In Marudá, Marapanim-PA, the Women's Group "Erva Vida", produces and markets the "Tintura de pata-de-vaca" consumed by tourists and locals to treat diabetes. This remedy is prepared from leaves of Bauhinia monandra Kurz (pata-de-vaca). Different metabolic classes of plant already had a proven hypoglycemic action, for example flavonoids. Thus, the aim of this study was to characterize the handmade medicine through pharmacobotanic parameters pharmacognostic, phytochemicals and chromatographic in compararação with a tincture of Bauhinia monandra, for the presence of flavonoids. Both the preliminary phytochemical analysis, and the analysis by Thin Layer Chromatography, phenolic metabolites detected, possibly flavonoids in handmade medicine and hydroethanolic extract of plant species. In the analysis by HPLC, it revealed the presence of rutin in handmade medicine as well as in hydroethanolic extract of Bauhinia monandra.Item Acesso aberto (Open Access) Análises cromatográficas por CLAE-DAD, termogravimétricas e morfo-anatômicas de amostras comerciais de Echinodorus macrophyllus (Kunth) Micheli (Chapéu-de-couro)(Universidade Federal do Pará, 2010) FREITAS, Cristian Barbosa de; BORGES, Fernanda Iikiu; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070For quality control of the commercial sample of Echinodorus macrophyllus were analyzed with botanical, pharmacognostic and physical-chemical properties. To achieve the anatomical cuts was rehydrated, sectioned with steel blade and fixed on slides semi-permanent. The sections were cleared in 20% NaClO and stained with astra blue-to-follow basic fuchsin. In cross section the epidermis is uniseriate with cells of rectangular shapes, heterodimensional, smooth and thin cuticle. For realization of thermogravimetry was used both drugs as solids at a heating rate of 5, 10 and 15 degrees Celsius. The chromatography was performed in HPLC and sample was obtained from column chromatography and eluted by gradient with methanol and acidic water. The palisade tissue consists of two layers of cells. The parenchyma spongy cells have thin walls, heterodimensional, different formats and well-developed intercellular spaces. The vascular bundles are collateral. The leaf is amphistomatic with stomata paracytic. Petiole in cross section, has a triangular to hexagonal. The epidermis is uniseriate with cells of polygonal shape, smooth walls, large amounts of aerenchyma which sometimes occur diaphragms. Samples of the leaf have characteristics of the species E. macrophyllus, but the anatomy of the petiole showed different structures from those described for the species, which belong to the genus Echinodorus, however suggests possible contamination of the material supplied with other plant species. Thermogravimetric analysis contributed significantly to establish parameters and characterize the sample.Item Acesso aberto (Open Access) Contribuição para o estudo microquímico e anatômico da casca e validação de um método espectrofotométrico para quantificação de alcalóides totais de Himatanthus lancifolius (Muell. Woodson(Universidade Federal do Pará, 2010-01-22) BARROS, Patricia Miriam Sayuri Sato; BORGES, Fernanda Iikiu; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070For the registration of phytomedicines and their availability to the users, ANVISA establishes quality, security and effectiveness parameters, stipulating control requirements similar to that ones applied to synthetic medicines. This work reports the investigation of the bark of Himatanthus lancifolius and its extracts aiming to contribute for the standardization of derivatives of this plant species, the assays were developed using commercial samples Q and S, and an authentic one provided by EMBRAPA – E. The microchemical, anatomical and chromatographic comparison of Q, S and E does not allow characterising both the commercial sample as H. lancifolius. The presence of ulein could not be characterised in any sample, even in E. The TLC and HPLC data show that the three samples have different chromatographic profile. The morphological and the microchemical characterisation of the barks of H. lancifolius shows cortex with groups of fibres of the primary phloem included, with crystals and lactifers in the region of the secondary phloem. The developed quantification method shows high selectivity at 281nm, what confers confidence for the detection of the alkaloids. The method is robust, according to the current regulation, and shows linearity, precision and accuracy, beside accessibility and is easy to execute. The total alkaloid fraction present in the aqueous extract of the collected sample (E) represents 0,219% of the dried extract. The author expects to have contributed for reducing the lack of methods for the quality control of phytomedicines from Himatanthus lancifolius.Item Acesso aberto (Open Access) Desenvolvimento de um método para caracterização do extrato hidroetanólico das folhas de Mikania lindleyana DC por Cromatografia Líquida de Alta Eficiência (CLAE)(Universidade Federal do Pará, 2011) OWITI, Alex Oselu; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070Mikania lindleyana DC (Asteraceae), vernacular “Sucuriju”, is a creeping, shrubby, perennial, evergreen plant, growing in the Amazon region where it is used as diuretic, anti-hypertensive, anti-inflammatory and anti-ulcerous medicine. The main aim of this study was to develop a method for characterization of hydro-ethanolic extract (tincture) from the leaves of M. lindleyana DC, using UV and High performance Liquid Chromatography (HPLC). The hydro-ethanolic extract (tincture) was prepared based on Brazilian Pharmacopeia V, 2010 and its dry residue was submitted to phytochemical analyses, Thin Layer Chromatography (TLC) and High Performance Liquid chromatography (HPLC). Among the substances observed in the phytochemical analysis was coumarin, alkaloids, amino acids, reducing sugars, phenols, tannins, steroids, terpenes, saponins and organic acids. TLC test was done by applying hexanic, chloroformic and ethyl acetate Fractions together with Crude hydro-ethanolic extract and aqueous solution of coumarin Aldrich® 0,1mg/ml on the same plate using toluene/dichloromethane/acetone (45:25:30) as the eluent system. The chromatogram was then sprayed with an alcoholic solution of KOH 5%, and then observed under UV light at 254nm and 365 nm, revealing a light green band at an RF-0.61 for chloroformic fraction, crude hydro-ethanolic extract and for coumarin samples. The HPLC analysis was conducted using methanol/water 53:47 as the eluent system. The chloroformic fraction(CL), coumarin and the crude hydro-ethanolic extract (CHE) showed peaks at Retention time around 6.00minutes and spectra with maximum absorptions between 270nm and 300nm.This clearly demonstrate the presence of coumarins in the samples (CL= 0.209mg/mL and CHE= 0.014mg/mL).Item Acesso aberto (Open Access) Investigação fitoquímica biomonitorada da tintura 70% de Arrabidaea chica (Humb. & Bompl.) Scandentia is a small shrub about 2.5 m high, occurring from southern Mexico to central Brazil, is common in the Amazon, where it is used by Indians for body painting.Popularly tea leaves is indicated for inflammation, anemia, intestinal cramps, regulate menstruation, diarrhea, among others. Recent studies have shown its antioxidant activity and wound healing in vivo and in vitro, and antifungal activity against Trichophyton mentagrophytes and trypanocidal effect. This paper aims to contribute to the validation of the claim of use and to standardize the form of employment A. chica herbal medicine as a resource, since it is in RENISUS. The ethanolic and aqueous extracts of the plant were investigated for their secondary metabolites. Parameters were determined for quality control of the plant drug, through grading, drying powder loss and total ash.Besides the chromatographic profiles obtained by TLC, CLV, LC-MS and LC - DAD. The plant meets the parameters of quality control in accordance with the literature. A CCD profile obtained with the eluent Hexane / Acetone / MeOH (67,5:27,5:5) and revealed with NP / PEG revealed a band with Rf = 0.17 with yellow fluorescence characteristic of flavonoids. In the profile of HPLC - DAD chromatograms were obtained with characteristics of quinones and flavonoids as well. From these results, the work was directed to the isolation, initially with the FAE by fractionation of CLV and analysis of resulting fractions by HPLC-DAD, and as a result suggested the presence of a flavonoid apigenin derivative of the flavonol. The microbiological results corroborate some of its popular uses thus contributing to validate the form of alleged use of the plant.Item Acesso aberto (Open Access) Investigação fitoquímica e atividade biológica das cascas de Luehea speciosa willd(Universidade Federal do Pará, 2017-12-21) NASCIMENTO, Maisa Carmen Batista do; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070The barks of Luehea speciosa, family Malvaceae, are used in traditional medicine to treat dysentery, rheumatism, tumors, bronchitis, skin wounds, weight loss and others diseases. However, studies give little information about the species in the literature. Thus, it is of great interest to investigate its chemical constitution and biological activities, since the scientific proof of its activity can promote, in the future, a therapeutic alternative. The present study analyzed the chemical constitution of the dry crude extract (EBS) and hexane (FHX), dichloromethane (FDM), ethyl acetate (AED) and methanolic (FME) fractions of the species by high-performance liquid chromatography (HPLC) with tandem mass spectrometric - LC/MS, as well as its antioxidant activities by bioautography and cytotoxic assays, in vitro, against HELA, HEP-2 and VERO tumor cell lines. The samples showed antioxidant activity, being the fractions FDM and AED results more relevant. The EBS and fractions demonstrated cytotoxicity on all cell lines, however the FDM fraction had greater cytotoxic activity against tumor cells. In the LC/MS analyzes, were detected phenolic compounds and suggestive characteristics of chicoric acid presence through m/z = 148, as in the UV spectrum. These results demonstrate that this species has antioxidant activity and suggest its cytotoxicity against tumor cells, being a promising species for future studies.Item Acesso aberto (Open Access) Investigação fitoquímica e avaliação da atividade anti inflamatória do extrato hidroetílico de folhas de salix humboldtiana em modelo zebrafish (Danio rerio)(Universidade Federal do Pará, 2021-09-25) RAMOS, Uêbem Fernandes; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070; https://orcid.org/ 0000-0002-2421-8245The leaves of Salix humboldtiana, family Salicaceae, are used in traditional medicine for the treatment of various inflammatory diseases, febrile states, among others. However, there is little information about the species in the literature. However, the genus Salix has a very significant diversity of uses in popular medicine, with Salix alba L. as its most renowned representative, which is included in the Brazilian Pharmacopoeia Form of Herbal Medicine. This work proposes to investigate chemical constituents with an emphasis on phenolic compounds, through pharmacognostic studies (analysis of physical-chemical quality); the phytochemical approach, the tests, served to demonstrate the presence of classes of secondary metabolites: reducing sugars, phenols and tannins, flavonoids, catechins, steroids and coumarin derivatives; fractionation, CCD and CLAE-DAD. In addition to biological testing on an animal model (D. rerio) to investigate anti-inflammatory activities. In this sense, the objectives of performing the phytochemical analyzes and the bioassay, the Salix humboldtiana Ethyl Extract (EBSh) was used in different doses in in vivo tests. Groups previously treated with 75 mg / kg EBSh showed 5% inhibition; 150 mg / kg showed 14% inhibition; 300 mg / kg showed 54% inhibition; 500 mg / kg showed 60% inhibition, 750 mg / kg showed 43% inhibition of abdominal edema compared to the negative control. Among the five different concentrations of EBSh tested and evaluated in the zebrafish model, the concentration of 500 mg, provided a statistically more expressive response regarding anti-inflammatory activity and anti-edematogenic degree after 5 h of treatment. This response was superior to the indomethacin standard when compared to the negative control, thus indicating the ability to reduce and / or inhibit edema in the peritoneal region. The animals subjected to the tests showed changes in normal behavior at all doses tested, these changes varied according to the concentration of EBSh administered. In the three lowest doses (75 mg, 150 mg and 300 mg), the fish showed only mild to moderate changes and less frequently, while in the two highest concentrations (500 mg and 750 mg), they manifested important behavioral changes, some with greater frequency and intensity.Item Acesso aberto (Open Access) Investigação fitoquímica e das atividades antioxidante e antiparasitária do extrato e frações de Aspidosperma excelsum Benth(Universidade Federal do Pará, 2013-09-30) NASCIMENTO, Myrth Soares do; VASCONCELOS, Flávio de; http://lattes.cnpq.br/3695753129639448; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070Neglected diseases are diseases that affect a large number of people around the world. These include Chagas disease, leishmaniasis and malaria, whose therapy used, in addition to reduced presents problems that hinder their treatment and show the need to search for new therapeutic options. Aspidosperma excelsum (Apocynaceae), popularly known as carapanaúba, is used in the popular herbal medicine for the treatment of malaria. Its chemical constitution and antiparasitic activity of other species of genus suggests its antiparasitic potential. To investigate their chemical constitution and possible biological activities, the lyophilized extract, obtained from barks of specie, and their alkaloidic fractions were analyzed by chromatographic techniques and evaluated for antioxidant and antiparasitic activity against Leishmania ssp., Trypanossoma cruzi e Plasmodium falciparum. The HPLC analyses showed peaks highly correlated to yohimbine in the tincture and in its alkaloidic fractions: FAlk7, FAlk10 and FAlk14. All the samples were active against L. chagasi and FAlk1, FAlk4 and FAlk7 show IC50 values lower than Amphotericin B against L. braziliensis. Fractions FAlk1 and FAlk4 were also active on P. falciparum, showing high selectivity to the parasite. Evaluation of antioxidant activity revealed important capacity of samples in capturing free radicals, characterizing them as antioxidants.Item Acesso aberto (Open Access) Isoeleuterol e isoeleuterina: potenciais marcadores químicos da tintura de Eleutherine plicata Herb (Iridaceae) e atividades microbiológica e antioxidante(Universidade Federal do Pará, 2008-12-30) MALHEIROS, Luiz Claudio da Silva; VIEIRA, José Maria dos Santos; http://lattes.cnpq.br/6807452375674442; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070Eleutherine plicata Herb. is an Iridaceae, popularly known as marupazinho, widely used by people to treat diarrhea. With the bulb of the plant is about a cup of tea, which is used to treat infestations caused by amoeba. The plant material used in this study was collected in Belém do Pará and their identification by comparison of botany was deposited in exsiccates Paraense Emilio Goeldi Museum under nº 10543. The ethanol extract was prepared by percolation from the bulb previously dried and crushed. After drying the ethanol extract was suspended in a solution hydroalcoholic (1:1) and subjected to partition with solvents of increasing polarity. With the ethanol extract and the fractions were performed eighteen tests to detect classes of secondary metabolites. The ethanol extract and the fractions hexanic and chloroform, showed a positive result for naphthoquinones, anthraquinones and steroids and triperpenoids. The analysis by thin-layer chromatography fractions of ethanol extract and hexane and chloroform, showed areas susceptible to methanolic solution of KOH 10%, indicating the presence of quinones in these samples. The evaluation of the antimicrobial activity of ethanol extract and fractions hexanic and chloroform with strains of C. albicans, S. aureus, E. coli and P. aeruginosa, showed that the chloroform fraction is the most active, presenting the largest halos of inhibition of microbial growth, possibly containing a higher concentration of active constituents. The chemical constituents isoeleutherol and isoeleutherine were isolated from chloroform fraction, and were characterized chemically using RMN 1H e 13C, compared with the literature. The ethanol extract, isoeleutherol and isoeleutherine were submitted to evaluation of their antioxidant activies, and showed weak activity when compared with BHT.Item Acesso aberto (Open Access) Isolamento e quantificação de flavonóides e abordagem das atividades antioxidante e antimicrobiana de Jatropha gossypiifolia L.(Universidade Federal do Pará, 2008-12-30) VEIGA, Andrex Augusto Silva da; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070Jatropha gossypiifolia L (Euphobeaceae), known in Pará as “Pião roxo” is used for treatment of hemorroids, burns, stomach pains among other diseases, the importance of this plant to this region has led to its research in order to improve its phytotherapeutic development. During the analysis, the chromatographic profiles of its Crude Ethanolic Extract (CEE)was determined using Thin Layer chromatography (TLC) and High Performance Liquid Chromatography (HPLC) methods. The total and isolated flavonoids quantities were determined. Antimicrobial activity essays of the crude ethanolic extract were also conducted and the fractions obtained (hexanic, chloroform, ethyl acetate and residual fractions). The antioxidant activities of the extract and the fractions were also characterized. The plant materials were collected from EMBRAPA medicinal plants garden in the east Amazon and then extracted through percolation to obtain crude ethanolic extract. The dry extract was obtained using low pressure rotative evaporator followed by liquid-liquid partition with hexane, chloroform and ethyl acetate successively. The fractions obtained were also concentrated at low pressure, The ethyl acetate fractions were re-fractioned, giving rise to acetone fraction (ActF-II), a new ethyl acetate (AF-II), methanolic fraction (MF-II) and a solid residue (SR-II). ActF-II was submitted to liquid chromatography at average pressure, resulting in sub-fractions like Fr ActS-II and Fr ActS3-II called Jg1 and Jg2, respectively. Chemical prospection of the Extract and the metabolite fractions were conducted in triplicates, leading to the observation of the presence of the following polyphenols: Catechines, Tannins and flavonoids. The analysis by thin Layer Chromatography (TLC) was conducted using eluent of ethyl acetate/formic acid/glacial acetic acid/water at (100:11:11:26) on a normal phase silica gel, showing the retention factors in the following zones:0.65, 0.72, and 0.77 in reaction to Fecl3 solution . The HPLC chromatogram of ethanolic crude extract presented three significant peaks with average values of RT: 13.2 min, 15.02 min, and 16.00 min whose spectra are characteristics of flavonoids. On calculating the flavonoid quantity using the average absorbance reading (A=0.874), 2.02% flavonoid was found. The concentrations for each isolated substances was 340ug for Jg1 and 406 ug for Jg2 per milliliter of extract solution. Regarding the biological activity tested AF-1 and Fr-1 inhibited growths of S. aureus and C.albicans and also manifested a great antioxidant potential. In the same way, in some states in Brazil official programs of medicinal plants have been registered and to follow the plant species used in the folk phytotherapy ethnoguided methodologies are applied mainly ethnobotany and ethnopharmacology. From the Pharmaceutical Sciences Ethopharmacy was elaborated, which has been worldwide used to approach medicinal plants and to evaluate the remedies prepared from them. The general aim of this work is to contribute for the consolidation of Ethnopharmacy and the implementation of the PNPMF in the state of Pará, bringing information collected about indication of medicinal plants, preparation and administration of vegetal remedies and problems related to the use of them. More specifically, it aims to determine the regional nosogeography, to identify and to select the used phytotherapeutical resources for the treatment of the prevalent illnesses, to know and to characterise from the pharmaceutical point of view the remedies made with vegetable base, as well as elaborating cards with information about selected vegetables, these are other objectives considered here. The tool to reach these objectives is the ethnopharmaceutical survey which associates elements of other ethnoguided methods with the content of the Pharmaceutical Care. Applied in eighty six communities of the city, at two moments, first in the year 2000 and in March/April 2008, the method supplied data which were tabulated using Excel to make tables and graphs. Thus, 91 illnesses, 236 plants species, distributed through 72 botanical families, and in 7 different forms of administration. Thirty eight adverse events could also be identified. Cards with botanical, chemical and ethnopharmaceutical information were elaborated for the four plants species with highest relative citation frequencies.Item Acesso aberto (Open Access) Utilização de técnicas hifenadas para o desenvolvimento de programações para analise de Acmella oleracea (L.) RK Jansen(Universidade Federal do Pará, 2016-08-31) SILVA, Ianna Dias Ribeiro da; REBELO, Monaliza Maia; http://lattes.cnpq.br/2064401199679916; BARBOSA, Wagner Luiz Ramos; http://lattes.cnpq.br/1372405563294070The use of hyphenated techniques that allow the separation and isolation of substances of a herbal extracts is necessary both for the knowledge of the chemical composition of a compound, as well as for the determination of a substance or a group that serve as marker of particular species. The species Acmella oleracea (l.) R. K. Jansen, grows in Legal Amazon, a geographic region in northern Brazil, where is known as Jambu, Agrião Bravo ou Agrião do Pará. It is used in folk medicine in the form of tea, syrups and tinctures and indicated for use as anesthetic and antiseptic. The aim of this study was to use the HPLC-UV-ELSD and HPLC-DAD-MS for the development of methods that can identify metabolites of interest on Acmella oleracea. The plant material was acquired in Tailândia, State of Pará, in the Santa Inês farm, city of Tomé-Açu, PA150 highway margins, positioned the 260 km from Belém, at 02° 56 ' 22 '' south latitude and 48° 57 ' 03 '' west longitude, and consisted of 10 packs of Acmella oleracea (jambu), composed of leaves, flower, root and stem, after cleaning the material was grinded and then macerated for 7 days with 92.8° GL ethanol at a ratio of 1:10. Hydroethanolic extracts, of each part of the plant, were filtered and concentrated at a low pressure and then lyophilized for further analysis with hyphenated techniques. The Spilanthol was registered in the analysis by HPLC-DAD-ESI-MS, Scan mode and Product ion, through the detection of ion m/z 222 in all analyzed organs. A substance which showed m/z 376.80 for analysis with etoh/water and m/z 388.20 for acn/water, which has promising profile was found through the analysis by HPLC-DAD-MS and needs specific experiments for confirmation of flavonoids. During the development of methods, it was observed that the best chromatographic profiles of EEAOFOC and EEAOR were obtained when the mobile phase was composed of ethanol and water.
