    Atividade antibacteriana de plantas medicinais frente á bactérias multirresistentes e a sua interação com drogas antimicrobianas

    Infection control of the multidrug-resistant microorganisms sometimes is ineffective even with the development of new antibiotics. Many herbal extracts have antimicrobial effects and may represent an alternative therapy for infectious diseases, mainly when associated with antibiotics of clinical use. The aim of this study was to evaluate the antibacterial activity of medicinal plants in multidrug-resistant microorganisms and their interaction with antimicrobial agents. We evaluate the antibacterial activity of plant extracts and fractions of Eleutherine plicata (“marupazinho”) Geissospermum vellosii (“pau-pereira”) and Portulaca pilosa (“amor-crescido”) against isolates of Oxacillin-Resistant Staphylococcus aureus (ORSA) and multi-resistant bacteria Pseudomonas aeruginosa, from human clinical isolates. Also we evaluate interaction of these plant extracts with antimicrobial agents of clinical use. The antibacterial activity was determined by disk diffusion on Mueller Hinton agar and the Minimum Inhibitory Concentration (MIC) by micro dilution plate technique using Muller Hinton broth as culture medium and 0.01% resazurin as a developer of bacterial growth. The extracts and fractions were tested at concentrations of 500, 250, 125, 62.5, 31.2 and 16.2 μg/mL dissolved in 10% DMSO. Plants E. plicata and G. vellosii demonstrated activity against ORSA isolates with MICs of 125 μg/mL, whereas P. pilosa had an effect on the isolates of P. aeruginosa with MIC of 250 μg/mL. There were 25% of synergism and only 5% of antagonism of all 120 plant and antimicrobial agents interaction tested. ORSA isolates had synergistic interaction with ciprofloxacin, clindamycin and vancomycin agents and with both plant derivatives of E. plicata and G. vellosii. The derivatives of P. pilosa potentiated the action of the aztreonam, cefepime and piperacillin + tazobactam agents compared to the isolates of P. aeruginosa multidrug-resistant. The results shows therapeutic potential of E. plicata, G. vellosii and P. pilosa in the control of bacterial infections involving multidrug-resistant phenotype (MDR) and its interaction with antibacterial agents may represent a new alternative in the therapy of these infections.

    Metodologias empregadas em estudos de avaliação da atividade alelopática em condições de laboratório: revisão crítica

    During the last decades, many allelopathy research groups have been formed, worldwide. As part of this trend, Brazil has published a large number of scientific articles in national and international periodicals. Allelopathy, a relatively new science, is in its first stage of development; thus, a proliferation of often not very sound techniques has occurred. The use of such techniques makes it difficult to interpret the results, their biological value and real meaning. Procedures must become more uniform, as the importance of allelopathy increases. Thus, this work has critically revised the bioassay protocols used to characterize the allelopathic properties of crude extracts and pure substances, presenting the strong points and limitations of each procedure, as well.

    Nitro compounds have been used in therapy from the 40s, when they were widely synthesized and tested against various diseases as well as the discovery of their cytotoxic effects. Thus different groups, derivatives and analogs which could prove safer and more effective were studied. Some studies have shown that nitrostyrenes, like the nitrofenileteno possess diverse biological activities such as antifungal, antibacterial, anti-inflammatory, antinoceptive, etc. Additonally, there is currently a growing interest in new antifungal agents. Among the fungi of the genus Aspergillus is the most common Aspergillus fumigatus which is transmitted through the air causing aspergillosis. Dermatophytes are clinically relevant and cause various skin diseases, Trichophyton mentagrophytes being quite prevalent. The objective of this study was to validate the chemical structure of synthesized nitrostyrenes and evaluate its antifungal activity in two species of fungi, including a filamentous dermatophytes and opportunistic as well as evaluation of the synergistic effects among nitrostyrenes and amphotericin B (AB). The results confirm the infrared spectral and nuclear magnetic resonance of 1H nitrostyrenes analogues as correspond with the proposed structure. Test microdilution demonstrated in vitro antifungal activity of nitrostyrenes with MIC values for 4'-methyl-1-nitro-2- phenylethene (7B) of 0,05 mg/ml against A. fumigatus. For T. mentagrophytes the most active compound was 4'-methoxy-1-nitro-2-phenylethene (7C) with MIC of 0,22 mg/mL. Regarding the analysis of the combination of AB and nitrostyrenes in vitro by the checkerboard test showed antagonist compound 7A and 7C. The 7B compound was indifferent against the standard strain of A. Fumigatus.
