Highly lethal, remais a serious public health problem because of difficulties in early diagnosis and the development of effective therapeutic measures. Recent in the field of molecular biology studies suggest that define the profile of miRNAs in hepatocellular carcinoma may considerably influence the identification of risk factors associated with oncogenes and suppressor genes. The objective is to evaluate the expression of miRNA 135b, miRNA 181a-5p and miRNA 181a-3p in samples of Hepatocellular Carcinoma and Chronic Hepatitis C and correlate them so likely to seek biomarkers related to the mechanism of carcinogenesis. The research was done in six patients with hepatocellular carcinoma and twenty four cases of Chronic Hepatitis C, Para, northen Brazil. All samples Hepatocellular carcinoma underwent microdissection for subsequent RNA extraction. For the extraction of total RNA and microRNA All=Prep the DNA / RNA FFPE kit (quiagem), quantified by the Qubit® 2.0 Fluorometer (Invitrogen) for final concentration of 5ng/μL standard equipment was used. The cDNA was obtained using TaqMan® MicroRNA Reverse Transcription (Applied Biosystems). Statistical analyzes were performed in softwares SPSS 17.0, using the Mann-Whitney test, with significat differences in the expression levels of the miR181a-3p and miR 181a-5p in hepatocellular carcinoma (average 3.94 and 17.9, respectively) compared with chronic hepatitis C (average 1.18 to 1.8, respectively) with P-value of 0.005 and 0.003. In this study, it was observed that miRNAs 181a-3p and 181a-5p, especially the 181a-5p way were significantly more highly expressed in hepatocellular carcinoma samples when compared to non-tumor liver tissue with chronic hepatitis C. Therefore, microRNAs have interesting characteristics that favor them as possible in biological screening for early diagnosis of tumors and targeted therapies selected markers.Item Acesso aberto (Open Access) Avaliação de microRNAs circulantes na esquizofrenia: da desregulação epigenômica a potenciais biomarcadores(Universidade Federal do Pará, 2021-03) RODRIGUES, André Luiz de Souza; BURBANO, Rommel Mario Rodriguéz; http://lattes.cnpq.br/4362051219348099; https://orcid.org/0000-0002-4872-234XIntroduction: Schizophrenia is a serious and complex pathology that affects about 0.5-1% of the world's population. For the clinical diagnosis of schizophrenia, there are clinical criteria to be evaluated, which include both positive and negative symptoms. In the origin of the disease, there is a close relationship between environmental stimuli, and strong evidence shows that these stimuli have the ability to act on epigenetic mechanisms, which act in the regulation of gene expression. MicroRNAs (miRNAs) are stable and potentially reliable biomarkers, and some miRNAs have been previously identified as potential biomarkers for schizophrenia in peripheral samples. Objective: To evaluate the expression profile of circulating miRNA's in patients with schizophrenia (hsa-miR-34a, miR-449a, miR-564, miR-432, miR-548d, miR-572 and miR-652) in relation to control individuals negative for the disease. Methods: Analytical, case-control, cross- sectional study using samples previously collected from patients diagnosed with Schizophrenia (N = 650) and control group (N = 924), who adequately met the inclusion criteria. The samples were analyzed after RNA extraction through its quantification and techniques for obtaining reverse transcriptase reaction and real-time quantitative polymerase chain reaction. All data were analyzed using IBM SPSS22 statistical program. Results: Using the peripheral blood collection method with the intention of finding possible biomarkers for schizophrenia, an increased expression of the miRNA’s miR-34a, miR-449a, miR-564, miR-432, miR-548d, miR-572 and miR-652 was observed in several scenarios analyzed, confronting the case and control groups, as well as variables within the case-group, demonstrating potential diagnostic value.Item Acesso aberto (Open Access) Repercussões das análises de bioinformática nos resultados da expressão diferencial de genes e seus reguladores miRNAs no câncer gástrico(Universidade Federal do Pará, 2018-09-13) ARAÚJO, Emily Vanessa Nascimento; SANTOS, Ândrea Kely Campos Ribeiro dos; http://lattes.cnpq.br/3899534338451625The cancer, according to Globocan, in 2018, was responsible for a rate of 8.2 million deaths and about 14.1 million new cases. Regarding the analysis of gastric cancer (GC), in the state of Pará, it was estimated that for the year 2018 the number of affected men would be 23.30 and 11.45 for women. In the development of gastric adenocarcinoma, several significantly increased molecular alterations were identified when compared to non-cancerous tissues. One of these elements are miRNAS, small non-coding RNAs that act on the regulation of gene expression and can act as oncogenes or tumor suppressors. The objective of this study was to investigate the possibility of genes from tumor samples (deposited in The Cancer Genome Atlas - TCGA database) to be modified by differentially expressed miRNAs. The analysis was performed by sequencing platform, RNA-Seq, and used the software R-peridot in differential expression analysis, whose result identified six miRNAs. Next, the miRTargetLink Human platform identified eight miRNA target genes between tumor samples and their corresponding adjacent CG tissues. Functional analyzes identified important genes that can be enriched by three pathways associated with cancer: focal adhesion, RAP1 signaling pathway and calcium signaling pathway. Finally, the use of the Z-Score calculation confirmed significant findings.
