The samples were tested by six serological methods: two of agglutination, two of indirect ELISA and two of competitive ELISA. To determine the relative sensitivity and specificity of different tests, animals with a positive result to competitive ELISA of the FAO/IAEA were considered as infected. The relative sensitivity of competitive ELISA, indirect ELISA with conjugate anti-bovine light chain monoclonal antibody labelled with HRPO, indirect ELISA with anti-bovine IgG conjugate, rose Bengal test and rapid slide agglutination test was 100%, 98.57%, 97.14%, 91.42% and 79.28%, and the relative specificity 99.33%, 97.33%, 95.66%, 94.00% and 86.33%, respectively. The value of the different serological tests for the diagnosis of brucellosis is discussed.Item Acesso aberto (Open Access) Brucella abortus detected in cheese from the Amazon region: differentiation of a vaccine strain (B19) from the field strain in the states of Pará, Amapá and Rondônia, Brazil(Universidade Federal do Pará, 2016-08) BRITO, Jacqueline da Silva; MORAES, Carina Martins de; SILVA, Cleyzer Lopes; SALES, Gustavo Aguiar; KEID, Lara Borges; MATOS, Paulo Cesar Magalhães; LARA, Ana P.S.S.; MORAES, Carla Cristina Guimarães deBrucellosis is an infectious-contagious disease responsible for significant economic losses to the meat and milk supply chain, because it causes reproductive disorders in animals and is a chronic anthropozoonosis. This study was designed to detect the DNA of Brucella spp. in cheese and to differentiate between a vaccine strain (B19) and the field strain. Sixty-six samples of different cheeses which are produced and marketed in three states of the Brazilian Amazon region (Amapá [5 samples], Pará [55 samples] and Rondônia [6 samples]) were evaluated. Thirty-nine of these samples were from cheeses made from cow's milk, and 27 were from cheeses made from buffalo milk. Four of the 66 samples were from cheeses produced in milk processing plants regulated by the Federal Inspection Service (Serviço de Inspeção Federal); nine of the samples were from cheeses produced in processing plants regulated by the State Inspection Service (Serviço de Inspeção Estadual); five of the samples were from artisanal cheeses; and the remaining 48 samples were from informally produced cheese. DNA was obtained from the samples following a DNA extraction protocol, and PCR was conducted using primers B4 and B5 to detect Brucella spp. Primers eri1 and eri2 were used to differentiate the field strain from the B19 vaccine strain. The results showed that 21.21% (14/66) of the samples were positive for Brucella spp., of which 21.43% (3/14) were positive for the B. abortus field strain, and 7.14% (1/14) were identified as harboring vaccine strain B19. Nineteen buffalo cows in various stages of pregnancy, serologically positive for brucellosis and subjected to culling were used. Fetal age was determined by ultrasound examination and the size of fetuses was measured at necropsy. Fragments of lymph nodes were collected for histopathology and qPCR. The detection of B. abortus DNA in the lymph nodes was checked from the fourth month of pregnancy in seven buffaloes and in a post-calving buffalo. Acute to chronic lymphadenitis was histologically diagnosed. B. abortus DNA was detected in all evaluated groups of lymph nodes; the mammary lymph nodes were the most affected.Item Acesso aberto (Open Access) Estudo soroepidemiológico de Brucella abortus, Toxoplasma gondii e vírus da artrite encefalite caprina em rebanhos caprinos nas unidades produtoras dos estados do Pará e Maranhão(Universidade Federal do Pará, 2009-01-30) PIRES, Claudina Rita de Souza; DIAS, Hilma Lúcia Tavares; http://lattes.cnpq.br/4501155453484456This study aimed at searching for the occurrence and frequency levels of the antibodies anti- Brucella abortus, Toxoplasma gondii and the Caprine Arthritis Encephalitis Virus (CAEV) in caprine from 14 distinct production units within the states of Pará and Maranhão. In the state of Pará, animals from the municipalities of Benevides, Castanhal, Santa Izabel do Pará and Moju have been analyzed, whereas in the state of Maranhão the study covered the municipality of Chapadinha. The serological tests accomplished with the purpose of diagnosing brucellosis disease were, the Buffered Acidified Plate Antigen test (BAPA), employed as triage test, and the 2-Mercaptoetanol (2-Me), as confirmatory test. For the toxoplasmosis analysis the Indirect Immunofluorescence Reaction (IIFR) and for CAEV the Agarose Gel Immunodiffusion (AGID) have been used respectively. The result of the brucellosis analysis has shown negativity in 100,0% of the studied samples. For toxoplasmosis and CAEV the respective incidences were 23,5% (97/412) and 21,6% (85/393). Statistical differences were observed in the relationship between anti-Toxoplasma gondii antibodies and the age of the caprines, showing that animals over 24-month old have undergone more risks of being infected when compared to animals at early ages OR=2,15 (IC95% 1,19 – 3,88). On the other hand, the risk factors found for CAEV were: lack of knowledge about the disease OR=6,45 (IC95% 2,88-14,47); the use of non - disposable materials, OR=10,85 (IC95% 4,85-24,28); the free-range farming system OR=10,85 (IC95% 4,85-24,28), the semi-extensive farming system OR=3,71 (IC95% 1,64-8,35) and handling procedures OR=11,4 (IC95% 5,51-23,60). Twenty buffaloes serologically positive for brucellosis were used and subjected to stamping for collection of material from the closed uterus of several months gestation. Fragments of lymph nodes were collected. Fetal age was determined by ultrasound examination and the size of fetuses was measured at necropsy. The samples were subjected to histopathology and qPCR. From the second month of pregnancy on it was possible to detect the presence of B. abortus DNA in amniotic fluid, allantoic liquid and uterus, and from the fifth month on in placenta, heart, spleen, kidney, lung, intestine, liver and lymph nodes of the fetuses. The main pathological findings were fibrinous suppurative necrotic placentitis, and chronic endometritis. The detection of B. abortus DNA in the lymph nodes was checked from the fourth month of pregnancy in seven buffaloes and in a post-calving buffalo. This study aims to evaluate the humoral and immune response of buffalo calves raised in the Middle Amazon Mesoregion, Pará state, vaccinated with B19 strain of B. abortus according the age recommended by the National Program for the Control and Eradication of Brucellosis and Tuberculosis (PNCEBT). Thirty-six female (test group) and six males (control group) aged 3-8 months were selected and monitored. On day 0 of the experiment, blood was collected from all animals, then all females received commercial vaccine with standard-dose sample B19 as recommended by PNCEBT with subsequent blood samples were taken at 30, 60, 90, 120, 150, 180, 210, 330, 360 and 390 days, respectively. The blood serums were evaluated for evidence of agglutination screening through Buffered Acidified Antigen - AAT and the confirmatory test through 2 Mercaptoethanol - 2ME. The results for all samples collected on day 0 were negative in the reaction, however at 30 and 60 days all females were 100% positive through test reaction AAT and titration 1:200, starting the decline through the titration at 90 days in both events. Females at 210 days still present reagents with similar results in both tests, however only one animal remained reactive in both tests up to 360 days. After 390 days of the begin of the experiment 100% of the females had a negative reaction in both events and throughout the experiment males were negative. Thus, by the evaluation of results, vaccination was effective for immunization, as well as an important tool in the prophylaxis of brucellosis in the species studied, as well as the tests applied to the diagnosis of this disease in buffaloes species at regional level.
