Item Acesso aberto (Open Access) Anticorpos IgG anti-Neospora caninum e Toxoplasma gondii em búfalas (Bubalus bubalis) criadas no estado do Pará(2010-05) SILVA, Sandro Patroca da; MOTA, Rinaldo Aparecido; CAVALCANTI, Erika Fernanda Torres Samico; SOUZA NETO, Orestes Luis de; ALBUQUERQUE, Pedro Paulo Feitosa de; DIAS, Hilma Lúcia Tavares; FARIA, Eduardo Bento deTo investigate the occurrence of antibodies IgG anti-Neospora caninum and Toxoplasma gondii, blood samples were collected from 374 adult female water buffaloes originated from 14 production units located in 13 counties of the state of Pará. The sera were subjected to indirect fluorescent antibody (IFA), using titers of 200 and 64 as IFA cut off points for N. caninum and T. gondii, respectively. Hundred and fifty three (40.9%) buffaloes were sero-positive for N. caninum, with 100% of the properties being sources of infection. Four buffaloes (1.1%) in four farms were seropositive for T. gondii, and one buffalo cow (0.27%) was seropositive for the two parasites. The presence of antibodies indicates that these protozoan parasites are circulating among buffaloes from the properties studied, representing a source of infection to other animals, as well as a possible cause of reproductive disorders in this species.Item Acesso aberto (Open Access) Avaliação sorológica e molecular da infecção por Toxoplasma gondii em bubalinos da Ilha de Marajó naturalmente infectados(Universidade Federal do Pará, 2016-02-29) SANTOS, Cássia Maria Pedroso dos; PEREIRA, Washington Luiz Assunção; http://lattes.cnpq.br/9266281009742277In order to analyze the occurrence of Toxoplasma gondii in buffalo Marajó Island by serological and molecular method, blood samples and brain tissue of 100 buffaloes were collected. Blood sera were tested by indirect immunofluorescence assay (IFA) using a 1:64 dilution as the cutoff point for T. gondii, these samples were tested for the presence of antibodies IgG anti-T. gondii. To confirm the presence of T. gondii B1 gene in genomic material of blood and brain tissue samples were subjected to amplification by polymerase chain reaction (nested PCR). Among the tested sera buffaloes, 14 (14%) were reactive against T. gondii by the IFA technique, while blood samples and brain tissue tested 4- (4%) were amplified DNA by the technique of T. gondii PCR, 3 (3%) in blood samples and 1 (1%) in brain tissue samples. Despite the low occurrence of antibodies anti-T. gondii in serum buffalo, it can be concluded that infection by this parasite is present in buffalo breed in Marajo, moreover, it was possible to detect the genomic material of T. gondii in blood samples and brain tissue these animals, thus there is the need for further research, using the molecular technique for the diagnosis of toxoplasmosis since the detection of the bioagent buffaloes becomes even more relevant to the importance of these animals in the epidemiological cycle of toxoplasmosis as possible sources of infection primarily for the human being.Item Acesso aberto (Open Access) Distribuição das enzimas NADPH-Diaforase e Tirosina Hidroxilase na substância negra de camundongos infectados pelo Toxoplasma gondii(Universidade Federal do Pará, 2010-06-29) MEURER, Ywlliane da Silva Rodrigues; PEREIRA JÚNIOR, Antônio; http://lattes.cnpq.br/3239362677711162; NASCIMENTO, José Luiz Martins do; http://lattes.cnpq.br/7216249286784978; MORAES, Antonio Marcos Mello; http://lattes.cnpq.br/2806117046911976Toxoplasma gondii is a worldwide intracellular protozoan parasite and an important cause of disease in both humans and domestic animals. In Brazil, up to 80% of the population may be infected, depending on the region. Acute infection is generally asymptomatic, with posterior development of tissues cysts located mostly in muscles, retina and the central nervous system. The latent infection can alter behavior in animals and provoke psychotic symptoms in humans. Studies suggest that this infection may contribute to the occurrence of both schizophrenia and Parkinson’s disease, which are associated with abnormalities of the dopaminergic system. In this study we evaluated immunoreactivity for tyrosine hydroxylase (TH) and NADPH-diaphorase activities in the substantia nigra of infected male swiss mice. The animals were inoculated with bradyzoites (10 cysts) from a Toxoplasma gondii (Me-49 strain). The brains were removed after 30 and 60 days, sectioned and submitted to protocols to reveal NADPH-diaforase activity and immunohistochemistry anti-TH. The densitometric analysis showed a lower reactivity to NADPH-diaforase in the substantia nigra of infected animals when comparated with non-infected controls. A similar reduction was also observed in immunoreactivity against TH in the substantia nigra. The study was conducted in all Para meso, they were harvested blood samples from animals in 2070, with 1750 and 320 bovine and buffalo, respectively, coming from 52 municipalities and 100 properties, a survey of the epidemiological aspects influencing the onset of the infectious agent. Sera of animals underwent the test of ELISA Kit – Enzyme-Linked Immunosorbent Assay and indirect immunofluorescence - IFA for the detection of specific IgG antibodies to T. gondii cattle and buffalo the Imunodot® company. The animals were considered positive in the IFAT test from the dilution 1:64. It was considered statistically significant difference p 0.005. The buffaloes had higher seropositivity that the bovine, as well as the IFA test was higher than the ELISA test, and Mesoregion more seropositive was the Lower Amazon. There were no significant differences in the frequency of T. gondii, in relation to sex, type of exploitation, but no significant association for age, size of property, number of animals in the property, creation cycle, and the properties that presented greater amount of cats and dogs were the ones that had higher sororreagência the anti - T. gondii.Item Acesso aberto (Open Access) Pesquisa de anticorpos anti-Toxoplasma gondii (Nicolle & Manceaux, 1909) em felídeos selvagens nos municípios de Capitão Poço e Belém, Pará(Universidade Federal do Pará, 2008-02-29) SILVA, Carolina Costa; SOUZA, Nazaré Fonseca de; http://lattes.cnpq.br/1616332835810182Toxoplasmosis is one of the most widespread zoonosis in the world caused by Toxoplasma gondii, a protozoan that has in felids its unique and definitive host. Twenty one animals of four species were avaluated, Jaguarundi (Herpailurus yaguarondi), Ocelot (Leopardus pardalis), Margay (Leopardus wiedii), Jaguars (Panthera onca) to investigate the occurrence of toxoplasmosis in two municipalities of Pará State, using two serological assays, indirect hemagglutination, (IHA), direct modified agglutination (MAT) and coproparasitologic examination. From the tested animals, 18 were positive (85,72%). Twelve (57,14%) animals by the IHA and 14 (66,66%) by the MAT, were seropositive. In both utilized techniques there were no statistical differences between seropositivity and the genus. In the Herpailurus genus 4,6% of seropositivity was found in both tests; in the Leopardus 19,05% by IHA and 28,57% by MAT; and in the Panthera 33,33% in both techniques. Similar results were verified in 11 animals. Comparing both techniques, there were no statistical differences. Higher titers were obtained from a margay (1024) by MAT. No oocysts of T. gondii were found in the studied animals. It was verified high occurrence of toxoplasmosis in the studied areas and both techniques are efficient for serologic diagnoses of this disease.Item Acesso aberto (Open Access) Prevalência de anticorpos anti-Toxoplasma gondii em búfalos (Bubalus bubalis) no estado do Pará(2013-05) SILVA, Jenevaldo Barbosa da; FONSECA, Adivaldo Henrique da; ANDRADE, Stefano Juliano Tavares de; SILVA, André Guimarães Maciel e; OLIVEIRA, Carlos Magno Chaves; BARBOSA NETO, José DiomedesThe aim was to study the seroprevalence of Toxoplasma gondii in water buffaloes (Bubalus bubalis) from State of Pará, Brazil. Three hundred and nineteen buffaloes were randomly selected into seven municipalities of Marajó Island. For comparative purposes, 128 buffaloes of five municipalities in the state of Pará were also evaluated. The seroprevalence of T. gondii was evaluated by Indirect Enzyme Linked Immunosorbent Assay (iELISA). The samples diagnosed as positive in iELISA were subjected to Immunofluorescence Antibody Test (IFAT). We evaluated risk factors: location, breed, pregnancy and co-infection with Brucella abortus or Mycobacterium bovis. The frequency of animals positive for T. gondii in iELISA were compared by chi-square (χ2) with 95% confidence. Variables with p <0.2 were subjected to logistic regression analysis; the model was built based on the "odds ratios" test. The prevalence of T. gondii in iELISA was 41,6% (186/447). In IFAT, 86,5% (161/186) had their positivity for T. gondii confirmed. The average prevalence in the municipalities of the Marajó Island and of the mainland was 32% (103/319) and 55% (70/128), respectively. The municipalities with the highest prevalence were Soure (53%) and Salvaterra (49%) in Marajó Island, and Castanhal (55%) and Thailândia (50%) in the Continent. The breed and co-infection with Brucella abortus or Mycobacterium bovis presented no influence on the prevalence of T. gondii. Additionally, pregnant animals were 57% more positive for T. gondii than nonpregnant animals. The presence of antibodies is an indicative of T. gondii in buffaloes in the state of Pará, and these findings represent a risk not only for farm animals, but to public health as a source of infection.
