    Application of the comet assay in erythrocytes of Oreochromis niloticus (Pisces): a methodological comparison

    The present study applied the comet assay to erythrocytes of Oreochromis niloticus with the aim of improving protocols to detect DNA damage in these cells, by using two distinct pHs (pH = 12.1 and pH > 13) and evaluating whether there is a correspondence between silver and ethidium bromide staining. Comets were visually examined and, the frequency of cells with and without damage was obtained, as well as the distribution of classes and scores. By using the Kruskal-Wallis test, our results revealed that pH 12.1 is more effective, although both pHs can be used. Our findings also suggest that silver staining can substitute ethidium bromide, an expensive and highly toxic stain that requires specific equipment for examination.

    Efeito do fotoperíodo sobre a composição do filé de juvenis de tilápia do Nilo

    This study aimed to evaluate the effect of the photoperiod on the fillet composition of Nile tilapia juveniles. We employed a completely randomized design with five treatments (0L:24D, 6L:18D, 12L:12D, 18L:6D, 24L:0D) and four replications. We used 160 Nile tilapia juveniles, distributed in 20 tanks containing 20 liters of water, in a recirculation systems, at controlled temperature of 27 ºC and density of eight fish per tank. Fish were fed twice a day with commercial extruded feed containing 40% crude protein. Samples of the fillet at the end of 75 days were collected and stored in a freezer at -80 ºC for subsequent analysis of chemical composition. There was no effect of photoperiod on moisture and ash content of the fillet (P>0.05). The period 18L:6D showed higher amounts of protein in the fillet, as well as lower amounts of fat (P<0.05). Therefore, it can be concluded that the photoperiod can alter Nile tilapia body composition, and the photoperiod 18L:6D contributed to the higher concentration of crude protein and lower fat deposition in Nile tilapia fillets.

    Fotoperíodo sobre parâmetros fisiológicos relacionados ao estresse em alevinos de tilápia-do-nilo

    The objective of this study was to evaluate the influence of different photoperiods on physiological parameters related to stress in Nile tilapia fingerlings (Oreochromis niloticus). A completely randomized design with five treatments (0L, 6L, 12L, 18L, 24L) and four replications was used, and the aquarium as the experimental unit. One hundred and sixty tilapia fingerlings, 3.21±0.05g and 4.35±0.07cm were distributed in 20 aquariums of 20L in a recirculation system with temperature controlled at 27 º C and density of eight fish per aquarium. Feeding was performed twice daily with extruded feed containing 40% crude protein, for a period of 75 days. At the end of experimental period, the data of survival, cortisol, glucose and hematocrit were subjected to analysis of variance and, subsequently, in case of significance (P<0.05) a Tukey test 5% was performed to compare means. Manipulation of photoperiod shows no alteration in the homeostasis of juveniles, since there is no difference in survival, blood glucose, hematocrit and plasma cortisol (P>0.05).
