Hepatotoxicity was investigated, concentrations of mercury in total blood were monitored each 30 days using atomic absorption spectrometry with cold vapor Hg201, aspartate transaminase (AST), alanine transaminase (ALT) and total bilirubin (BT) were determined. Liver was fixed by formaldehyde 10% and prepared by histopathology protocols. Significant difference was observed in groups exposed and control about total mercury (Hgtotal) in the periods of 60, 90 (P < 0,05) and 120 days (P < 0,01). The histopathology revealed moderate steatosis and hydropic degeneration, common in methylHg exposed in other species. No Significant difference between the levels of AST (p= 0.38), ALT (p= 0.83) and BT (p= 0.07) in groups exposed and control. The Pearson correlation with Hgtotal was negative (AST r= -0,7; ALT r=0,07; BT r= -0,3 e p > 0,05), suggests another studies to clarify the alert levels of mercury concentrations and liver dosages.Item Acesso aberto (Open Access) Alterações histopatológicas dos rins de macacos prego, Cebusapella (Linnaeus 1758) após exposição crônica a baixas doses de metilmercúrio(Universidade Federal do Pará, 2014-02-28) SOUSA, Andréa do Socorro Campos de Araújo; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219Mercury has been a major environmental and occupational risk and it still remains a problem for human health in the Amazon region. Although studies have shown that mercury affects various tissues and organs, kidneys are the target organs to the metal toxicity. Thus, the aim of this study was to investigate the effects of chronic exposure to low doses of methylmercury on renal parenchyma of Cebusapella, adult males exposed during 120 consecutive days with daily oral doses of 1.5 μg in the diet. The concentrations of total mercury in the animals’ blood were monitored every 30 days using a cold vapor atomic spectrophotometer (201 Hg), compared to the control group. The method used for histopathological analysis was the immersion in paraffin for staining with hematoxylin and Eosin, Masson's CAB and PAS. The immunohistochemical investigations included reactions for detection of smooth muscle actin ( IA4 ), muscle actin ( HHF35 ) and cytokeratin (AE1 and AE2). The results showed that treatment with mercury caused significant differences (P < 0.001) between the exposed and control groups. As for total Hg levels, histopathologicalchanges just likehydrops in Proximal tubuleswere observed, a common finding in methylmercury exposure in other species, with no significant changes in creatinine and urea concentrations. The Person correlation test showed a strong negative relationship between mercury concentration and animal body weight loss (P < 0.0001). Another important finding was the decrease in mesangial cells number, which suggests that methylmercury executed its nephrotoxicity, affecting not only the renal tubular system, as well as the glomerular mesangium cells, making necessary a greater amount of experimental studies to clarify which mercury concentration alert level is capable of triggering aggression mechanisms and kidney injury in subjects exposed to methylmercury.Item Acesso aberto (Open Access) Avaliação da razão sinal ruído em eletrorretinografia multifocal: descrição do método e aplicação em toxoplasmose ocular(Universidade Federal do Pará, 2009) CARVALHO, Aline Correa de; SOUZA, Givago da Silva; http://lattes.cnpq.br/5705421011644718; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219The purpose of this work was to develop a quantitative analysis that allows, in healthful and patient with visual loss, to separate registers with signal correlated with the visual stimulation, of the registers that contain noise. System VERIS Science v6.0.5d was used to extract kernel first-class of the registers, and posterior exportation of the data for analysis in the MATLAB. In this environment of programming was carried the analysis of the main components (PCA), that is used to reduce the dimensionalidad of the data conserving its variation and minimizing the influence of the noise; the signal-to-noise analysis (SNR) of the multifocal responses (mfERG) using cumulative distribution SNR carried through in two intervals of time, one understanding only the signal and another one only the noise, for classification of the valid, diminished or absent responses. We have found that in a group healthy subjects, the noise and signal SNR cumulative distributions have not overlapped and occupy distinct ranges of SNR values in all subjects. By using a 1% criterion for false positives and false negatives, it was possible to define the borders of noise, low response, and signal SNR ranges. We have also applied this protocol to a three subject with visual loss due to ocular toxoplasmosis scares. In this case, there was a degree of overlapping between the noise and signal SNR distributions. It was still possible to separate the noise, low response, and signal SNR range by using the confidence limits for the distance between noise and signal boundaries obtained from the group of healthy subjects. The results of SNR evaluation were then used to display the mfERG waveforms in topographical plots that discriminate between reliable responses (signal), low responses, and no responses (noise). Using this approach, features of retinal topography, such as the optic nerve head, are more easily discriminate, whilst artifacts such as an erroneous central peak, are more easily rejected. The combinations of PCA to reconstruct mfERG waveforms with SNR evaluation are useful tools to analyze retinal topography both in healthy and impaired conditions.Item Acesso aberto (Open Access) Avaliação psicofísica da visão espacial cromática e acromática de pacientes pós-operatórios de microcirurgia vascular para aneurismas intracranianos do sistema carotídeo(Universidade Federal do Pará, 2017-07-01) BASTOS, Albedy Moreira; RODRIGUES, Anderson Raiol; http://lattes.cnpq.br/4030747999301402; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219The intracranial saccular aneurysms constitute the main cause of spontaneous meningeal or subarachnoid hemorrhage. The complications derived from the sprouting of such aneurysms hemorrhage, classified as sensorial (acuity and visual field), motor (motility of extra-ocular muscle), and perceptual, are considered as well studied by neurosurgery due to relative similarity between cases. However, little is known about possible postoperative perceptual and cognitive alterations. In the present study, the visual sensitivity of patients in postoperative condition of intracranial aneurysm of the carotid system to achromatic and chromatic spatial stimuli was evaluated. The contrast sensitivity to luminance spatial gratings was measured in eleven spatial frequencies (0,2, 0,5, 0,8, 1, 2, 4, 8, 10, 15, 20, and 30 cycles/degree). The color discrimination ability was measured by a version of the Farnsworth- Munsell test with 85 hues, divided in four series of stimuli presented sequentially, being one serie with 22 stimuli and three series with 24 stimuli. For the measure of color discrimination thresholds, 5 MacAdam’s ellipses were determined and, for each one of them, discrimination thresholds had been measured in 20 directions of the CIE 1976 color space, through the test of Mollon-Reffin. The studied variables were compared with the statistical norms of agematched control subjects. The results show that, individually, there are not significant losses of spatial contrast sensitivity. However, individual losses in color discrimination ability and individual alterations in color discrimination thresholds in all color confusion axis (protanopic, deuteranopic and tritanopic) were detected. Thus, it can be concluded that color vision have sensorial parameters which measurement can be useful in the evaluation of patients postoperative of intracranial aneurysm of the carotid system.Item Acesso aberto (Open Access) Eletrorretinografia multifocal: desenvolvimento de método avaliativo e aplicação em pacientes com toxoplasmose ocular e investigação funcional da função retiniana em pacientes com malária, infectados pelo P. The first purpose was the development of a reliable method to classify the electroretinographic responses and to validate this method in ocular toxoplasmosis. The second purpose was to investigate the effect of the acute use of chloroquine in the retina of vivax malaria patients, which received the standard treatment of the Ministry of Health of Brazil. The experimental tool of both studies was the multifocal electroretinogram (mfERG). In the first study the Veris system was used to test 10 healthy subjects to calculate a signal to noise ratio analysis in order to find the best values to separate normal responses from impaired retinal responses. The final results of this analysis were applied in three patients with visual loss due ocular toxoplasmosis. The results showed an optimal signal to noise ratio of 0.47 to separate waveforms with and without signal. The signal to noise ratio of 0.47, 0.44, 0.48, 0.57, 0.78, 1.21 were optimal to separate normal responses from progressively attenuated responses. The application of the signal to noise ratio analysis into ocular toxoplasmosis patients was able to map the retinal area with no activity evoked by the light. The second study was performed using mfERG to evaluate 48 vivax malaria patients which received chloroquine-based treatment. Their data were compared to 37 healthy subjects. The results showed that number of infections ranged between from 01 to 18 times. During the treatment, the mean dosage of intaken chloroquine was 5429 ± 782.3 mg. The cumulative dosage normalized to the body mass ranged from 14.58 to 545.45 mg/kg. No difference was observed in the mfERG amplitude and implicit time obtained from malaria patients and control group at any concentric rings of the visual field. There was no significant correlation between the retinal activity with the chloroquine cumulative dosage. It was concluded that the signal to noise analysis developed in this thesis can be used as a reliable method to study impaired retinal function, including the mapping of ocular toxoplasmosis consequences; and the acute use of chloroquine for malaria treatment caused no impairments to retinal activity recorded by mfERG.Item Acesso aberto (Open Access) Interferentes bio-sócio-ambientais na exposição ao mercúrio em crianças ribeirinhas de diferentes regiões da Amazônia(Universidade Federal do Pará, 2011-06-27) AMORAS, Walter Wanderley; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219The present study examined possible bio-socio-environmental factors interfering in mercury exposure in children riverside. 103 children participated in the regions of the Tapajos and Acará Rivers and Marajó Island. The type of study was performed cross-sectional analysis. Hair samples were collected for analysis of the Hgtotal levels, blood (hemoglobin and hematocrit analysis) and faeces. Anthropometric indices were used to analyze children's growth. In the evaluation of psychomotor development test was applied modified Denver II screening. The highest average of Hgtotal levels in hair samples of children was in the Tapajós region (5.58 mg / g) and 0.65 g / g in other locations. The prevalence of Hgtotal> 10 mg / g was 25% and 7.5% in São Luiz do Tapajós and Barreiras. Daily consumption of fish by the families of the children as it relates to levels of Hgtotal p (value) was <0.05. The prevalence of breastfeeding duration in the groups of children under 12 months and greater than 6 months for locations in the Tapajos River, when Hgtotal related contents in hair of children with p (value) <0.05. The prevalence of anemia in the Tapajós region was 46.7% and when related to Hgtotal levels p (value) <0.0001. The parasitological tests indicated that 68.3% were multiinfested frequent when related Hgtotal p (value) <0.05. The profile of growth observed through the nutritional indicators status was 82.6% normal weight for age and 14.5% with very low weight and low. Psychomotor performance assessed by the Denver II screening test had changed five children as suspicions of developmental delays. It was concluded that in locations around the gold mining activities riverine children are at risk of mercury contamination. Bio-social- environmental factors interfering in exposure to mercury such as daily consumption of fish, length of breastfeeding, anemia, intestinal parasites showed significant statistical relationships with Hgtotal in the children's hair.Item Acesso aberto (Open Access) Investigação de danos visuais em pacientes diagnosticados com meningite criptocócica não associada à imunossupressão(Universidade Federal do Pará, 2014-03-20) LACERDA, Eliza Maria da Costa Brito; SOUZA, Givago da Silva; http://lattes.cnpq.br/5705421011644718; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219Cryptococcal meningitis is a severe infectious disease caused by Cryptococcus spp., which has high lethality and causes sensorial sequels. The most important sequels are visual impairments. The aim of this study was to describe the visual losses suffered by patients without immunosupression history, diagnosed with cryptococcal meningitis in order to indicate a possible mechanism and risk factors for the visual sequels. The work was composed by a case series study with cryptococcal meningitis without immunosupression history (n =7 patients, 14 = eyes) and an epidemiological study of all the cases of cryptococcal meningitis without immunosupression history notified during 14 years in the reference hospital of Para State (n = 113 cases). In the case series study, the visual functions of a sample of patients were studied by ophthalmological, psychophysical, and electrophysiological evaluation. The epidemiological study analyzed the medical records data focusing on visual impairment. It was observed that the studied patients in the case series study had visual acuity impairment. Even in patients without clinical complaints, color discrimination, luminance contrast sensitivity, and visual field were impaired. The results indicated central retina commitment as the principal responsible for a cascade of alterations that prevented the normal upstream image processing, at the visual cortex level. It is suggested that the observed visual dysfunction was due not only to optic nerve damage. The principal risk factors for the visual alterations observed in the epidemiological study were disease time before the beginning of the treatment and patient immunological response.Item Acesso aberto (Open Access) Investigação do potencial cortical provocado visual para padrão reverso em pacientes diagnosticados com epilepsia parcial e generalizada(Universidade Federal do Pará, 2011-12-02) DUARTE, Regina Célia Beltrão; SOUZA, Givago da Silva; http://lattes.cnpq.br/5705421011644718; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219The present work evaluated the visual evoked cortical potential of children with clinical history of epilepsy in order to identify electrophysiological marker indicating cortical changes in epilepsy. Thirty-four subjects with history of epilepsy along the lifetime (18 subjects diagnosed with partial epilepsy and 16 subjects diagnosed with generalized epilepsy). The control group was composed by 19 subjects age-matched with no history of epilepsy. Visual evoked cortical potential components for pattern-reversal presentation of chessboards were evaluated in amplitude, implicit time, and amplitude ratio of components. It was observed that generalized epilepsy patients had larger N75 than other two groups, whilst N75/P100 and P100/N135 ration were smaller in partial epilepsy patients than other groups. There was weak correlation between the electrophysiological parameters and the age of seizures onset or time of antiepileptic drugs intake. In conclusion, N75 amplitude and amplitude ratio can be good electrophysiological markers to cortical changes in epilepsy.Item Acesso aberto (Open Access) Parasitoses intestinais e desnutrição em crianças expostas ao mercúrio na Amazônia(Universidade Federal do Pará, 2003) XAVIER, Fábio Branches; SILVEIRA, Luiz Carlos de Lima; http://lattes.cnpq.br/9383834641490219The study show the prevalence of intestinal parasitosis, malnutrition and mercury exposure in childish population of Barreiras riverside community, Itaituba district: western of Pará state. To detect intestinal parasitosis in excrement was used Hoffmann Method. For diagnosis of malnutrition admit the recommended method by World Health Organization (WHO). The proportion of total mercury in specimen hair was obtained to Atomic Spectrofotometry Absorption. The results showed incrase of prevalence of Ascarís fumbrícoídes and Ancylostoma duodenafe. Malnutrition acute cases to total 2,4% of population. Children with high levels of total mercury come down to 43,5%. It is also part of the clinical diagnosis the presence of yellowish-white color specific macular lesions found in the parafoveal and foveal regions, besides lipofuscin deposition generating hypofluorescence and atrophy of choriocapillaris and retinal pigment epithelium. Visual acuity of the Stargardt’s degeneration carriers decreases with their progress and tend to stabilize between 20/200 (1.0 logMAR) to 20/400 (1.3 logMAR). This thesis aimed to investigate the occurrence of association between allelic variants (mutations) and ABCA4 gene expression levels with the peripheral granulocytes morphology and clinical phenotypes, electrophysiological and retinal structure in patients harboring Stargardt’s maculopathy. Four non-synonymous and three synonymous allelic variants were identified among eleven selected subjects. The allelic combination L1395P/D1817E was found in 64% of the 22 chromosomes analyzed suggesting the occurrence of founder effect. There were no association between genotypes and clinical phenotypes analyzed. The differential values obtained for ABCA4 gene expression between patients and controls, as well as the prevalence of banded neutrophils in the peripheral blood circulation suggest the possibility they could serve as biomarkers for Stargardt. The eye fundus and angiographic images were used for the classification in the stages I, II and III of retinal impairment in subjects investigated. The thickness values of the central region of the macula among patients were much lower than those obtained for the controls, indicating a loss of photoreceptor layer. Full-field electroretinographic findings of light adapted eyes during the course of time enabled the characterization of the functional losses in investigated subjects.
