Item Acesso aberto (Open Access) Atividade antimicrobiana do óleo essencial de Piper aduncum L. e seu componente, dilapiol, frente a Staphylococcus spp. multirresistentes(Universidade Federal do Pará, 2012-08-31) BRAZÃO, Maria Angélica Bolini; MONTEIRO, Marta Chagas; http://lattes.cnpq.br/6710783324317390The essential oil of Piper aduncum (EPO) and its main component, the dilapiol (76.5%), were evaluated for antibacterial activity against different strains of Staphylococcus spp. ATCC and multiresistant. For testing the antibacterial activity of EPO and dilapiol were determined Minimum Inhibitory Concentration (MIC) and minimum bactericidal concentration (MBC), the technique of microdilution and by counting Colony Forming Units (CFU) using concentrations of 250, 500 , 750 and 1000 μg/ml of EPO and concentrations of 100 and 1000 μg/ml dilapiol. The bacterial inoculum used was adjusted to 1x104 range from 0.5 Mc Farland. As a negative control inoculum was used along with Tween 20, solubilising the essential oil and dilapiol, and as positive control we used the chloramphenicol 0.05 mg/mL. These compounds were tested against strains of S. aureus ATCC 25923, S. epidermidis ATCC 12228, MRSA hospital, and S.epidermidis, S. lentus multiresistant nosocomial. The results showed that the EPO showed the MIC 90% of 500 μg/mL and MBC of 1000 μg/mL against S. aureus ATCC 25923, whose concentration of 500 μg/mL was able to inhibit 60% of bacterial growth. The strain MRSA, the EPO showed a small inhibition (10%) at a concentration of 750 μg/mL, being obtained in the MIC90% 1000 μg/mL. In S. epidermidis ATCC 12228, the OEPA showed antimicrobial activity with MIC90% at 500 μg/mL and MBC at 750 μg/mL. For the strain S. epidermidis multiresistant, the PEO was able to inhibit only 35% growth of this strain at a concentration of 750 μg/mL, but the value obtained in the MIC90% 1000 mg/mL. As for dilapiol, the compound showed antimicrobial activity against a strain of S. aureus ATCC 25923 at a concentration of 1000 μg/mL, 100% growth inhibiting (CBM). On the other hand, had no antimicrobial effect on MRSA strain nor S. lentus multiresistant. Furthermore, the dilapiol inhibited only 20% of the growth of S. epidermidis ATCC 12228 and S. epidermidis multiresistant concentration of 1000 μg/mL. Thus, the data show a moderate antibacterial activity of the essential oil, and the dilapiol showed weak in vitro antimicrobial activity.Item Acesso aberto (Open Access) Avaliação de atividade antimicrobiana e perfil fitoquimíco de plantas medicinais utilizadas por comunidades remanescentes de quilombos no Marajó(Universidade Federal do Pará, 2020-12-28) SILVA, Suzana Helena Campelo Nogueira da; MONTEIRO, Marta Chagas; http://lattes.cnpq.br/6710783324317390; https://orcid.org/ 0000-0002-3328-5650; SILVA, Consuelo Yumiko Yoshioka e; http://lattes.cnpq.br/8337688339279747; https://orcid.org/ 0000-0002-9120-1910The ethnopharmacological survey is recognized as one of the most viable methods in the search for new medicinal plants, with the ultimate purpose of producing medicines of natural or semi-synthetic origin. In this sense, the remaining communities of Amazonian quilombolas (Marajó-PA) carry with them a great deal of knowledge about the use of medicinal plants, which has been passed down for generations in Mararajo soil, promoting the value of popular knowledge and its applicability in future studies. The objective of this study is to provide scientific support for the traditional use of plants in the treatment of dermatological diseases in quilombola communities in Marajó that do not yet have an adequate chemical and / or pharmacological study. During the fieldwork carried out between 2017 and 2018, 13 communities were interviewed, in which 7 plants with use in skin diseases were cited. Such plants were collected, and their exsiccates were prepared. After botanical identification by a qualified professional, an extensive bibliographic review was carried out, after which 3 plants were selected for phytochemical and pharmacological study (in vitro in Microsporum and Staphylococcus aureus). In addition, they were subjected to ORAC antioxidant and TP total polyphenols tests. The phytochemical profile was analyzed using liquid chromatography coupled to mass spectrometry (LC-MS) providing 17 chemical constituents, belonging to the flavonoid class. The results of the in vitro tests showed an antibacterial potential of the ethanolic extract of the leaves of C. alatus (MIC) of 0.625 µg / mL and (CBM) of 0.734 µg / mL and of the ethanol extract of the roots D. floribunda (MIC) of 125, 0 ug / mL and 200.0 μg / mL (CBM) compared to S. aureus, with emphasis on the F3 fraction (MIC) 25.0 μg / mL and (CBM) of 132.0 which presented the highest bacterial inhibition. Therefore, the results contributed to the validation of popular use and chemical characterization of the species that presented antimicrobial potential, which can be a promising candidate for herbal medicine.Item Acesso aberto (Open Access) Efeito da temperatura e molaridade na avaliação das atividades antimicrobiana, citotóxica e antioxidante do bio-óleo da semente do açaí (Euterpe oleracea, Mart.)(Universidade Federal do Pará, 2024-08) SILVA, Iago Castro da; MACHADO, Nélio Teixeira; http://lattes.cnpq.br/5698208558551065; MONTEIRO, Marta Chagas; http://lattes.cnpq.br/6710783324317390; https://orcid.org/0000-0002-3328-5650Açaí, a fruit from the Amazon, is valuable both economically and nutritionally. Its seeds, which are typically discarded, can be converted into bio-oil through pyrolysis (a process of thermochemical degradation of residual biomass), offering a sustainable alternative to fossil fuels. This study explores how temperature and molarity with Potassium Hydroxide (KOH) and Hydrochloric Acid (HCl), which are chemical impregnation reagents in the process, affect the antimicrobial, antioxidant, and cytotoxic activities of the produced bio-oil. Tests were conducted using Gas Chromatography coupled with Mass Spectrometry (GC-MS) and assays to evaluate antimicrobial, antioxidant, and cytotoxic activities at different temperatures (350, 400, and 450 °C) and molarities (0.5 M, 1.0 M, and 2.0 M). Phenolic compounds were the most abundant in the bio-oil (55.70%), followed by cyclic and aromatic hydrocarbons (11.89%) and linear hydrocarbons (9.64%). Despite a reduction in oxygenated compounds, the bio-oil retained bacteriostatic activity against Escherichia coli and Staphylococcus aureus across various temperature ranges, with notable effectiveness at 350 °C. Antioxidant activity was highest at 350 °C and at lower molarities. Furthermore, lower concentrations of acidic impregnation exhibited cytotoxic effects at high temperatures. Thus, bio-oil from açaí seeds generated through pyrolysis shows potential for antioxidant and antimicrobial activities, suggesting feasibility for further testing in dilutions with lower cytotoxicity.Item Acesso aberto (Open Access) ICB/PPGBAIP Estudo da ocorrência e perfil de suscetibilidade aos antimicrobianos de Staphylococcus aureus isolados de pacientes e profissionais de saúde na Unidade de Terapia Intensiva de hospital público de Rio Branco-AC(Universidade Federal do Pará, 2011-02-07) LAVIOLA GARCÊZ, Poliana Torres; LOUREIRO, Edvaldo Carlos Brito; http://lattes.cnpq.br/2685418720563351The nosocomial infection is a serious public health problem worldwide, mainly in patients admitted to the Intensive Care Unit, which are subject to greater risk due to the severity of clinical symptoms, constant use of broad spectrum antibiotics and frequency of use of invasive procedures. Staphylococcus aureus is a major pathogen that colonizes healthy individuals and is also responsible for infections in hospitalized patients. This study aimed to identify the resistance profile, main sites affected by infection and possible risk factors associated with infection or colonization by S. aureus isolated from patients and healthcare professionals from the Intensive Care Unit of Hospital Emergency and Emergency, Rio Branco (HUERB) – Acre. We developed a cross-sectional study, conducted between January to August 2009. To search for carriers, biological samples were collected from microbiota of patients and professionals and professionals hand washing. For a survey of cases of patients with nosocomial infection were collected biological samples from sites suspected of being affected, 72 hours from the date of admission until discharge, transfer or death. Of the 62 patients enrolled in the study, 19.3% were carriers and 6.4% developed nosocomial infections by S. aureus, and 35 professionals, 28.6% were carriers of S. aureus. It was the second most bacterial species isolated from patients, and was the fifth most isolated from cases of nosocomial infection. There was no statistical evidence for the variable state of coma, use of invasive procedures and state of carrier patient are considered risk factors for acquiring nosocomial S. aureus in this study. The anatomical sites affected by IH by S. aureus were the respiratory tract ( n=2), followed by blood (n=1). The sample catheter tip was responsible for one insulated. One (1.6%) patient developed IH by MRSA, and 5 (8,1%) patients and 2 ( 5.7%) professionals were MRSA carriers, low occurrence as it relates to the results of the rest of Brazil and world. We also emphasize the incidence of MRSA over MSSA and low antomicrobial resistance of MRSA demonstrating that the UTI-HUERB, the IH S. aureus does not constitute a public health problem. There were no isolates of S. aureus resistant to vancomicina, wich can be considered a therapeutic option for cases of IH by MRSA. It is worth emphasizing the importance of this study in Acre State, for being the first of its kind in the UTI, involving S. aureus and MRSA.
