This strain is interesting in the sense that it has been found together with non-tuberculous mycobacteria (NTMs) which could nevertheless play a role in the lesion formation. In this work, we describe a set of features of C. pseudotuberculosis P54B96, together with the details of the complete genome sequence and annotation. The genome comprises of 2.34 Mbp long, single circular genome with 2,084 protein-coding genes, 12 rRNA, 49 tRNA and 62 pseudogenes and a G+C content of 52.19%. To present the whole-genome sequence, a de novo assembly approach using 33 million short (25-bp) mate-paired SOLiD reads only was applied. Differential identification of these bacteria in the clinical microbiology laboratory by the most commonly used biochemical tests is challenging, and normally requires additional molecular methods. Herein, we present the annotated draft genome sequences of two isolates of “difficult-to-identify” human-pathogenic corynebacterial species: C. xerosis and C. minutissimum. The genome sequences of ca. 2.7 Mbp, with a mean number of 2,580 protein en coding genes, were also compared with the publicly available genome sequences of strains of C. amycolatum and C. striatum. These results will aid the exploration of novel biochemical reactions to improve existing identification tests as well as the development of more accurate molecular identification methods through detection of species-specific target genes for isolate’s identification or drug susceptibility profiling.Item Acesso aberto (Open Access) Ecological studies of mayflies (Insecta, Ephemeroptera): can sampling effort be reduced without losing essential taxonomic and ecological information?(Instituto Nacional de Pesquisas da Amazônia, 2018-06) SHIMANO, Yulie; MATOS, Mylena Neves Cardoso; JUEN, LeandroThe present study evaluated the potential for the reduction of sampling effort in studies of ephemeropteran nymphs in Brazilian Amazon streams, Pará State, Brazil, without the loss of ecological information (species composition, abundance, and richness), and the congruence of different levels of taxonomic resolution (morphospecies, genus, family and functional group). Test groups of 15, 10 and five subsamples were selected from the 20 subsamples collected per stream (40 streams sampled), and were compared in terms of their species richness and abundance (ANOVA), and composition (Procrustes). Taxonomic resolution was also analyzed in Procrustes. Species abundance (F(3, 156) = 25.426; p < 0.001) and richness (F(3, 156) = 13.866, p < 0.001) varied significantly among sample groups, while the results of the 15-S group were statistically similar, in both cases, to those of the 20-S group. A similar pattern was found for species composition. The genus-level taxonomic resolution produced results 99% similar to those found for the species-level data. The results indicate that the reduction in sampling effort from 20 to 15 subsamples per site and a genus-level taxonomic resolution would not affect the reliability of analyses significantly. A reduction of five samples per site would result in a decrease of effort in the field and the amount of material to be processed, reducing laboratory time. In addition to a reduction in the time and resources needed to identify specimens, the adoption of a genus-level taxonomic resolution could help minimize errors of under- or over-estimation in the processing of the results.Item Acesso aberto (Open Access) Effects of marginal vegetation removal on Odonata communities(Associação Brasileira de Limnologia, 2013-03) CARVALHO, Fernando Geraldo de; PINTO, Nelson Silva; OLIVEIRA JUNIOR, José Max Barbosa de; JUEN, LeandroAIM: Here we assess the effects of habitat degradation on individuals of the two suborders of Odonata community of Borecaia river sub-basin. More specifically, we tested the hypothesis that Anisoptera richness would be positively affected by removal of vegetation; on the other hand, Zygoptera richness would be adversely affected by virtue of their ecophysiological requirements; METHODS: We selected 10 streams of similar orders, six preserved and four degraded. Streams characterized as preserved had values of Index of Habitat Integrity (HII) above 0.70 (0.77 ± 0.07, mean ± SD) and continuous forest on both sides with a minimum width of 70 meters. Each site was sampled three times on different days. The effect of vegetation removal on richness was assessed using richness estimated by first order Jackknife; RESULTS: Decreased physical integrity (measured with IIH) of streams had no significant effect on the estimated richness to Odonata in general. However, the estimated richness of Anisoptera showed an inverse relationship with the integrity (r2 = 0.485, P = 0.025), i.e., there was a reduction in their species richness with increasing integrity; DISCUSSION: As a general pattern, Anisoptera presents higher richness in an altered site; on the other hand, Zygoptera presents higher richness in a preserved one. This pattern suggests that Odonata needs to be considered at the sub-order level to access the effects of habitat degradation on these insects. Because of its restrictions ecophysiological Odonata varied widely in their composition and species richness between the two types of environments, it reinforces the potential of the order of studies and environmental monitoring also shows that Zygoptera be more affected by changes in habitat. However, further studies including more samples and different streams are need to confirm this pattern, being an interesting line of research for future works.Item Acesso aberto (Open Access) Fatores de risco cardiovascular e fatores associados em escolares do Município de Belém, Pará, Brasil(Escola Nacional de Saúde Pública Sergio Arouca, Fundação Oswaldo Cruz, 2014-03) RIBAS, Simone Augusta; SILVA, Luiz Carlos Santana daThis cross-sectional study aimed to identify risk factors for cardiovascular disease in a stratified cluster sample of 557 schoolchildren (6-19 years) in Belém, Pará State, Brazil. Potential risk factors were obesity, hypertension, dyslipidemia, diabetes, smoking, physical inactivity, and atherogenic diet. Socio-demographic and lifestyle variables were tested in a binary logistic regression model. The most prevalent risk factors were overweight (20.4%), dyslipidemia (48.1%), and physical inactivity (66.2%). Children below ten years of age and those from higher-income families and with higher maternal schooling showed greater odds of developing overweight; meanwhile, those with overweight were more prone to developing hypercholesterolemia and hypertriglyceridemia. The findings point to the need to implement strategies to prevent overweight in early childhood, through balanced nutrition and regular physical activity, in order to effectively reduce the prevalence of risk factors in schoolchildren.Item Acesso aberto (Open Access) Feeding ecology of immature Lithodoras dorsalis (Valenciennes, 1840) (Siluriformes: Doradidae) in a tidal environment, estuary of the rio Amazonas(Sociedade Brasileira de Ictiologia, 2015-06) BARBOSA, Thiago Augusto Pedroso; BARTHEM, Ronaldo Borges; MONTAG, Luciano Fogaça de AssisStudies of feeding ecology are important for the evaluation of interactive processes in fish communities. This study evaluated the feeding ecology of Lithodoras dorsalis (Doradidae) from streams within the Amazon estuary delta (Brazil), a macro-tidal area, on different pluviometric periods. A total of 371 young specimens was collected during 12 months of sampling (July 2010 to June 2011). The species diet was composed of 28 food items analyzed by Repletion Index, Alimentary Index and Niche Breadth. Young L. dorsalis was classified as herbivore with a frugivory tendency due to the high importance of fruit and seeds in its diet. Food intake varied among sampled months, with the lowest intake being recorded during the rainy-dry season transition period, and the highest at the beginning of the dry season. The importance of food items and the composition of the diet were different throughout the year, probably due to the daily tides that allow fish to access new environments and the pluviometric periods. These results provide important data on the feeding ecology of Amazonian doradids. The study also emphasized the importance of allochthonous resources, derived from the riparian forest, which reinforces the importance of this habitat for the conservation of Neotropical freshwater fishes.Item Acesso aberto (Open Access) Feeding ecology of Serrasalmus gouldingi (Characiformes: Serrasalmidae) in the lower Anapu River region, Eastern Amazon, Brazil(Instituto Nacional de Pesquisas da Amazônia, 2016-09) PRUDENTE, Bruno da Silveira; MARINHO, Pedro Carneiro; VALENTE, Roberta de MeloStudies on feeding ecology of fishes are important to understand the relationship between species and environmental seasonal variations. In tropical rivers, these relationships are mainly modeled by hydrological patterns. Thereby, this study aimed to assess the influence of fluviometric variation and life stage (juveniles and adults) in the feeding ecology of Serrasalmus gouldingi in the lower Anapu River region, located in Eastern Amazon, Pará, Brazil. Specimens were collected bimonthly, considering four different hydrological periods. We assessed the diet composition, feeding intensity and niche breadth of the species. Thirty-two dietary items were identified and grouped into ten categories. A total of 279 stomachs were analyzed, showing a predominance of fish fragments, followed by fruits and seeds. The diet composition of S. gouldingi differed only between drought and flood season, although it did not differ between juveniles and adults. An increase in feeding intensity was recorded during the rise in the water level, with a lower feeding intensity observed during transitional season. Serrasalmus gouldingi showed lower niche breadth during flood season, attributed to the high consumption of fruits and seeds, presenting an omnivorous diet with high tendency towards piscivory. The objective of the research was to determine the age at which male collared peccaries reach puberty. Testicular samples of 15 animals, aged 7 to 16 months, distributed into five groups (G1, G2, G3, G4 and G5) were used. The testes showed considerably increased weight, length and width (p < 0.05) from G1 to G3, whereas, from this group onward, the development of this organ was slower. There was positive correlation (p < 0.001) between the following testicular parameters: weight and length (r = 0.97), weight and width (r = 0.88), length and width (r = 0.92). Regarding the diameter of seminiferous tubules, an increase was observed (p < 0.05) from G1 to G4. The total number of spermatogenic cells increased significantly (p < 0.05) until G3 and then it stabilized. There was also positive correlation between testis weight and tubular diameter (r = 0.99, p < 0.001), and testis weight and spermatogenic cells (r = 0.98, p < 0.001). The number of Sertoli cells decreased significantly (p < 0.05) from G1, when they were undifferentiated as support cells, to G5, when they occurred together with the complete line of spermatic cells. The results demonstrate that the reproductive development of peccaries can be classified into the following stages: impuberty (G1, 7-8 months); pre-pubertal (G2, 9-10 months); pubertal (G3, 11-12 months); post-pubertal 1 (G4, 13-14 months); and post-pubertal 2 (G5, 15-16 months). Based on the histological analyses, puberty in the male collared peccary was determined to occur between 11 and 12 months of age.Item Acesso aberto (Open Access) The role of Lithodoras dorsalis (Siluriformes: Doradidae) as seed disperser in Eastern Amazon(Sociedade Brasileira de Inctiologia, 2017) BARBOSA, Thiago Augusto Pedroso; MONTAG, Luciano Fogaça de AssisIchthyochory is an important process responsible for the high diversity of plant species in tropical flooded forests. Thus, this study aimed to investigate the role of a catfish species, Lithodoras dorsalis, as seed disperser in the flooded forests at the Amazon River mouth, Brazil. Analyzing the stomach contents of 371 individuals of Lithodoras dorsalis, the Germination Potential (GP%) and Germination Speed Index (GSI) of seeds that were removed intact were investigated. This allowed us to evaluate the germination performance of two important species of plants in Amazonia, Euterpe oleracea (Açaí) and Montrichardia linifera (Aninga), after passage through the digestive tract of this catfish species. Given that digestion by L. dorsalis reduced the germination viability of M. linifera and that seeds were often destroyed during consumption, we suggest that L. dorsalis may have a limited role as seed disperser of M. linifera and instead mostly act as seed predator. However, for the species E. oleracea, L. dorsalis was a potential disperser, since the performance of germination of these seeds was improved after digestion. In this article, we report the genome sequence of Corynebacterium pseudotuberculosis strain Cp162, collected from the external neck abscess of a camel in the United Kingdom.
