    Anemia hemolítica causada por Indigofera suffruticosa (Leg. Papilionoideae) em bovinos

    The aereal parts of Indigofera suffruticosa Mill. (family Leg. Papilionoideae) were force-fed fresh to 6 bovines in daily doses of 10 to 40 g/kg. Cattle breeders of various parts of the Northeast of Brazil accuse this plant as the cause of a non-fatal disease characterized by hemoglobinuria. The disease occurs only in years when the plant proliferates well invading the native pastures. All experimental animals had hemoglobinuria, which was transitory, inspite continuation of the administration of the plant. Two animals had no further manifestations, a third animal showed only slight other manifestations, and the other three had additional symptoms of moderate intensity. These were apathy, whitish visible mucous membranes, rough hair coat, anorexia, descrease in frequency and intensity of the ruminal movements, tachycardia, positive venous pulse and dispnoea. Before the occurrence of the hemolytic crisis the urine had a bluish-green colour. None of the experimental animals died, but one was euthanized whilst showing hemoglobinuria. Post-mortem findings were anemia, the bladder containing wine-red urine, swollen dark-brown kidneys, liver on the outside and on the cut-surface bluish and with perceptible lobular design. The main histological changes were found in liver and kidney. In the liver there was coagulative necrosis and cloudy swelling and/or cytoplasmatic microvacuolization of the hepatocytes; in the kidney there was severe nephrosis, associated with large amounts of filtrate and/or hemoglobine in the Bowman spaces, in the tubules and also in the cytoplasm of the epithelial cells.

    Aspectos epidemiológicos e clínico-patológicos comparados da intoxicação por Arrabidaea bilabiata (Bignoniaceae) em búfalos e bovinos

    Experiments showed that the clinical and pathological pictures were essentially the same, but the buffalo was at least twice more resistant than the bovine to the toxic effect of Arrabidaea bilabiata (Sprague) Sandw. It was also shown that the young leaves are twice (October, end of the dry season) or one and a half (May, end of the rainy season) more toxic than the mature leaves, and that the plant is more toxic in October. These data indicate that the smaller incidence of poisoning by plants of the group that causes sudden death in buffaloes in the Amazon Region is, in part, due to the greater resistance of this animal species to the toxic action of the plant. Also important seems to be the coincidence of the preferential habitat of the buffalo (flooded areas) with the habitat of A. bilabiata, less toxic than Palicourea marcgravii St.Hil. which is a plant of the non-flooded areas, the preferential habitat of cattle.

    Avaliação comparativa da ultraestrutura e propriedades físicas do esmalte bovino, bubalino e humano

    This study aimed to compare the morphology and physical properties of the enamel structure of bovine, buffalo and human teeth. Analysis of this tissue was performed by scanning electron microscopy, mineral composition, microhardness and surface roughness of enamel in 41 buffalo incisors (Bos taurus indicus), 41 bovine incisors (Pelorovis antiques), and 30 human permanent incisors. The results showed a significant similarity between the ultrastructure of enamel in these animal species and the one found in human samples. The chemical elements which presented higher concentration in bovine and buffalo enamel were: O,Ca and P, precisely those that form hydroxyapatite crystals - Ca10(PO4)6(OH)2. Knoop microhardness values showed no statistically significant differences between the three species. However, the surface roughness of buffalo enamel (2.16µm ±0.23) was significantly higher when compared with human (0.36µm ±0.05) and bovine teeth (0.41µm ±0.07). It is concluded that the characteristics and properties of bovine and buffalo enamel, as obtained from our analysis and testing, showed a similar morphology to that of humans. They showed a similar ultrastructural architecture, microhardness and mineral composition equivalent to the human dental tissue, becoming reference models for research.

    Avaliação do ganho de peso e aspectos reprodutivos de primíparas zebuínas suplementadas com uréia no período seco

    This study aimed to evaluate the productive and reproductive performance of cow cutting, supplemented with urea in the dry season. We used 58 females Tabapuã x Nellore crossbred, pregnant, with a mean age of 39 months and average weight of 399 kg. The worm and experimental animals were divided into two groups, in an area of 50 ha of grass Brachiaria brizantha cv Marandu, with 25 ha for each group, one group (A) supplemented with mineral and another (B) with mineral enriched with urea, the ratio of 80:20 (salt: urea). During the months of August-November, when most of the year in the dry, the grass had on average 5,800 kg of dry matter per hectare to 6.2% crude protein. These mixtures were administered in mineral cochos covered in order to meet ad libitum consumption during the dry period. The experimental design was completely randomized to two treatments. Difference was observed for final body weight (PVF) between the treatment with salt and mineral supplementation with urea, with average values of 360.1 kg and 385.8 kg, respectively. Difference was also observed for average daily gain, with an average of 0.465 kg/an/day for the treated group and 0.284 kg/an/day for the control group, postpartum with a gain of 41.2 and 26.5 kg/animal, respectively. The best performance of the supplemented animals should be related to the increase in dry matter intake promoted by supplementation with urea (12.610 kg to 8.744 kg). The best body growth (weight and body score) to the beginning of the amounts reflected in the expression of heat these animals, whereas 75% of cows supplemented with urea had expressed estrus during the first 60 days from the amounts to only 53, 33% of nonsupplemented. When we evaluated the whole period from the mount was not observed difference between the expression of estrus in females supplemented or not, a fact related to good body score submitted by the end of the animal to mount, 3.7 in females supplemented and 2, 9 in non-supplemented.

    Brachiaria spp. poisoning of ruminants in Brazil

    Brachiaria species are the most important grasses for cattle production in Brazil. However, a limiting factor for the use of Brachiaria spp. is their toxicity. Most outbreaks of hepatogenous photosensitization are caused by B. decumbens; however B. brizantha, B. humidicola and B. ruziziensis can also cause poisoning. The poisoning affects cattle, sheep, goats and buffalo. Sheep are more susceptible than other animal species and the young are more susceptible than adults. There are differences in susceptibility among animals of the same species and it has been suggested that this resistance is genetic. Also has been suggested that buffalo and probably some sheep are resilient, i.e. when poisoned these animals have histologic lesions and high GGT serum concentrations, but do not show clinical signs. In general, saponin concentrations are higher in growing plants, but outbreaks occur all over the year, probably due to unexplained rise in saponin concentration in the plant. A clinical syndrome of progressive weight loss and death, without photosensitization, has been reported in cattle poisoned by B. decumbens. Main preventive measures are based on the selection of resistant or resilient animals and on the development of Brachiaria species or varieties with low saponin concentration.

    Co-cultivo de embriões bovinos com células-tronco adultas derivadas de tecido adiposo

    The in vitro embryo production (IVP) is a biotechnology used to increase the reproductive potential of genetically superior animals, embryos produced in vitro are inferior in quality when compared in vivo. Techniques trying improve the rates of in vitro produced embryos. One technique is the co-culture system using somatic cells that remove toxic metabolites and protect against oxidative stress. Mesenchymal stem cells derived from adipose tissue (CTA) are multipotent cells that secrete growth factors and cytokines. The stem cells were used in in vitro co-culture of bovine embryos at different concentrations in order to improve the protocol PIVE. CTAs underwent differentiation into three mesenchymal lineages, and was performed immunophenotyping of specific membrane markers of MSCs. The cleavage rate was assessed on the second day after fertilization and blastocyst rate on the seventh day when they were stored for counting the total number of cells and gene expression. The results were analyzed by ANOVA, T-test and post-test of Fisher, adopting a significance level of 5%. Treatment of co-cultivation with CTAs significantly influenced the formation of the blastocyst, the total number of cells in embryos and correlated gene expression pluripotency and carbohydrate metabolism. These results showed increased production rate and quality of embryos produced in vitro co-culture with CTAs compared to co-culture with granulosa cells. These results also indicate that the constant presence of CTAs in co-culture is superior to conditioning with CTAs. Checked the effects of CTAs can occur through soluble factors or via exosomes secreted by CTAs. Future studies are needed to clarify the possible cause for positive effects observed in this work by the CTAs in co-cultivation.

    Comparação da sensibilidade de bovinos e búfalos à intoxicação por Palicourea marcgravii (Rubiaceae)

    In order to verify if buffaloes are more resistant than cattle to poisoning by Palicourea marcgravii, the plant was given per os simultaneously to buffaloes and cattle. Seven buffaloes and three head of cattle were used. Doses of 0.5, 1.0 and 2.0 g/kg did not cause signs of poisoning in buffaloes; but doses of 3.0, 4.0 and 6.0 g/kg caused death. In cattle 0.25 g/kg did not cause symptoms; but 0.5 and 2.0 g/kg were lethal. The influence of exercise on the onset of symptoms, the time which elapsed from the beginning of the administration of the plant to the appearance of symptoms, and the symptoms themselves, were similar in the two animal species. The clinical course was longer in buffaloes; whilst in cattle the period between the onset of severe symptoms and death lasted 9 to 17 minutes; in buffaloes it varied from 10 minutes to 1 hour 28 minutes. It can be concluded, that buffaloes are about six times more resistant than cattle to the toxic action of P. maracgravii. The lower incidence of deaths in buffaloes by poisonous plants in the Amazon region is, at least in part, due the lower sensibility of buffaloes to poisoning by this plant. Another factor responsible for the smaller number of buffaloes which die due to plant poisoning in the Amazon region, is possibly that buffaloes prefer the lowlands, e.g. temporarily flooded areas, which are the habitat of Arrabidaea bilabiata, the second most important poisonous plant of the Amazon region. This plant is less poisonous than P. marcgravii, and its habitat are the higher not flooded areas. So it would be safer, in order to diminish losses, to breed buffaloes instead of cattle in areas where P. marcgravii occurs. The reason for the greater resistance of buffaloes should be further investigated as a possible prophylactic measure to avoid poisoning by P. marcgravii in cattle. On the other hand, Australian investigators modified genetically the ruminal bacteria Butyrivibrio fibrisolvens by introducing a gene, isolated from a Moraxella sp, which codifies a dehalogenase, capable of hydrolyzing fluoroacetate, the poisonous principle of P. marcgravii. Transferring the genetically modified B. fibrisolvens into the rumen of animals, which consume plants that contain fluoroacetate, would be a viable method to control that poisoning through a ruminal detoxifying process for the toxin. Through preliminary contacts, the director of the consorcium in Australia, responsible for the modification of the bacteria, declared that he is interested to sell the technology to our country, but it would be necessary to know if it is permitted to introduce into Brazil the genetically modified bacteria. If allowed, it would be necessary to perform investigations on the viability and the methodology for using this bacteria to control P. marcgravii poisoning in our country.

    Comparação de peso e rendimento de carcaça entre vacas bovinas e bubalinas

    Records of alive weight, weight of carcass, weight of primary cuts and income of carcass, proceeding from the Company Scope Ltda, located in the district of Icoaraci, Located in Belém-Pará, had been used to discriminate possible differences between bubaline and bovine, abated females during the year of 2007. The registers of alive weight had been used to separate the sample in light and weighed females, separately in each species, using as threshold the corresponding value the median of the alive weight in each species. Thus, four groups of females, bubalinas and bovine, light and weighed had been composites. For comparison between female vectors of averages of the weights alive, weights hot carcass, left and right hot carcass, weight of the cold carcass, front weight, back weight, weight tip-of-needle and weight leather, between bubaline and bovine, light and weighed, it was used procedures of multivaried inference. The results had disclosed the heterogeneities for the weight of abate of the animals, inside and between species, having a clear-cut standard of ideal weight to it does not abate it. Bubaline had gotten greaters weights for carcass component, when compared with the females of the bovine group. Excepting the weight of the cold carcass and the tip of needle, bubaline of the group weighed, had gotten comparative average greater when to the light group. Inside of the group of bovine females it did not have difference statistics for the weight of carcass cold (P<0,01). It is verified that, in average, bovine had presented greater income, except for the front. The intra-racial Groups had been presented significantly different between itself (P). On the other hand, it was not verified intra-racial differences for the incomes of carcaças hot, cold carcass, front and back pernil.

    Deficiências minerais em vacas em lactação da bacia leiteira do município de Rondon do Pará, estado do Pará

    A study of mineral deficiencies was performed in lactating cows from dairy herds belonging to 13 farms of Rondon do Pará, state of Pará, Brazil. We determined the levels of phosphorus (P) in the bone and levels of copper (Cu), cobalt (Co), selenium (Se) and zinc (Zn) in the liver of 47 dairy cows in lactation 2. The samples were collected by means of biopsies taken on the top third of the 12th rib of the right side and the caudal edge of the caudate liver lobe, respectively. The herds were composed of crossbred (Holstein x Zebu) cattle, kept in extensive production system on Brachiaria brizantha cv. Marandu pasture and received mineral supplementation. The mineral mixture on 12 farms were commercial type, called "full" and the owners added a quantity of salt above the manufacturer's recommendations on ten farms. The mineral mixtures were provided in troughs without covering on seven farms and on other eight, providing of the mineral mix was not performed daily. On 11 farms there were clinical histories consistent with mineral deficiencies in livestock, and placenta retention and osteophagia were the most frequently reported ones. The results of chemical analysis showed deficiency of P on five farms, of Co on three farms, of Se on nine farms and of Zn on ten farms. It is concluded that the mineral supplementation performed on the farms do not supply the daily demands of P, Se and Co, based on the estimated daily consumption of 30g of NaCl per animal; the little adequate or inadequate troughs for proper supplementation, as well as inconstant supply of mineral mixtures contribute to the deficiency of one or more minerals.

    Degenerative joint disease in cattle and buffaloes in the Amazon region: a retrospective study

    A retrospective study of the epidemiological and clinic-pathological aspects of cattle and buffaloes with degenerative joint disease (DJD) was conducted in the state of Pará, Brazil. From 1999 to 2014, eleven cattle and 24 buffaloes were evaluated. All the treated animals with suspected DJD underwent a clinical examination of the musculoskeletal system. In seven cattle and eight buffaloes with clinical signs of the disease postmortem examination was performed. The common clinical signs observed in both species were chronic lameness, stiff gait, postural changes, audible crackles in the affected limb, prolonged recumbency, difficulty in getting up and progressive weight loss. The lesions observed at necropsy were: irregular articular surfaces, erosion of the articular cartilage and the underlying bone tissue, and proliferation of the periarticular bone tissue with formation of osteophytes. The most affected joints in cattle and buffaloes wereof the hind limb. In buffaloes, the main predisposing factor to the onset of DJD was phosphorus deficiency. In cattle, defects of the anatomical conformation of the hind limbs, chronic trauma due to the activities performed, such as semen collection, and advanced age possibly contributed to the emergence of the disease.

    Dermatite por lambedura em bovinos no estado do Pará

    A condition of 13 cattle from 10 farms in six districts in the state of Pará, Brazil, characterized by ulcerative skin wounds was studied. The lesions were observed in regions of the body to which the cattle had access with their own tongue; the animals licked the wounds frequently. Epidemiological studies, pathological findings and the differential diagnosis indicate that the dermatitis is caused by licking. The wounds healed after restraining the head of the animals that then were unable to lick themselves.

    Desempenho e características da carcaça e da carne de novilhos não-castrados alimentados com ou sem adição de monensina e/ou probiótico à dieta

    The objective of this experiment was to evaluate the performance, carcass and meat characteristics of non-castrated cattle with or without monensin (M) and/or probiotic (P) (Saccharomyces cerevisiae) to the diet. The animals were divided into individual stalls, being left 145 days in feedlot. The roughage used was corn silage + 1.2% of the weight live of concentrate in natural matter basis. There was no effect of the addition of the addictives supplied isolated or associated over feed intake, weight gain and fed conversion. Feed intake, weight gain and feed conversion averages presented quadratic behavior with the increase of the period feedlot. Although the addition of M or P to the diet resulted in the numeric increase (P>.05) of the intake (4.2%) the average of weight gain reduced (5.8 and 5.3%, respectively), resulting in smaller fed conversion (P>.05). The association of M+P already increased (P>.05) the food intake to the order of 9.5% with concomitant increase (P>.05) of the weight gain (6,4%) when compared to diet control. The animals fed with M+P presented better carcass finish (5.5mm), followed by the control (4.7mm), and smaller values, it was verified in the carcasses of the animals that received M (3.7mm) and P (3.5mm). The addition of monensin and/or probiotic for feedlot steers did not improve the performance, carcass and meat quality.

    Development and assessment of a latex agglutination test based on recombinant MSP5 to detect antibodies against Anaplasma marginale in cattle

    The recombinant protein MSP5 has been established as an important antigen for serological diagnosis of Anaplasma marginale by enzyme-linked immunosorbent assay (ELISA). However, due to the high cost of specialized equipment, this technique is not accessible to all laboratories, especially in developing countries in areas where the disease is endemic. The present study describes the standardization of a latex agglutination test (LAT) to detect antibodies against A. marginale based on recombinant MSP5. Compared with indirect enzyme-linked immunosorbent assay (iELISA), the relative sensitivity and specificity of the LAT were 95.21% and 91.86% respectively, with an almost perfect agreement between tests (kappa index = 0.863). These results can be considered important for the serological diagnosis of A. marginale, as they indicate that the test represents a rapid and low cost alternative to ELISA.

    Diagnóstico da paratuberculose em bovinos de corte do estado do Pará - Brasil

    514 blood samples from beef cattle were examined of to find anti- M. avium (subsp.) paratuberculosis antibodies, using commercial ELISA test kit. The animals from samples were all compous from cross between Indicus x Taurus race, between male and female, with two age class, over 36 months old and below 36 months old, from of the 23 municipality of Para State. Also were collected 100 samples of small intestine and mesenteric lymph nodes of bacteriological test through Ziehl-Neelsen method to detect the M. avium (subsp.) paratuberculosis. Were at random collected in slaughterhouse of Belem. In ELISA test, through these 514 samples, 182 (35,4%) were seropositive and 332 (64,6%) samples were seronegative. All the middle region were found soropositives animals. The female with over 36 months old presented larger response test, of the 92,06% seropositives animals. Among male the prevalency larger (76,79%,) were obtained between the most new, with below 36 months old. Of 100 samples stained through Ziehl-Neelsen method, not were found either Mycobacterium spp.. The conclusion, is the biggest number of seropositive animals of anti- M. avium (subsp.) paratuberculosis sign of disease is presented in all the regions studied, watching the risks of this disease with the elaborate more covering stud ies about the paratuberculosis with the appling of directs and indirects diagnosis methods.

    Direct detection of Mycobacterium tuberculosis complex in bovine and bubaline tissues through nested-PCR

    Post-mortem bacterial culture and specific biochemical tests are currently performed to characterize the etiologic agent of bovine tuberculosis. Cultures take up to 90 days to develop. A diagnosis by molecular tests such as PCR can provide fast and reliable results while significantly decreasing the time of confirmation. In the present study, a nested-PCR system, targeting rv2807, with conventional PCR followed by real-time PCR, was developed to detect Mycobacterium tuberculosis complex (MTC) organisms directly from bovine and bubaline tissue homogenates. The sensitivity and specificity of the reactions were assessed with DNA samples extracted from tuberculous and non-tuberculous mycobacteria, as well as other Actinomycetales species and DNA samples extracted directly from bovine and bubaline tissue homogenates. Regarding the analytical sensitivity, DNA of the M. bovis AN5 strain was detected up to 1.5 pg by nested-PCR, whereas DNA of M. tuberculosis H37Rv strain was detected up to 6.1 pg. The nested-PCR system showed 100% analytical specificity for MTC when tested with DNA of reference strains of non-tuberculous mycobacteria and closely-related Actinomycetales. A clinical sensitivity level of 76.7% was detected with tissues samples positive for MTC by means of the culture and conventional PCR. A clinical specificity of 100% was detected with DNA from tissue samples of cattle with negative results in the comparative intradermal tuberculin test. These cattle exhibited no visible lesions and were negative in the culture for MTC. The use of the nested-PCR assay to detect M. tuberculosis complex in tissue homogenates provided a rapid diagnosis of bovine and bubaline tuberculosis.

    This study aims to determine the prevalence of antibodies immunoglobulin class G (IgG) specific of this disease in bovine created, by contributing to the understanding of the epidemiology of this zoonosis in the state of Pará. The study was conducted in all Para meso, they were harvested blood samples from animals in 2070, with 1750 and 320 bovine and buffalo, respectively, coming from 52 municipalities and 100 properties, a survey of the epidemiological aspects influencing the onset of the infectious agent. Sera of animals underwent the test of ELISA Kit – Enzyme-Linked Immunosorbent Assay and indirect immunofluorescence - IFA for the detection of specific IgG antibodies to T. gondii cattle and buffalo the Imunodot® company. The animals were considered positive in the IFAT test from the dilution 1:64. It was considered statistically significant difference p 0.005. The buffaloes had higher seropositivity that the bovine, as well as the IFA test was higher than the ELISA test, and Mesoregion more seropositive was the Lower Amazon. There were no significant differences in the frequency of T. gondii, in relation to sex, type of exploitation, but no significant association for age, size of property, number of animals in the property, creation cycle, and the properties that presented greater amount of cats and dogs were the ones that had higher sororreagência the anti - T. gondii.

    Diversidade genética e eficiência de DNA microssatélites para o controle genealógico da raça Nelore

    The genetic variability and paternity testing values in Nelore breed were estimated using 11 ISAG/FAO microsatellites. The markers were organized into 4 amplification groups for semi-automated fluorescence genotyping. All markers were highly polymorphic, with an average of 8.2 alleles per locus. With a mean value of 0.48, the observed heterozygosity was lower than the expected for 10 of the loci. A significant deficit of heterozygotes was observed for 9 loci, resulting in a lack of Hardy-Weinberg equilibrium in the studied population. Polymorphism information content values exceeded 0.5 for 10 loci. The power of discrimination was >0.999 and paternity exclusion probabilities when a mother, her offspring and a putative sire are compared or when one or other parental genotype is unavailable for the combined set of markers were, respectively, >0.999 and >0.989. The set of 11 microsatellite markers proved to be an efficient tool for paternity testing in Nelore cattle.

    Efeito do cortisol na produção in vitro de embriões bovinos

    The aim of this thesis was to evaluate the effect of cortisol in oocyte maturation and bovine embryo culture. In Chapter 1 we analyzed the distribution and location of glucocorticoid receptor (GR) by immunocytochemistry in oocytes, cumulus cells, embryonic cells 2-4, 8-16 cell, morula and blastocyst, and was also verified the presence of mRNA (qualitative RT-PCR) for GR in the stages. The results showed that the GR is present in all cells analyzed. In order to verify the functionality GR in preimplantation embryo development, translation of mRNA for GR zygotes was silenced in the RNAi technique, and subsequent embryo development was analyzed. The embryonic development decreased (P <0.05) after silencing of GR mRNA. The presence of GR oocyte and the cumulus cells indicates that these cells are sensitive to the use of CG. Given this, in Chapter 2, the effects of different cortisol concentrations during in vitro maturation (IVM) of bovine oocytes and embryonic development, apoptosis rate and gene expression (NRF1, COX, TFAM, GLUT1, FASN and HSP70) were analiyzed. The concentrations of cortisol used were 0.01, 0.1 and 1 mg / mL . There was no statistical difference in the number of cells and cleavage rate, but the concentration of 0.1 mg/ml of cortisol increased the blastocyst rate when compared to the control group (without cortisol in IVM) (41 ± 10 versus 21 ± 1.2; p <0.05, respectively). The rate of apoptosis and gene expression in oocytes, cumulus cells and blastocysts was only assessed at a concentration of 0.1 mg / ml of cortisol. There was no statistical difference in the apoptotic index, and not with respect to gene expression in oocytes and cumulus cells for COX genes, NRF1, HSP70 and FASN (p> 0, 05). Regarding embryonic gene expression, only the measurements relative mRNA FASN, GLUT1 and HSP70 were increased in blastocysts treated with 0.1 g/ml during IVM when compared to embryos of the Control group (p <0.05), the other genes showed no change (p> 0.05). In Chapter 3, we investigated the use of cortisol during in vitro culture (IVC) of bovine embryos. As in Chapter 1 was identified GR in all embryonic stages and when the GR expression was silenced and embryonic development was impaired. Thus, in the experiment three different concentrations (0.01, 0.1 and 1 mg / mL) of cortisol in IVC and embryonic development was evaluated same parameters of Chapter 2. There was no statistical difference in the embryos treated with cortisol in different concentrations when compared to the control for the parameters analyzed (p> 0.05). The concentration of 0.1 mg/ml was chosen to evaluate other parameters of embryo quality. Thus, IVC embryos with or without 0.1 mg/mL of cortisol, were analyzed for apoptosis rate and gene expression, not being statistically significant difference in any of the analyzes (p> 0.05). After these studies conclude that oocytes and embryos are responsive to GC, and the addition of cortisol in IVM improves oocyte competence, but the supplementation of IVC with cortisol may not have influence on embryo development.

    Efeito do estradiol e da progesterona no desenvolvimento e na qualidade de embriões bovinos produzidos in vitro

    The development and quality of bovine embryos co-cultivated with bovine oviduct epithelial cells (BOEC) supplemented or not with estradiol and progesterone were evaluated. Oocytes were matured and fertilized in vitro (MIV/FIV) (5%CO2/38.5oC). The BOEC were cultivated in TCM-199 with or without estradiol (E2) (24h), in the same conditions of MIV/FIV. Presumptive zygotes were transferred to BOEC in suspension after in vitro maturation and fertilization of bovine oocytes with thawed percoll-selected sperm. The zygotes were cultivated in CR2 medium containing or not progesterone (P4) (G1=P4+E2), (G2=E2), (G3=P4), and (G4=control). No significant differences (P>0.05) were found in the cleavage rates among G1, G2, and G4 (53.5%, 56.3%, and 51.7%) as well as in relation to the blastocystis (BL) rates (29.3%, 31.2%, and 28.7%). However, significant differences (P<0.05) were observed in the G3 for both variables (34.5% and 16.4%). G1 and G2 showed hatching rates higher (P<0.05) than the other groups (23.3%; 23.2%), being G4 (19.3%) different from G3 (16.1%) (P<0.01). In the groups G1, G2, and G3, the total cell number of the BL increased (125.9, 128.4, and 123.6) (P<0.05) compared to G4 (112.5). These results demonstrate that the treatment of the BOEC with estradiol in first the 24 hours of culture can be used separately or in combination with the progesterone to improve the quality of bovine embryos produced in vitro.
