Item Acesso aberto (Open Access) Aspectos clínico-patológicos do envenenamento botrópico experimental em equinos(2011-09) SOUSA, Melina Garcia Saraiva de; TOKARNIA, Carlos Maria Antônio Hubinger; BRITO, Marilene de Farias; REIS, Alessandra dos Santos Belo; OLIVEIRA, Carlos Magno Chaves; FREITAS, Nayra Fernanda de Queiroz Ramos; OLIVEIRA, Cairo Henrique Sousa de; BARBOSA NETO, José DiomedesThe clinical and pathological alterations in horses, experimentally inoculated with Bothropoides jararaca, Bothrops jararacussu, Bothrops moojeni and Bothropoides neuwiedi poisons, were studied with the purpose to supply subsidies for the diagnosis of the poisoning. The liofilized poisons were diluted in 1ml of physiologic solution and subcutaneously administered to six horses, at doses of 0.5 and 1mg/kg (B. jararaca), 0.8 and 1.6mg/kg (B. jararacussu), 0.205mg/kg (B. moojeni) and 1mg/kg (B. neuwiedi). All horses, less those that received the poison of B. jararacussu, died The clinical signs began between 8min and 2h10min after the inoculation. The clinical course varied, in the four cases of lethal exit, from 24h41min to 70h41min, and was 16 days in the two horses that recovered,. The clinical picture, independent of the poison type and doses, was characterized by tumefaction at the site of inoculation, dragging on the ground with the hooves of the inoculated leg, inquietude, apathy, decrease of reaction to external stimuli, pale mucous membranes and hemorrhages. Laboratory exams revealed normocytic normochrômic anemia with progressive decrease in the number of erythrocytes, of hemoglobin and of the hematocrit, and leucocytosis due to neutrophilia. There was increase of alamina aminotransferase, creatinaquinase, lactic dehydrogenase, ureia and glucose, as well increase of the time of activation of protrombin and partial tromboplastina. At postmortem examination, the main findings were extensive hemorrhagic areas in the subcutaneous tissue, with the presence of non-coagulated blood, to a large degree associataed with edema (hemorragic edema), which extended from the inoculation site of the venom to the cervical, thoraxic and scapular region, and to the leg. In the periphery of the hemorragic areas existed gelatinous edema. There were great amounts of sanguinary liquid in the thoracic, pericardic and abdominal cavities. The purpose of this study was to contribute to the understanding of this type of snake accident in horses. The lyophilized venom was diluted into 1ml of a 0.9% saline solution and was inoculated subcutaneously into five horses, at the doses of 0.12mg/kg (one horse), 0.066mg/kg (two horses) and 0.03mg/kg (two horses). The venom caused death of the horse that had received 0.12mg/kg, and of one horse of the two that had received the dose of 0.066mg/kg. The clinical course varied from 27h27min to 52h29min. The second horse inoculated with 0.066mg/kg recovered within 12 days after inoculation. The dose of 0.03mg/kg had a course of 6 to 10 days but did not cause fatal envenomation. The clinical picture in the horses was characterized by swelling of the inoculation site and adjacent areas, by apathy and lowered head, locomotory alterations shown by dragging of the hoves on the ground, decubitus and difficulty to get up, reduction of auricular, palatal, upperlip and threat reflexes, and increase of heart and breathing frequency. The laboratory examination revealed leukocytosis and lymphocytosis in two horses. There was increase of the creatine-kinase (CK), lactic dehydrogenase (DHL) and ureia; reduction in the seric levels of calcium, phosphorus and magnesium. The time of activated partial tromboplastina (TTPA) increased in the horses that died. Post mortem findings were edema of the subcutaneous tissue at the inoculated shoulder, suffusions in the epicard of left and right heart ventricles, bladder replete with urine containing a white-yellowish sediment, and hemorrhagic areas in its mucosa. Histopatologic examination revealed the liver parenchyma with diffuse moderate vacuolation affecting predominantly the intermediate area of the hepatic lobe, and slight dilation of the sinusoides in some areas; in the kidney slight dilation of the uriniferous tubules mainly in the cortex.Item Acesso aberto (Open Access) Aspectos clínico-patológicos e imuno-histoquímicos de equídeos infectados pelo vírus da raiva(2010-11) PEDROSO, Pedro Miguel Ocampos; COLODEL, Edson Moleta; GOMES, Danilo Carloto; VARASCHIN, Mary Suzan; BEZERRA JÚNIOR, Pedro Soares; BARBOSA NETO, José Diomedes; TOKARNIA, Carlos Maria Antônio Hubinger; DRIEMEIER, DavidThirteen horses and one mule from four regions of Brazil with clinical and histological diagnosis of rabies were analyzed. The clinical course lasted four days, with motor incoordination, paresis and paralysis of the legs, and decubitus. The main histopathological findings were lymphoplasmacytic meningoencephalitis and non-suppurative meningomyelitis associated with characteristic Negri bodies in 64.28% (9/14) cases, mainly in the cervical spinal cord and in Purkinje neurons of the cerebellum. In 55.55% (5/9), the animals examined were positive by for the fluorescent antibody test for rabies. All animals were positive in immunohistochemistry for rabies, whose immunolabeling were more evident in the temporal cortex, occipital cortex and cervical spinal cord. The diagnosis can be confirmed either directly by bacterial isolation and PCR or by ELISA, which is an indirect method based on the detection of serum antibodies. The aim of this study was to clone, express and characterize the SeM protein of Streptococcus equi subsp. equi, evaluate its use as antigen in indirect ELISA and determine its performance to distinguish sera of negative, vaccinated and positive animals. This was initially performed by cloning the gene encoding the SeM protein and its expression in Escherichia coli. Subsequently, the protein produced was characterized and used as antigen in ELISA. Serum samples for evaluation were taken from 40 negative foals, 46 horses vaccinated with a commercial vaccine against strangles and 46 horses diagnosed with the disease. The test showed high specificity and sensitivity, allowing discrimination between negative and positive, positive and vaccinated animals, and vaccinated animals and negative sera. Thus, it was concluded that the protein produced rSeM, which can be used as antigen for disease diagnosis, and the described ELISA might be helpful to evaluate the immune status of the herd.Item Acesso aberto (Open Access) Encefalomielite equina Leste na Ilha de Marajó, Pará(Universidade Federal do Pará, 2012-02-29) CAMPOS, Karinny Ferreira; BARBOSA NETO, José Diomedes; http://lattes.cnpq.br/1516707357889557Nine cases of equine encephalomyelitis were studied in the Marajó Island, State of Pará, Brazil. The animals had difficulty in maintaining a station, walk in a circle, marked depression, eyelids closed, tongue paralysis, muscle tremors, bruxism, anorexia and dehydration. Some had their ear and eyelid reflexes diminished, decreased tongue tone and tachycardia. Position of self-hearing was observed frequently. The animals were often found leaning on tree trunks and fences to keep themselves on station. At necropsy, they showed hemorrhage of the meninges and spinal cord, and some animals also showed adhesion of the meninges. Histologically there was diffuse encephalitis affecting mainly the gray matter, with meningitis and choroiditis. It was observed the presence of perivascular cuffs consisting of mononuclear inflammatory cells. In two animals it was possible to identificate the Eastern equine encephalitis virus by semi-nested reverse transcription polymerase chain reaction (semi-nested RT-PCR).Item Acesso aberto (Open Access) Equine infectious anemia on Marajo Island at the mouth of the Amazon river(Universidade Federal do Pará, 2015-12) FREITAS, Nayra Fernanda de Queiroz Ramos; OLIVEIRA, Carlos Magno Chaves; LEITE, Rômulo Cerqueira; REIS, Jenner Karlisson Pimenta dos; OLIVEIRA, Fernanda Gonçalves de; BOMJARDIM, Henrique dos Anjos; SALVARANI, Felipe Masiero; BARBOSA NETO, José DiomedesEquine infectious anemia (EIA) is a transmissible and incurable disease caused by a lentivirus, the equine infectious anemia virus (EIAV). There are no reports in the literature of this infection in Equidae on Marajo Island. The objective of this study was to diagnose the disease in the municipalities of Cachoeira do Arari, Salvaterra, Santa Cruz do Arari and Soure, on Marajó Island, state of Pará, Brazil. For serological survey samples were collected from 294 horses, over 5-month-old, males and females of puruca and marajoara breeds and from some half-breeds, which were tested by immunodiffusion in Agar gel (AGID). A prevalence of 46.26% (136/294) positive cases was found. EIA is considered endemic in the municipalities studied, due to the ecology of the region with a high numbered population of bloodsucking insect vectors and the absence of official measures for the control of the disease.Item Acesso aberto (Open Access) Fotossensibilização hepatógena em eqüinos pela ingestão de Brachiaria humidicola (Gramineae) no estado do Pará(2006-09) BARBOSA NETO, José Diomedes; OLIVEIRA, Carlos Magno Chaves; TOKARNIA, Carlos Maria Antônio Hubinger; PEIXOTO, Paulo Fernando de VargasData on the clinical-pathological aspects and on the epidemioloy of a disease in horses, characterized by phototsensitization, which occurs in northeastern of Pará, Brazil, are presented. Of a total of 40 horses examined clinically, post-mortem examination was performed on seven. Due to the characteristic lesions found on the non-pigmented skin, the macroscopic and histological alterations in the liver, and knowing that Brachiaria grasses contain saponins with toxic properties, which cause liver lesions and photosensitization in cattle and sheep, it is concluded that this disease in horses is caused by grassing exclusively Brachiaria humidicola.Item Acesso aberto (Open Access) Freqüência de anticorpos anti-Borrelia burgdorferi em eqüinos na mesorregião metropolitana de Belém, Estado do Pará(Universidade Federal do Pará, 2006) GALO, Katiany Rocha; FONSECA, Adivaldo Henrique da; http://lattes.cnpq.br/4411441162862608; BARBOSA NETO, José Diomedes; http://lattes.cnpq.br/1516707357889557Lyme borreliosis is a multisistemic disease caused by the spirochete Borrelia burgdorferi latu sensu and transmited by ixodide ticks, affecting both humans beings and domesticated animals and having wild animals as Its natural reservoirs, besides of being a zoonosis of wide geographic distribution. We picked 300 blood samples of apparently health horses from the cities of Ananideua, Belém, Benevides, Castanhal, Marituba and Santa Izabel of the Pará, all included in the metropolitan mesoregion of Belém – Pará. The samples were picked up by the jugular vein and the serum analised through indirect Enzyme-Linked Immunosorbent Assay (ELISA) in the Laboratory of Parasitic Diseases of the Federal Rural University of Rio de Janeiro. The aim of this work was to verify the serum epidemiology of Borrelia burgdorferi in the metropolitan mesorregion of Belém. The frequency of serum positivity was 26,67% (n=80), being 72 (24%) with title of 1:800, six (2%) with 1:1600 and two (0,6%) with 1:3200. The serum frequency by gender was approximately 13,67% of females and 13% of males. The thoroughbred and the half-breed animals had 9% and 18% of frequency, respectively. There was no significative difference in the serum positivity with regard to cities, gender, race and age. The frequency found corroborates the hypothesis of occurrence of Borrelia sp. in the studied region.Item Acesso aberto (Open Access) Freqüência de anticorpos homólogos anti-Borrelia burgdorferi em eqüinos na mesorregião metropolitana de Belém, Estado do Pará(2009-03) GALO, Katiany Rocha; FONSECA, Adivaldo Henrique da; MADUREIRA, Renata Cunha; BARBOSA NETO, José DiomedesSpirochaetes transmitted by ticks are microorganisms of worldwide distribution, which infect wild, domestic animals and human beings. A total of 300 equine sera from four municipalities: Ananideua (58), Belém (61), Castanhal (131), and Santa Izabel do Pará (50), were evaluated for Borrelia burgdorferi by an Elisa test. There were no significant differences (P<0.05) among municipalities, breed, sex or husbandry. A total of 80 (26.7%) horses were B. burgdorferi positive with titles of 1:800, 72 (90%) horses, 1:1.600, 6 (7.5%) horses, and 1:3.200, 2 (2.5%) horses. The results were similar to those in the USA, where related frequencies ranged from 7 to 75% in asymptomatic seropositive horses. The presence of anti-B.burgdorferi homologous antibodies in horses from four municipalities in the metropolitan mesorregion of Belém suggests the possibility of occurrence of human cases in the region.Item Acesso aberto (Open Access) Inibição e reversão da agregação plaquetária de eqüinos in vitro com o uso de ketoprofeno, fenilbutazona e flunixim meglumine(2009-10) LEME, Fabiola de Oliveira Paes; WURZINGER, Laurenz; ALVES, Geraldo Eleno Silveira; BICALHO, Adriane Pimenta da Costa-Val; BARBOSA, Marcos Pinott; PAES, Paulo Ricardo de Oliveira; OLIVEIRA, Marcos Enê ChavesSeveral diseases may lead to platelet pre-activation and hypercoagulability states in horses. The activity of many drugs against platelet aggregation may, not only contribute to the evaluation of a disease but also its response to the therapy. With the aim to study in vitro prevention and reversion of platelet aggregation, the non steroidal anti-inflammatory drug (NSAID): ketoprophen, phenylbutazone and flunixin-meglumin were evaluated. The comparison demonstrated that phenylbutazone and ketoprophen prevented platelet aggregation induced by ADP better than flunixin-meglumin, in a superior manner to the monoclonal antibody Reopro, and in a better way than the membrane receptor blockers Ro-438857 and RGDS. The reversion of platelet aggregation demonstrated that even phenylbutazone or ketoprophen have a dose-dependent effect.Item Acesso aberto (Open Access) Investigação soroepidemiológica para brucelose e leptospirose em equideos de tração e seus tratadores nos municípios de Belém e Ananindeua - Pará(Universidade Federal do Pará, 2007-06-18) SANTOS, Wilson Rogério Rodrigues dos; DIAS, Hilma Lúcia Tavares; http://lattes.cnpq.br/4501155453484456The objective of the work was the detention of antibodies anti - Brucella abortus and anti - Leptospira interrogans in serum of equines and equine workers of the quarters of the cities of Belém and Ananindeua, enclosing the months from April to August of 2005, using for this end, 195 sanguineous serum of eqquines and 70 sanguineous serum of men that manipulated the animals direct or indirectly. For the research of reagents serum of animals to B. abortus, the tests used had been Antigen Acidified Test (AAT), as a selection test, and slow seroagglutination (SAL) and and the 2-mercaptoetanol (2-ME) as confirmatory tests. To Leptospirosis, the test used was of microscopical seroagglutination being carried through the selection of the serum front to the 25 serovars of L. interrogans, considering positive those samples with titulation equal or bigger that 100. Of 195 samples of equine sanguineous serum, 184 (94.87%) had been positive for all serovars analyzed, being that more frequent found: Patoc, Automnalis, Ictehaemohragiae, Pyrogenes and Bratislava. For the sanguineous samples of men, the positivity was of 49 (70%) of reacting serum, with the most detected serovars: Patoc, Ictehaemohragiae, Bratislava, Butembo, Copenhageni and Automnalis. Of the positive samples of equine and its respective workers, 47/70 (67.14%) were similar for same serovars of Leptospira spp., being that 2/70 (2.86%) samples had been negative in both the searched groups, 2/70 (2.86%) were only positive in humans and 19/70 (27.14%) were exclusively positive in the samples of equine serum. The quarters of Coqueiro, Guamá, 40 horas, Barreiro and Bengui had presented the biggest percentage of positive cases serum. It did not have significant difference in relation to the other studied variants, as: climate, age (animal and man), time of service (animal and man), species of the animal, props up corporal of the animal and degree of instruction of the man. These were visits to the farm, epidemiological studies, blood sample collections, skin biopsies, and inspection of the pasture. The study included 25 Equidae, 14 males and 11 females, six months to eight years old. The animals showed ulcerative lesions of irregular borders on the head (nose, muzzle, upper and lower lips), in the oral cavity (buccal vestibule and gum) and on the limbs (billets, metacarpals and metatarsals and scapular-humeral joint). The histopathological examination revealed foci of cutaneous erosions with epidermal necrosis, spongiosis and vesicular degeneration of the remaining epidermis, and mild inflammatory infiltrate in the underlying dermis, consisting predominantly of macrophages and, to a lesser degree, eosinophils. The inspection of the pasture, which consisted of Brachiaria humidicola, revealed a heavy invasion by two plants provided with spines, Mimosa pudica and Mimosa debilis, of the Leguminosae Mimosoideae family. It was concluded that the skin lesions were caused by traumatic action of the spines of Mimosa pudica and Mimosa debilis.Item Acesso aberto (Open Access) Lesões traumáticas na pele causadas pelos espinhos de Mimosa pudica e Mimosa debilis em equídeos(Universidade Federal do Pará, 2012-02-24) REIS, Alessandra dos Santos Belo; BARBOSA NETO, José Diomedes; http://lattes.cnpq.br/1516707357889557We studied traumatic injury of the skin in horses caused by traumatic plants, popularly known as "sleep-mary", "Poppy", "scraper", "malice" and "do not touch me". The study was conducted on a farm in the district of Castanhal, northeastern of the state of Para, where there were technical visits, epidemiological study, blood samples, biopsies of affected skin and collection of plants. The study included 25 horses, 14 males and 11 females, aged between six months and eight years. The pasture consisted of Brachiaria humidicola and was heavily invaded by traumatizing plants. The animals showed ulcerative lesions of irregular borders, on the head (nose, muzzle, upper and lower lips and chamfer), oral cavity (buccal vestibule and gum) and limbs (billets, metacarpals and metatarsals and scapular-humeral joint). The histopathological examination revealed foci of cutaneous erosions, characterized by loss and epidermal necrosis with spongiosis and vesicular degeneration of the remaining epidermis and mild inflammatory infiltrate in the underlying dermis, consisting predominantly of macrophages and, to a lesser degree, eosinophils. In this paper, plants causing mechanical injury to ruminants and horses in Brazil are reviewed, and an outbreak in sheep of mechanical injury of the skin by Stipa sp. in Uruguay is reported. The sheep, mainly lambs, showed large number of fruits of Stipa sp. in the wool, some of them penetrating the skin, panniculus adiposus, and cutaneous trunci muscle, causing multifocal dermatitis and panniculitis.Item Acesso aberto (Open Access) Prevalência da anemia infecciosa equina na Ilha de Marajó, estado do Pará, Brasil(Universidade Federal do Pará, 2014) FREITAS, Nayra Fernanda de Queiroz Ramos; BARBOSA NETO, José Diomedes; http://lattes.cnpq.br/1516707357889557The aim of the study was to determine the prevalence of equine infectious anemia in the municipalities of Cachoeira do Arari, Salvaterra, Santa Cruz do Arari and Soure, in the Marajó Island, State of Pará, Brazil. For serological survey were selected 349 samples collected from June 2012 to March 2013, tested by agar gel immunodiffusion. Blood count was performed in 65 samples and biochemical examinations in 70, in which was researched urea, creatinine, aspartate aminotransferase, alanine aminotransferase, gamma glutamyl transferase, alkaline phosphatase, total bilirubin and direct bilirubin. A prevalence of 24.06% (84/349) was verified. The average number of red blood cells was significantly lower in seropositive animals in relation to the seronegative ones and there was no significant difference in mean results of hematocrit, hemoglobin, mean corpuscular volume, mean corpuscular hemoglobin concentration, platelets, in the white blood cell count, as well as in the biochemical examination. The clinical observed was poor nutritional status, apathy, pale mucous membranes, dehydration and elevation in heart and respiratory rate. At necropsy, the primary findings in all horses were jaundiced housing, small accumulation of fluid in the abdominal cavity, as well as hepatomegaly and splenomegaly. The histopathological examination showed spleen and liver with hemosiderosis. The enlargement of varied extension was uni or bilateral. The cheek mucosa of the labial commissure showed slight erosions. During chewing there was loss of small amounts of grass and saliva through the oral cleft. The affected horses were in good nutritional condition. Histopathological studies of tissues obtained by biopsia, revealed a superficial epidermitis. The pastures consisted of Panicum maximum grass (varieties Tanzânia, Mombaça, Tobiatã and Colonião) which was mature, tall, lignified, with leaves of cutting edges. Based on epidemiological, clinical and histopathological data, it was concluded that the lesions were caused by the hard grass, favored by the way horses pull the tall grass and chew it.Item Acesso aberto (Open Access) Reemerging of natural infection by Trypanosoma evansi in horses in Arari, Marajó Island, Brazil(Universidade Federal do Pará, 2016-12) SILVA, Josileide Araújo da; DOMICIANO, Tarcísio Oliveira; MONTÃO, Daniele Pina; SOUSA, Paulo Geovani Silva; RAMOS, Leandro Lopes; PAREDES, Laura Jamille Argolo; GONZALEZ MONTEIRO, Silvia; RIVERO, Beatriz Riet Correa; SCOFIELD, Alessandra; BEZERRA JÚNIOR, Pedro; BEZERRA, Isis Abel; CERQUEIRA, Valíria DuarteTwo outbreaks of trypanosomiasis by Trypanosoma evansi in horses in the municipality of Chaves, Marajó Island, Pará State, Brazil are reported. The first outbreak occurred in April 2011 in a farm with 147 horses; of these, 47 (31.97%) got sick, and 40 (27.21%) died. The second outbreak occurred in May 2012 and involved nine properties. Of a total of 679 horses, 209 (30.07%) got sick, and 183 (26.97%) died. The main clinical signs observed in these horses were weight loss, abdominal edema, toe dragging, and pelvic muscle atrophy. Necropsy was performed in two horses, one from each outbreak. Macroscopic alterations were not seen in the horse from the first outbreak; however, the horse from the second outbreak exhibited emaciation, pallor and icterus, as well as enlarged spleen with prominent white pulp at cut surface. Histopathology of the central nervous system of both animals revealed mild to severe, diffuse lymphoplasmacytic encephalitis, with presence of Mott cells. Immunohistochemistry for T. evansi revealed structures similar to intralesional trypomastigote forms. Between November and December of 2013, an epidemiological survey was carried out in the municipalities of Cachoeira do Arari, Santa Cruz do Arari, Salvaterra, Soure, and Chaves. Only Santa Cruz do Arari and Chaves had reported cases of the disease. The affected horses had difficulty to stand, walked in circles, with marked depression, closed eyelids, tongue paralysis, muscle tremors, bruxism, anorexia and dehydration. Some had their ear and eyelid reflexes diminished, decreased tongue tone and tachycardia; laid down frequently they kept their head on the chest. Often they were seen resting their head on tree trunks or fences. At necropsy, hemorrhages of the meninges and spinal cord, and in some animals also adhesion of the meninges were found. Histologically there was diffuse encephalitis affecting mainly the gray matter, with meningitis and choroiditis. Presence of perivascular cuffs consisting of mononuclear inflammatory cells was observed. From two horses the Eastern equine encephalitis virus was identified by semi-nested reverse transcription polymerase chain reaction (semi-nested RT-PCR).
