GII.6 (n=3), GII.9 (n=2), GII.12 (n=1), GII.14 (n=1), GI.1 (n=1) and GI.4 (n=2). Due to low quality of sequences obtained, eight samples could not be genotyped for GI and three for GII. Of 96 samples with concentration of thermotolerant coliforms above the recommended, 34 (35.4%) were also NoV positive. Increase on conductivity and total dissolved solids was observed in materials from Ver-o-Peso and Tucunduba, as well as the turbidity was notably higher in these places and the Acai’s Port. In the less rainy period (July to November) there was a trend in positivity increasing for NoV, and in the highest rainfall (December to June) a decrease in the incidence of this agent was noted. The results obtained in the present study indicate the circulation of NoV GI and GII in aquatic environments in Belem, revealing the degradation that these water bodies have suffered, as a result of poverty or lack of sanitation in our city, allowing the permanence of pathogens in these ecosystems, along with its effluents.Artigo de Periódico Acesso aberto (Open Access) Development and characterization of a packaging cell line for pseudo-infectious yellow fever virus particle generation(Universidade Federal do Pará, 2018-02) QUEIROZ, Sabrina Ribeiro de Almeida; SILVA JÚNIOR, José Valter Joaquim; SILVA, Andrea Nazare Monteiro Rangel da; OLIVEIRA, Amanda Gomes de; SANTOS, Jefferson José da Silva; GIL, Laura Helena Vega GonzalesIntroduction: Pseudo-infectious yellow fever viral particles (YFV-PIVs) have been used to study vaccines and viral packaging. Here, we report the development of a packaging cell line, which expresses the YFV prM/E proteins. Methods: HEK293 cells were transfected with YFV prM/E and C (84 nt) genes to generate HEK293-YFV-PrM/E-opt. The cells were evaluated for their ability to express the heterologous proteins and to package the replicon repYFV-17D-LucIRES, generating YFV-PIVs. Results: The expression of prM/E proteins was confirmed, and the cell line trans-packaged the replicon for recovery of a reporter for the YFV-PIVs. The HTLV 1 shows a bigger occurrence in south west of Japan, Caribbean, Central America, in different regions of South America and in parts of Central, as well as in western Africa, while HTLV-2, seems to assault distinct groups of people, such as Indians native people from North, Central and South America, Pigmies from Central Africa, Mongols in Asia as well as in infecting drug users. The present work had as objective describing HTLV molecular epidemiology, in three different populations the State of Amapá, they are: HIV/IDS positive patients, Afro-descenig population and individuals assisted of Public Health Center Laboratory of Amapá – LACEN-AP, directed for diagnosis of HTLV. The samples were tested for the presence of virus using serological (ELISA and Western blot) and molecular assays (gene amplification and restriction fragment length polymorphism from pX and env for the analysis of polimorfismo o restriction fragments for endonuclease action. The obtained results in different populations are In the population of HIV infected people, all the samples were negative; in the Afro-desceding population, only one sample was positive confirmed by serological test (ELISA), but negative according to western blot test and submitted to the molecular analysis, there wast not amplification. However, among samples of individuals directed for diagnosis of HTLV, 06 (six) were positive, 5(five) out of 6/them were confirmed by western blot test. In literature There are few studies on experimental neuropathology Marabá virus induced, despite 30 years of isolation. A single study, however, revealed that viral infection in newborn mice causes necrosis and pyknosis in neurons in several regions of the central nervous system (CNS). The objective of this study was to investigate the distribution of Marabá virus in the CNS, reactive microglia and astrocytes, histopathologic features, and the expression of cytokines and nitric oxide (NO), in Marabá virus-induced encephalitis in BALB / c mice. Thus was performed processing of samples for histopathological examination; Immunohistochemistry for observation of microglia, astrocytes and viral antigen; tests for quantification of cytokines and NO, and statistical analyzes. The results showed that infected animals (Ai) 3 days after inoculation (d.p.i.) with discrete labeling of viral antigen, as well as the activation of microglia and astrocytes in the CNS. Moreover , in Ai 6 d.p.i. marking the viral antigen was observed in almost all brain regions, with intense microglial activation in these locations, although was less astrogliosis. Edema, necrosis and apoptosis of neurons were mainly observed in the olfactory bulb, septum and frontal cortex of Ai 6 d.p.i. Quantification of IL - 12p40, IL- 10, IL- 6, TNF- α, INF- ү, MCP-1 and NO, showed significant increases in Ai 6 d.p.i., when compared with controls animals and Ai 3 d.p.i. On the other hand, TGF-β, important immunosuppressant, has not been significant in all groups and evaluated times (3 and 6 d.p.i.). The present study investigated the association between the Mbl gene polymorphism and the susceptibility to HIV-1 infection. A fragment of 349 bp from the exon 1 of the Mbl gene was amplified by PCR and then submitted to RFLP analysis using the endonucleases BanI and MboII, aiming the identification of the variant alleles. The study of 145 seropositive patients and 99 healthy controls showed the presence of alleles Mbl-*A, Mbl-*B and Mbl-*D, with frequencies of 69%, 22% and 9% among patients and 70.2%, 13.6% and 16.2% among healthy controls, respectively. The analysis of the genotype frequencies showed a high prevalence of the genotypes carriers of variant Mbl-*B among patients seropositive as compared to the healthy controls. Furthermore, the genotype B/B was six times more frequent among patients than the observed to the healthy controls (χ2=4.042; p=0.044). The mean viral load was lower in HIV-1 seropositive patient carrying the Mbl-*A allele than those carrying the variant Mbl-*B allele (5,821 copies/mL vs. 52,253 copies/mL; p= 0.05). Furthermore, patients carrying the allele Mbl-*A showed a significant reduction of the viral load (p<0.001), that was not observed among those carrying the variant Mbl-*B (p=0.999). One of the objectives of this study was to describe the prevalence of infections HIV-1, HTLV1/2, VHB and CMV in pregnant adolescents, attended in a Reference Center in the State of Pará. To achieve the objectives, it was collected blood samples from 324 pregnant adolescents, who came from several cities of the State, from November of 2009 to February of 2010. The samples were submitted to an immuno-enzymatic test (ELISA), in order to detect antibodies Anti- HIV, Anti- HTL V1/2, Anti- VHB and Anti- CMV IgM/ IgG. The serological analysis revealed one serum positive sample for HIV-1, two positives samples for HTLV1/2, while the majority of samples showed antibodies Anti-CMV-IgG, although the occurrence of high infection was low (2.2%). The prevalence of infection for VHB in pregnant adolescents was 0.62%, however a great number (83.3%) of adolescents are susceptible to the infection by VHB, what means they probably were not immunized. The majority of adolescents (63.4%) continued their studies, even though they know about their pregnancy and 34.6% started prenatal later, so they only had a minimum number of four consultations of prenatal. The result reinforces the hypothesis that pregnant adolescents have prevalence for those viral type infections, what is similar to the national pregnant women rates. Because older animals have less functional decline in adaptive immunity T cells, we tested the hypothesis that AE old mice would show higher number of deaths and more intense clinical signs than age-matched IE animals, and this would be associated with greater expansion of T lymphocytes. To test these hypotheses we established scheme of multiple inoculations in adult animals of 9 and 18 months of age. Two regimens of inoculation were tested: multiple injections of single serotype (VDEN3 genotype III) infected brain homogenate (SS) or alternatively multiple injections with that infected brain homogenate followed 24h later by inoculation of heterologous antibody (SSHA). In both cases multiple i.p. inoculations were done. It was found significant differences in the temporal progression of the disease in the animals submitted to one or another scheme of inoculation: SSHA group (Kaplan -Meier log-rank test, p = 0.0025); IUS (Kaplan -Meier log-rank test, p = 0.089). The survival curves of AE and AP under SSHA regime were extended after a single injection of glucocorticoids, reducing the symptoms and the number of deaths, and these effects were greater in the EE group than in the IE (Kaplan-Meier log-rank test, p = 0.0162). In SSHA scheme, EE group showed clinical signs more intense than the AP and those included dyspnea, tremor, hunched posture, immobility, pre-terminal paralysis, shock and eventual death. Compared to the IE group, the AE group regardless of age showed higher mortality and more severe clinical signs. These more severe clinical signs in EE animals under SSHA regime were associated with increased hyperplasia of T lymphocytes in the spleen and increased infiltration of these cells in the liver, lungs and kidneys. Although lymphocytic hyperplasia and infiltration have been more intense in older than in younger animals, immunostaining for viral antigens in target organs was higher in young than in the aged mice. The presence of the virus in various infected organs were confirmed by real time PCR. Taken together the results suggest that the enriched environment life style exacerbates the subsequent inflammatory response to infection, and that is associated with more severe clinical symptoms, higher mortality and increased T cell expansion. However, a few is known about the Central Nervous System (CNS) response against amazonic rhabdovirus. The virus Itacaiunas e Curionopolis were classified preliminary as members of the Rhabdoviridae family. It was suggested for these virus to be included in a new genus, Bracorhabdovirus. They are neurotropic virus and little is known about the inflammatory response of the CNS against these virus. This study examined aspects of the immune response of newborn mice CNS resident cell after experimental infection by Itacaiunas and Curionopolis virus, in vivo and in vitro. To achieve these goals, histochemical and immunohistochemical techniques were used to detect activated microglia and astrocytes, respectively, in sections of the brains of infected mice. The quantification of these cells were performed in hippocampal regions using stereological techniques. Furthermore, was investigated the expression of proinflammatory and anti-inflammatory cytokines produced by CNS cells in primary cultures infected with virus Itacaiunas and Curionopolis by using enzyme immunoassay (ELISA) from supernatants co-cultures enriched of microglia/neurons and astrocytes/neurons. The expression of nitric oxide was also analyzed by using cytochemical reagent DAF-FM diacetate (4-amino-5-methylamine-2,7-difluorofluorescein-diacetate). As a result, age matched control animals of each group did not present activated microglia or astrocytosis, as well as, both Curionopolis and Itacaiunas infected subjects early after the inoculation (2d.a.i. and 4d.a.i respectively). However at 5th d.a.i., Curionopolis infections significant increase in the number of activated microglias and reactive astrocytes were detected whereas at the 7th d.a.i. after Itacaiunas infection only minimal activation of microglia and reactive astrocytosis was detected. The results from immunoassays showed great production of IL-12p40 in cultures of CNS cells infected with both virus, 48 and 96 hours after infection (h.a.i.). Low levels of TGF-β and IL-10 were detected after 48 hours of infection with both virus, however, was observed increased expression of this cytokines after 96 hours. CNS cell cultures infected with Curionopolis virus 48h.p.i. and Itacaiunas virus 48 and 96h.a.i. showed a slight production of nitric oxide, but it was increased in cultures infected with Curionopolis virus 96h.a.i. The samples were inoculated in RD and HEp-2 cells line, and the positive were identified by neutralization test or indirect immunofluorescence. Out of 249 samples, 33 (13.2%) were positive 57.6% (n = 19) in patients under 11 years of age (p <0.03) and predominantly (72.7%) in males (p <0.008). The serotypes isolated were: Echovirus 30 (n = 31), Coxsackievirus B5 (n = 1) and Echovirus 30 and 4. RSV has been associated with annual respiratory disease outbreaks and bacterial co-infection has also been reported. This study is the first RSV study in young children hospitalized with community-acquired pneumonia (CAP) in Belém city, Pará (Northern Brazil). It had the objective of determining the prevalence of RSV infection and evaluating the patients’ clinical and epidemiological features. Methods. We conducted a prospective study across eight hospitals from November 2006 to October 2007. In this study, 1,050 nasopharyngeal aspirate samples were obtained from hospitalized children up to the age of three years with CAP, and tested for RSV antigen by direct immunofluorescence assay and by Reverse Transcription Polymerase Chain Reaction (RT-PCR) for RSV subtype identification. Levels of C-reactive protein (CRP) and results of bacterial infection were also obtained. Results. RSV infection was diagnosed in 243 (23.1%) children. The mean age of the RSV-positive group was lower than the RSV-negative group (12.1 months vs 15.5 months, both ranged 1-36 months, p<0.001) whereas gender distribution was similar. The RSV-positive group showed lower CRP mean levels when compared to the RSV-negative group (15.3 vs 24.0 mg/dL, p<0.05). Radiological findings showed that 54.2% of RSV-positive group and 50.3% of RSV-negative group had interstitial infiltrate. Bacterial infection was identified predominantly in the RSV-positive group (10% vs 4.5%, p<0.05). Rhinorrhea and nasal obstruction were predominantly observed in the RSV-positive group. A co-circulation of subtypes A and B was noted, with a predominance of subtype B (209/227). Multivariate analysis revealed that age under 1 year (p<0.015), CRP levels under 48 mg/dL (p<0.001) and bacterial co-infection (p<0.032) were independently associated with the presence of RSV as opposed to RSV-negative group, and in analyze of symptoms, nasal obstruction were independently associated with RSV-positive group (p<0.001). Conclusion. The infections have universal distribution. Many studies have demonstrated the strong association of infection by species of high-risk cervical cancer. Therefore, this study sought to determine the prevalence of HPV infection in a group of women screened for cervical cancer. From january 2008 to december 2009 were collected cervical samples from 180 women seen at the Laboratory of Cytopathology of Universidade Federal do Pará (UFPA), with 162 eligible for the study. Each participant was collected two samples, one for the manufacture of cytological smears for subsequent analysis by the Papanicolaou method and the other for analysis of molecular biology, in order to investigate the presence of HPV, using the primers MY09 and MY11. The species of HPV have been identified by PCR sequencing of nucleotide bases of the L1 ORF of HPV. The analysis of the epidemiological profile of the study group showed that the average age accounted for 37.5 years. The majority (49.38%) were married, 37.65% had initiated sexual activity between 13 and 17 years old and 58.64% did not use condoms during sex. The overall prevalence of HPV was found to be 18.52%. Thirteen different types were identified, with the majority (66.67%) was a low-risk group, in which HPV-11 was more frequent. Among the high risk group (30%) HPV-31 was found in four cases. The species distribution of HPV according to the cytology showed that the prevalence of infection was higher in the group of women whose smear was free of abnormal cytology (43.33%). In the group of women with premalignant changes only observed infections by species of low risk (10%). HPV infection was significantly associated with sexual activity and the frequency in the completion of screening. The prevalence in this study corroborates other findings in the medical literature. The prevalence of infection in women with normal cytology reinforces the idea that the infection is mostly asymptomatic and the Papanicolaou method is less efficient in detecting the infection than the techniques of molecular biology. Other factors also affect the variability of the env gene: for example function of neutralizing antibodies - Nabs (FROST et al., 2005), the variation in glycosylation sites; (LEAL et al, 2012 LEAL et al., 2008), binding to target cell receptors (i.e, CD4, CXCR4, CCR5), and also the switch from CCR5 to CXCR4 receptor (MILD et al., 2013). Thus, the relationship between viral levels diversification and CD4 + T cells in the env gene can be circumstantial. The vif gene, on the other hand, it is retained and not subject to bias present in the env gene (i.e, glycosylation sites, etc.). Thus, to study the influence of CD4 + T cells levels in HIV variability, the estimate of the selective regimes was used (Nonsynonymous Substitutions - dN and Synonymous Substitutions - dS) by a codon-based model and phylogenetic analysis of unrelated individuals (inter-host). HIV-1 sequences were used of the not-correlated individuals, obtained from the Los Alamos Database. The sequences were separated into CD4+ levels of categories and then analyzed. The analysis revealed no direct correlation between CD4+ T cell levels and evolutionary rates in gp120 and vif gene from HIV-1, subtypes B and C in a population approach.
