Tese Acesso aberto (Open Access) Análise comparativa da alteração dimensional de materiais de moldagem elastoméricos, através da técnica do casquete(Universidade Estadual de Campinas, 2000-09-18) ANDRADE, Cícero Almeida de; SILVA, Frederico Andrade e; http://lattes.cnpq.br/2282864112869742The present study assessed comparatively the dimensional change of four fluid-consistence elastomeric impression materiais (one addition silicone, two condensation silicones and one mercaptan) using the headcap technique. A metal cast (pattern) was made, enclosing two teeth with metal-crown preparation. On this cast, with a 0,3-mm metal relief, were obtained the headcaps using Neelon's technique (brush method). The impressions were carried out with a static-weight press, where a strength of 39.2 N (newton) was pressing the trays over the metal cast. The occlusal surfaces of the prepared teeth had cross-wise grooves. In the extreme of each groove there were small points where the reading started. The linear reading, in a comparing microscope, was initially done at the occlusal surface of each teeth distinctly, so following it was read matching the points between the two teeth. The biological behavior of the lesions is distinct because the ameloblastoma is more aggressive and significant rate of tumor recurrence. CCOT is a less aggressive tumor and recurrence rarely there and therefore was used as a control in the study. Therefore, the complete elucidation of the mechanisms by which these odontogenic tumors show such biological behavior remains a challenge for researchers. The ADAMTS (A Disintegrin and Metalloproteinase with thrombospondin) are metalloendopeptidases who are dependent on zinc in its catalytic domain. These enzymes have catalytic activity against a broad range of substrates including proteoglycans (aggrecan, brevican and versican), which are proteins present in the extracellular matrix (ECM). The ADAMTS exhibit structural features that confer great potential to display multiple functions. Exhibit crucial role in various processes such as proliferation, adhesion, invasion and cell signaling. Changes to these enzymes are present in various tumors, suggesting that these proteins may be involved in the carcinogenic process in different ways. Specifically, ADAMTS-1 has been correlated with tumorigenesis of some cancers such as in breast, lung and pancreatic cancer. Like ADAMTS, aggrecan, versican and brevican are expressed in various tumors and altered regulation of proteoglycans may contribute to the development of carcinogenesis. In this work ADAMTS-1, aggrecan, brevican and versican in ameloblastoma and CCOT were studied, 20 cases of ameloblastoma and 6 cases of TOCC, used as controls were included. We evaluated the expression of ADAMTS-1, aggrecan, brevican and versican by immunohistochemical study and the marking areas were measured and analyzed. To correlation analysis between the studied proteins used the Spearman test. All samples of ameloblastoma expressed ADAMTS-1, aggrecan, brevican and versican. All samples TOCC also expressed the same proteins, but in significantly less than the amount ameloblastoma. The difference in expression of ADAMTS-1 and brevican in the epithelium of ameloblastoma and of TOCC was statistically significant (p<0.0105). As the expression of aggrecan and versican, between ameloblastoma and TOCC, in the epithelium was also statistically significant (p<0.0067) and (p<0.0148), respectively. There was no correlation between the proteins studied.Tese Acesso aberto (Open Access) Comparação do planejamento em implantodontia por radiografia panorâmica e tomografia computadorizada de feixe cônico(São Leopoldo Mandic, 2017-03-09) ANDRADE, Wagner Almeida de; JUNQUEIRA, José Luiz CintraThe aim of this research was to evaluate the intra-examiner agreement in the planning of implant planning with panoramic radiography and cone beam computed tomography (CBCT). The research was a retrospective study of the 34 patients, in which panoramic radiography and CBCT were evaluated. The sample used in this study was 76 edentulous sites, with one or multiple spaces in different regions of the jaws, which performed the installation of implants. Inclusion criteria were patients attend in the period from 2013 to 2015, with panoramic radiography and CBCT, with the clinical planning, and discussed between students and teachers before the surgical procedure. The follow up of the patients who did not attend the inclusion were excluded. The radiographic and tomographic planning was compared with clinical planing. The experts answered a questionnaire with the indication of the area to be planned and type of implant, according to an interactive catalog. The data were analyzed in a descriptive statistic, and the intraclass correlation coefficient (ICC) was applied to evaluate the similarity between the measurements of the width and length of the implants by the computerized planning and the initial planning. The level of significance was 5%. The results showed that in the radiographic planning, the evaluation of the reproducibility using the CCI, the observed values were considered from average to good in the posterior maxillary. In tomographic planning, the anterior maxillary height coincided with the initial surgical planning; In the evaluation of the plans of the posterior maxillary and height of the posterior mandible, the CCI found were considered satisfactory. It was concluded that there were intra-examiner disagreements in the planning of the initial and tomographic implants. Smoking and alcohol are the main causes of oral cancer and larynx, but a part of the population develops the disease without being exposed to these risk factors, suggesting the existence of other causes such as genetic predisposition, alteration of tumor suppressor genes, diet and viral agents, particularly human papillomavirus (HPV) and Epstein-Barr virus (EBV). The proposition of this study was to verify prevalence of HPV and EBV in normal oral mucosa, cancer of oral cavity and larynx, and what types are most prevalent in these two situations. For this study we established two groups: one consisting of 70 specimens embedded in paraffin, with a confirmed diagnosis of oral and larynx cancer and other with 166 individuals without lesions in the oral cavity. Laboratory analysis for the viral detection and typing HPV and EBV (EBV type 1 or 2) were performed by conventional PCR (Polymerase Chain Reaction). The typing of samples positive for HPV types (6, 11, 16, 18, 33, 35, 38, 52 and 58) was performed by real time PCR using probes specific for each type. The EBV and HPV prevalence found in the oral and laryngeal cancers was 78.6% for HPV and 84.3% for EBV and 24.1% and 45.8% for EBV and HPV, respectively, in individuals without oral lesions. The most prevalent HPV types were HPV 58 (50.9%), HPV 6 (9.1%) and HPV 16 (9.1%) in cancer group and HPV 18 (12.5%), HPV 6 (7.5 %) and HPV 58 (2.5%) in the group with no lesions. The EBV 2 was more prevalent in both the cancer lesions than subjects without lesions, with a frequency of 94.9% and 82.9%, respectively. There was no association of HPV infection with EBV and sex, the prevalence being similar for men and women. Association was observed between the prevalence of HPV and EBV and its co-infections with the group that developed cancer. The prevalence of HPV and EBV and the odds ratio in the occurrence of cancer was 8.86 (p <0.0001) in individuals infected with HPV and 4.08 (p = 0.0004) in patients infected by EBV. The probability value estimated for prevalence of HPV and EBV co-infection and the occurrence of cancer has shown that individuals infected by both viruses have 65.72% likelihood of developing cancer, while infected with HPV has 31.94% and infected with EBV 17.79%. The results of this study may suggest that viral agents (HPV and EBV) are important risk factors for the development of carcinogenesis, and HPV is more effective than EBV in triggering the disease.Dissertação Acesso aberto (Open Access) Expressão de metalotioneína e sua influência no comportamento biológico in vitro do carcinoma mucoepidermoide(Universidade Federal do Pará, 2017-02-22) AQUIME, João Rafael Habib Souza; PINHEIRO, João de Jesus Viana; http://lattes.cnpq.br/1365260779826770The Mucoepidermoid Carcinoma (MEC) is the most common tumor of salivary glands, presenting considerable rates of recurrence and distant metastasis due to high invasive capacity of tumor cells, probably favored by metallothionein (MT) actuation, an zinc storage protein that supply this element to protease activity and synthesis of proteins and nucleic acids. So, the aim of this paper was to characterize a cell line from MEC and to correlate expression of MT with the Transforming Growth Factor-α (TGF-α), Tumor Necrosis Factor-α (TNF-α) and Matrix Metalloproteinases (MMPs). Indirect Immunofluorescence assay was realized to detect expression to epithelial and mesenchymal markers. Besides, it was realized cytogenetic analysis to verify cellular chromosomal alterations. The metallothionein2A (MT2A) gene silencing was obtained by small-interference RNA (siRNA) assay, and then, western blot technique was performed to correlate the expression of growth factors and MMPs studied with MT-2A silencing. Indirect immunofluorescence revealed expression to cytokeratines 19 and AE1/AE3, fibronectin, vimentin andα-smooth muscle actin. Cytogenetic evaluation demonstrated several structural and numerical alterations, among which the translocation t(11;19) (q21;p13), characteristic of MEC. After siRNA assay, a decreased expression of TGF-α and MMP-9 was visualized, while TNF-α became more expressed and MMP-2 kept its expression unaltered. Our findings suggest that MT presents important role in tumor invasion mechanism in MEC, because it interferes with expression of proteins directly involved in this process.Dissertação Acesso aberto (Open Access) Perfil clínico-epidemiológico de saúde bucal em pacientes de hanseníase(Universidade Federal do Pará, 2009) SANTIAGO, Roberta Souza D'Almeida Couto; BARROSO, Regina Fátima Feio; http://lattes.cnpq.br/6485426116453902; SALGADO, Claudio Guedes; http://lattes.cnpq.br/2310734509396125The aim of this study was to evaluate the clinical-epidemiological profile of oral health in leprosy patients at multidrug terapy (MDT) through the oral mucosa clinic assessment, the VPI (visible plaque index), GBI (gum bleeding index), DMFTI (decayed, missing and filled teeth index) and salivary flow rate measure by SSI (salivary secretion index) as epidemiological indicators, well over buffeing capacity by salivary pH. The sample was 80 patients, aged from 8 to 73 years submited to the analysis of the oral conditions and stimulated saliva collection, divided in study group (SG) constituted of 40 leprosy patients in MDT carried in the CRTDS Dr. Marcello Candia Marituba-Pa, Brazil and control group (CG) of 40 no leprosy patients. The results pointed 62,5% unspecific oral lesions, among the most frequent was melanin pigmentation of the attached gingiva, mordiscatum line of the buccal mucosa, gingival inflammation, tongue varicose veins and trauma for use of prosthesis, without statistical significant difference (p=0,14) with the CG. The average of the VPI was 50,4 % of the dental surfaces with accumulation of bacterial plaque, without statistical significant difference (p=0,40) with the CG. The VPI was bigger in the individuals without access to the odontologic service and while it increased the degree of incapacity of the patients, juvenile was the access to the service. The average of the GBI was 29,7% of the dental surfaces with gingival bleeding, without statistical significant difference (p=0,35) with the CG. The middle DMFTI was of 11,6 varying from 4,0 to 24, which increases with the advancement of the age. The biggest accumulation of plaque, the gingival bleeding and the consumption of foods cariogenic out of the time-table of the meal also contributed to the increase of the DMFTI. Another factor made a list to the increase of I DMFTI was no access it to the odontologic service for 70 % of the leprosy patients, having statistical significant difference is (p=0,0005) with the CG and the lack of education of oral hygiene in 60 %, having statistical significant difference (p=0,01) with the CG. The middle SSI was of 0,9ml/min and did not present association with the MDT and not even with the use of prednisolone. The SG patients, 25 % presented hiposalivation, but there was no increase of DMFTI and none of the groups revealed alteration of salivary pH , varying from 5,85 to 7,34, with buffeing capacity inside the standard of normality. Conclusion that the clinical-epidemiological profile of leprosy patients is similar to the control group, do not have find specific oral lesions for leprosy, which does not cancel the possibility of the buccal cavity to be a fountain of infection for leprosy, needing histopathological confirmation and / or PCR for detection of viable M.leprae. Material and method: A total of 99 patients who wore partial or total acrylic resin-based upper dentures were included in this study. The subjects completed an epidemiological data form that includes the patient’s gender, age, local factors (hygiene habits, remove denture to sleep, use of mouthwash, present condition of the denture, age of the denture) and functional factors (vertical dimension at rest, vertical dimension of occlusion, occlusion, retention, and static and dynamic stability). To detect yeasts, samples were collected from the inner surface of the dentures and from the palatal mucosa in contact with it. Subsequently, the samples were cultured on Sabouraud dextrose agar, observing macro and microscopic characteristics. Result: In the present study, we did not find any significant relationship between the gender and disease onset. Based on the Newton classification, 36.3% of the patients presented with DS and 89.0% were colonized by yeasts; of these subjects, 50% had type I lesions, 33.3% had type II lesions, and 16.6% had type III lesions. All of the qualitative and local factors, except the use of mouthwash, were clinically relevant to the development of disease. Conclusion: Denture stomatitis in denture users in northern Brazil was multifactorial, involving local, functional and microbiological factors.
